Klorprifos, tarım alanlarında verimin arttırılması amacıyla bitkilerin zararlı organizmalardan korunması için kullanılmaktadır. Klorprifos öğrenme ve hafıza mekanizmalarında fonksiyonel hasarlar yapmaktadır. NMDA reseptör subünitleri kognitif fonsiyonlarda rol almaktadırlar. Bu çalışmada klorprifos'un olumsuz etkilerinin mekanizmalarına bir yaklaşım sağlayabilmek maksadıyla NR2A ve NR2B düzeylerindeki etkisi araştırıldı. Dört adet grup oluşturdu. Genç klorprifos grubuna 40 mg/kg dozunda, erişkin klorprifos grubuna 70 mg/kg dozunda ciltaltı klorprifos uygulandı. Her iki grubun kontrollerine ise kendi ilaç gruplarıyla aynı hacimde serum fizyolojik uygulandı. Uygulamanın 2. ve 14. günlerinde dekapitasyon ile deney sonlandırıldı. Plazmada AChE; beyin dokusunda MDA, SOD, GSH-Px ve CAT; hipokampüste NR2A ve NR2B düzeyleri çalışıldı. Klorprifos plazma AChE düzeylerini genç sıçanlarda 2. günde, erişkin sıçanlarda 2. ve 14. günde anlamlı olarak azalttı. Klorprifos beyin dokusundaki MDA ve SOD düzeylerinde anlamlı bir değişikliğe neden olmadı. MDA düzeylerinde artış olmaması, uyguladığımız dozlarda beyin dokusunda lipid peroksidasyonunun olmadığını göstermektedir. Klorprifos beyin dokusundaki CAT aktivitesinin genç sıçanlarda 2. ve 14. günde azalmasına neden olurken, erişkin sıçanlarda GSH-Px aktivitesinde 2. günde artışa sebep olmuştur. Bu etkiler klorprifos'un enzimler üzerine direkt etkisinden kaynaklanıyor olabilir. Klorprifos genç sıçanlarda 14. günde NR2A ve NR2B reseptör düzeylerini arttırmıştır. Erişkin sıçanlarda 2. günde NR2A ve NR2B düzeylerini arttırırken, 14. günde ise yalnızca NR2B düzeylerini arttırmıştır. Reseptör düzeylerindeki bu artışlar kolinerjik ve glutamaterjik sistem arasındaki etkileşimin bir sonucu olabilir. Çalışmamızda yalnızca NR2A ve NR2B düzeyleri değerlendirildiği için, diğer NMDA reseptör subünitlerindeki değişikliklerin de tespiti, klorprifos'un NMDA reseptörleri üzerine olan etkisinin anlaşılmasına ve çıkan sonuçların yorumlanmasına katkıda bulunacaktır. Kolinerjik sistem ve glutamaterjik nöronal sistem arasındaki etkileşim mekanizmaları henüz net olarak ortaya konulamadığından, daha ileri araştırmalara ihtiyaç vardır. Chlorpyrifos is used for protecting the plants against harmful organizations in order to increase the productivity in agricultural industry. Chlorpyrifos makes functional damages on mechanisms of learning and memory. NMDA receptor subtypes have a role on cognitive functions. In this study the effects of chlorpyrifos on levels of NR2A and NR2B was investigated to get an approach to the mechanisms of negative effects. Four groups were constituted. Chlorpyrifos was applied subcutaneously to juvenile group as 40 mg/kg dose and to adult group as 70 mg/kg dose. Isotonic sodium chloride solution was applied to both of the controls of these two groups as the same volume as their own drug groups. The examination was finalized by decapitation on 2nd and 14th days of the administration. Levels of AChE in plasma; MDA, SOD, GSH-Px and CAT in brain tissue; NR2A and NR2B in hippocampus were investigated. Chlorpyrifos reduced significantly the plasma's AChE levels of juvenil rats in 2nd whereas of adult ones in 2nd and 14nd days. Chlorpyrifos did not cause a significant change in levels of MDA and SOD in brain tissue. No increase in MDA levels show that there has been no lipid peroxidation in brain tissue throughout the doses we applied. Chlorpyrifos decreased the CAT activities in brain tissues in 2nd and 14nd days for juvenil rats whereas increased the GSH-Px activities in 2nd days for adult rats. These effects may be rised from chlorpyrifos' direct effects on enzymes. In juvenile rats chlorpyrifos increased the NR2A and NR2B receptor levels in 14nd day. In adult rats it increased the NR2A and NR2B levels in 2nd day whereas only NR2B levels in 14nd day. These increases in receptor levels may be the result of the interaction between cholinergic and glutamatergic systems. In our work only NR2A and NR2B levels were evaluated so that determining the changes in other NMDA receptor subtypes levels may help to understand the effects of chlorpyrifos on NMDA receptors and to interpret the results obtained. Since the mechanisms of the influences between cholinergic system and glutamatergic neuronal system have not been identified clearly, there is a need for more advanced studies.
