Aspirin ve Nimesulid nonsteroid antiinflamatuvar özellikleriyle beraber antioksidan ve antiagregan özelliklere sahiptirler. Bu deneysel çalışmada antioksidan özelliği olan Aspirin ve Nimesulid'in streptozo to sinle oluşturulan diyabetik ratlarda, vasküler sistem ve böbrek dokusu üzerine hasarını azaltmasını araştırdık. Çalışmamızda her bir grupta 10'ar adet rat olmak üzere 4 grup vardı. Gruplar; Kontrol, DM (STZ 50 mg/kg, i.p tek doz), DM ? Asp (10mg/kg/gün, gavaj,6 hafta) ve DM ? Nim (18 mg/kg/gün, gavaj, 6hafta) şeklindedir. Reaktif oksijen radikallerinin (ROS) göstergesi olan biyokimyasal parametrelerden MDA, GPx, CAT ve SOD düzeylerine bakılırken, aort ve böbrek dokusu histopatolojik olarak da değerlendirildi. DM ve Kontrol grupları karşılaştırıldığında; DM grubunda MDA düzeyi Kontrol grubuna göre anlamlı olarak daha yüksek (p<0,05) bulunurken, GPx ve SOD düzeyleri açısından istatistiksel açıdan anlamlı fark saptanamamıştır (p>0,05). Aspirin verilen DM'li grupta ise MDA ve GPx düzeyi anlamlı olarak düşmüştür (p<0,05). Nimesulid ise sadece MDA'yı anlamlı olarak azaltmıştır (p<0,05). DM'li grupta CAT düzeyi anlamlı olarak düşmüş ve Aspirin ve Nimesulid ise bu düşüşü anlamlı olarak önlemiş ve Kontrol düzeyine çıkarmıştır. Biyokimyasal bulgularımıza paralel olarak, diyabetik ratlarda böbrek dokularının histolojik incelemelerinde oksidatif hasarın göstergesi olabilecek bulgular izlendi. DM grubu ile Kontrol grubu karşılaştırıldığında bu bulgular anlamlı olarak (p<0,05) farklı iken Aspirin verilmiş DM'li grupta bu histopatolojik değişiklikler hafif derecede gözlendi. Ancak Nimesulid verilen DM'li grupta ise yukarıdaki histopatolojik değişikliklerin DM'li gruptakine benzer olduğu gözlendi. Yani Nimesulid Aspirin kadar diyabetin sebep olduğu histopatolojik bozuklukları düzeltemedi. Sonuç olarak Aspirin'in diyabetli ratlarda damar ve böbrek dokusunda oluşan hasarda Nimesulid'e göre daha etkili olduğu görüldü. In this experimental study, the reduction of damage on vasculer system and kidney tissue of rats that were induced diabetic with STZ, was investigated by using Aspirin and Nimesulide which have antioxidant properties. In this study, there were four groups and each of them consisted of ten animals. These four groups classified as; Control, DM (STZ 50 mg/kg, i.p single dose), DM ? Asp (10mg/kg/day, po,6 weeks) ve DM ? Nim (18 mg/kg/day, po, 4 weeks). While the level of MDA, GPx, CAT and SOD, biochemical parameters of ROS, were gazing at, the kidney tissue and aort were also evaluated as hystopatological situation. When DM and Control groups were compared, it was established that the level of MDA in DM group was significantly higher than Control group (p<0,05). On the other hand, according to the levels of GPx and SOD and from the point of statically, significant difference wasn't established. However, in DM group treated with Aspirin, levels of MDA and GPx decreased significantly (p<0,05). Nimesulide was only decreased MDA level significantly (p<0,05). In DM group, the level of CAT decreased significantly but Aspirin and Nimesulide prevented decrease significantly and increased it to Control levels. Together with biochemical findings, according to the hystological investigation on kidney tissue of diabetic rats, it was observed that there could be signs of oxidative damage. When DM and Control groups compared, while these findings were significantly different (p<0,05), in DM group treated with Aspirin, this hystopatological damages observed lightly. However, in DM group treated with Nimesulide the same hystopatological damages with DM group were observed. It means that Nimesulide didin't cure the hystopatological damages that were caused by diabetic as Aspirin. As a result, it has been determined that Aspirin was more effective than Nimesulide about improving the damages on vascular and kidney tissue of diabetic rats.
