MTX çeşitli kanser türlerinde ve bazı inflamatuvar hastalıklarda, antineoplastik ve antiinflamatuvar ajan olarak yaygın şekilde kullanılmaktadır. Metotreksat'ın karaciğer ve böbrekler üzerine toksik etkileri olduğu bilinmektedir. Bu çalışmanın amacı Metotreksat'ın karaciğer ve böbrekler üzerinde oluşturduğu hasara karşı AG ve L-NAME'nin etkilerinin incelenmesidir. Bu çalışmada toplam 54 adet Wistar albino türü erkek sıçan kullanıldı. Sıçanlar 6 gruba ayrıldı: Grup I (kontrol, n=9), Grup II (20 mg/kg MTX, n=9), Grup III (20 mg/kg MTX? 20 mg/kg AG, n=9), Grup IV (20 mg/kg MTX? 3,5 mg/kg L-NAME, n=9), Grup V (20 mg/kg AG, n=9) ve Grup VI ( 3,5 mg/kg LNAME, n=9). Çalışmanın beşinci günü anestezi altında deney sonlandırılarak sıçanların karaciğer ve böbrek dokuları alındı. Histolojik incelemeler için alınan dokular % 10'luk nötral formaldehit içerisinde tespit edildi. Biyokimyasal inceleme için alınan dokularda lipid peroksidasyonu ürünü olan malondialdehid (MDA) seviyeleri incelendi. Histolojik incelemede; kontrol, AG ve L-NAME gruplarında karaciğer ve böbrek dokuları normal histolojik görünümde izlendi. MTX grubunda kontrol grubu ile karşılaştırıldığında, karaciğer ve böbrek dokularında hasarlanma tespit edildi. MTX-AG ve MTX-L-NAME gruplarında bu hasarlanmanın azaldığı gözlemlendi (p<0,01). Biyokimyasal olarak kontrol ve diğer grupların böbrek dokusu MDA düzeylerinde bir farklılık bulunamadı (p>0,01). Karaciğer dokusunda ise kontrol grubu ile karşılaştırıldığında, MTX-AG grubunda bir artış olduğu (p<0,01) diğer gruplarda bir farklılık olmadığı gözlendi (p>0,01). L-NAME ve Aminoguanidin'in, metotreksat kaynaklı karaciğer ve böbrek toksisitesi üzerine sadece yapısal olarak koruyucu bir etkisinin olduğunu gözlemledik. Methotrexate (MTX) is widely used as an antineoplastic and antiinflammatory agent in a variety of cancers and certain inflammatory disorders. MTX's toxic effects on the liver and kidneys are known. The purpose of this study is to examine Aminoguanidine's (AG) and NG Nitro L-Arginine Methyl Ester's (LNAME) effects on the MTX's liver and kidney damages. In this study 54 male Wistar albino rats were used. The rats were divided into six groups: Group I (control, n=9), Group II (20 mg/kg MTX, n=9), Group III (20 mg/kg MTX? 20 mg/kg AG, n=9), Group IV (20 mg/kg MTX? 3,5 mg/kg LNAME, n=9), Group V (20 mg/kg AG, n=9) and Group VI (3,5 mg/kg L-NAME, n=9). At the end of the fifth day, liver and kidney tissues of the rats were collected under the anesthesia. Tissues taken for histological examination, were fixed in 10 % neutral formaldehyde. Lipid peroxidation product malondialdehyde (MDA) levels were examined in the tissues taken for biochemical analysis. When compared with the control group's tissues, damage were present in the MTX group's liver and kidney tissues, in the tissues of the MTX-AG and MTX-LNAME groups, damage were decreased, AG and L-NAME group's tissues were histologically normal (p<0,01). Biochemically control's and other groups kidney tissue MDA levels were not different from each other (p>0,01). When compared with the control group, MDA levels in the liver tissue's of MTX-AG group, a significant increase (p<0,01) and other groups were not different from each other (p>0,01) was observed. We observed only morphological protective effect's of Aminoguanidine and L-NAME on methotrexate induced liver and kidney toxicity. Keywords : Aminoguanidine, kidney, liver, L-NAME, methotrexate.
