Bu çalışmada sıçanlarda deneysel gentamisin (GM) nefrotoksisitesinde oksidatif stresin rolü ve olası oksidatif stres üzerine kafeik asit fenetil ester.in (CAPE) etkisi araştırıldı. Çalışmamıza 40 dişi Wistar-albino sıçan alınarak, her grupta 10 sıçan olacak şekilde 4 gruba ayrıldı. Kontrol grubuna 1 ml/kg/gün serum fizyolojik (SF), CAPE ve GM+CAPE gruplarına 10 .mol/kg/gün intraperitoneal (i.p.) CAPE 12 gün boyunca verildi. GM grubuna 2 gün boyunca günde bir kez 1 mL/kg SF i.p. yoldan verildi, 8 gün GM 100 mg/kg/gün dozda i.p. , son 2 gün boyunca günde bir kez 1 mL/kg SF i.p.olarak enjekte edildi. GM+CAPE grubuna ilk 2 gün, günde bir kez CAPE 10 .mol/kg/gün i.p, 8 gün boyunca günde bir kez 10 .mol/kg/gün CAPE i.p+ GM 100 mgr/kgr/gün i.p, kalan 2 gün boyunca da günde bir kez 10 .mol/kg/gün CAPE i.p olarak toplam 12 gün süreyle enjekte edildi. On ikinci gün tüm sıçanların yaşamına son verilerek kan örnekleri ve böbrekleri alındı. Sol böbrek dokusunda histopatolojik değerlendirme yapıldı. Sağ böbrekte ise malondialdehit (MDA), nitrik oksit (NO) ve glutatyon peroksidaz (GSH-Px) , süperoksit dismutaz (SOD) ve katalaz (CAT) enzim aktiviteleri ölçüldü. Serumda BUN ve kreatinin değerleri ölçüldü. GM ve GM+CAPE gruplarında histopatolojik değerlendirmede nefrotoksisiteye ait bulgular gözlendi. GM grubunda kontrol grubuna göre serum BUN ve kreatinin değerlerinde anlamlı artış saptandı (p<0.0001). GM grubunda, kontrol grubuna göre böbrek dokusunda antioksidan enzimlerden SOD, GSH-Px, CAT.ın aktivitesinin düşük (p<0.0001) olduğu tespit edildi. NO ve MDA seviyeleri ise yüksek olarak saptandı (sırasıyla p<0.0001, p<0.0001). GM.ne CAPE.nin eklendiği grupta ise GM.nin renal dokuda oluşturduğu oksidatif stresin azaldığı tespit edildi. Bu bulgular GM nefrotoksisitesinde oksidatif stresin önemli bir mekanizma olabileceğini ve CAPE kullanımının, GM nefrotoksisitesi sonucunda oluşan oksidatif stresin önlenmesinde etkili olduğunu göstermiştir.

In this study, the role of oxidative stres in experimental Gentamicin (GM) induced nephrotoxicity and the effect of caffeic acid phenethyl ester (CAPE) on possible oxidative stres in rats were investigated. Forty famele Wistar rats were randomly divided into four groups. Each group consisted of 10 rats. Control group received 1 ml/kg serum physiologic (SF) intraperitoneally (i.p.) once a day, CAPE and GM+CAPE groups received 10 .mol/kg/day CAPE i.p. for 12 days. GM group; injected i.p wiht 1 ml/kg SF for 2 days before GM treatment and after the GM and afterwards i.p wiht 100 mg/kg GM for 8 days, continued only injected i.p wiht 1 ml/kg SF for 2 days. GM+ CAPE treated group; injected i.p with 10 .mol/kg CAPE for 2 days before GM treatment and daily during GM treatmend and after the GM treatment for 2 days. On the twelveth day of the study all rats were sacrificed and then blood samples and kidneys were taken. Left kidneys were used for histopathological evaluation. Malonyldialdehyde (MDA) and nitric oxide (NO) levels, glutathione peroxidase (GSH-Px), superoxide dismutase (SOD) and catalase (CAT) enzyme activities were determinated in right kidneys of rats. Levels of BUN and creatinin were studied in serum. Hystopathologic evaluation showed nephrotoxicity findings in GM and GM+CAPE groups. Serum BUN and creatinin leves of GM group was significantly higher than control group (p<0.0001). In group, SOD, GSH-Px, CAT enzyme activities were lower (p<0.0001) than the control group. NO and MDA levels were higher (respectively p<0.0001, p<0.0001) than the control group. Where as in GM plus CAPE given group, it has been observed that the oxidative stress amount was decreased in renal tissue. According to these findings, it could be concluded that oxidative stress may be a critical mechanism in GM nephrotoxicity and using of CAPE may be an effective treatment option in prevention of oxidative stress due to GM nephrotoxicity.
