Kemik iyileşmesi biyolojik, mekanik ve sistemik faktörlerin etkisinde olan karmaşık bir süreçtir. Sigaranın kemik iyileşmesi üzerine olumsuz etkileri olduğu bilinmektedir. Sigaranın neden olduğu bu hasarı önlemek için çeşitli gıdalar, vitaminler, antioksidanlar ve ilaçlarla çalışmalar yapılmaktadır. Ancak bunların hepsi ya yalnızca osteoblastlara ya da osteoklastlara etki etmektedir. Polifenol yapıda bir antioksidan olan resveratrol, antioksidan özelliğinin yanı sıra hem osteoblast stimülasyonu ve hem de osteoklast inhibisyonuna neden olmaktadır.Bu çalışmanın amacı deneysel olarak sigara dumanına maruz bırakılan ratlarda kronik sigara içiciliği modeli oluşturarak, sigaranın ve resveratrolün kemik iyileşmesi üzerine etkilerinin ve oksidatif stres ile kemik iyileşmesi arasındaki ilişkinin araştırılmasıdır. Bu amaçla denekler 4 hafta süresince 6 sigara/gün sigaraya maruz bırakılarak kronik sigara içiciliği modeli oluştuktan sonra trefin frezle femurda 3 mm çapında defekt açılmıştır. Defektin oluşturulduğu gün başlanarak yeni kemiğin oluştuğu 28 günlük dönemde deneklere 20 mg/kg dozunda resveratrol verilmiştir. Histomorfometrik incelemede osteoblast, osteoklast sayıları, yeni kemik alanı, biyokimyasal incelemede kemik-alkalen fosfataz, serumda ve dil dokusunda total oksidan/antioksidan kapasite değerlendirilmiştir.Bu çalışmanın sonuçları sigaranın kemik iyileşmesini olumsuz etkilediğini ve resveratrol verilmesinin bu olumsuz etkiyi azaltmakta yardımcı olduğunu göstermiştir. Resveratrolün total oksidan/antioksidan kapasite üzerine etkilerinin değerlendirilebilmesi için daha farklı dozlarıyla çalışmalar yapılabilir. Bone healing is a complex process that is influenced by biological, mechanical and systemic factors. Cigarette is known to have negative effects on bone healing. A variety of nutrients, vitamins and drugs are being studied to prevent bone damage that caused by cigarette. But all these materials effect only osteoblastogenesis or osteoclastogenesis. Resveratrol is a polifenolic antioxidant that modulate bone metabolism that have both osteoblastic stimulation effect and leads to inhibition of osteoblasts.The aim of this experimental study is to investigate the effects of smoking and resveratrol on bone healing and relationship between oxidative stress and bone healing in rats exposed to cigarette smoke by creating models of chronic cigarette smoking. For this purpose, rats exposed 6 cigarettes/day during 4 weeks to establish chronic cigarette smoker model and defects was created in the femur by 3 mm diameter trephine bur. Starting from the day of creation of the defect to 28-day period that new bone healing occured, the subjects were given 20 mg/kg resveratrol.Histomorphometric examination of osteoblast, osteoclast numbers, new bone area, bone-alkaline phosphatase biochemical examination, total oxidant/antioxidant capacity of serum and tongue tissue was evaluated.The results of this study are;smoking have adverse effects on the bone healing and administration of resveratrol might help to reduce these negative effects. In order to asses the effects of resveratrol on total oxidative/antioxidative capacity further studies with different resveratrol doses are needed.
