HIF-1 ilk olarak, hipoksiye cevaben eritropoietinin (EPO) seviyelerinin artmasına sebep olan transkripsiyonel kompleks olarak tanımlanmıştır. Sonraki çalışmalar, hipoksik koşullar altında HIF-1 aktivitesinin çeşitli eritropoietin üretmeyen hücre serilerinde de bulunduğunu göstermiştir. Bu durum, HIF-1’in hipoksiye cevap olarak gen ekspresyonu aktivitesinde genel bir role sahip olduğunu ortaya koymuştur. HIF-1 heterodimer yapıdadır. HIF-1, sürekli eksprese olan HIF-1 β ve hipoksi ile ilişkili HIF-1 α alt ünitelerini içerir. Yapılan çalışmalarda HIF-1’in hedef genlerinin kodladığı protein ürünlerinin, anjiyogenez/vasküler şekil değişikliği, glukoz metabolizması, eritropoez, hücre çoğalması, canlılık, hücre göçü, hormonal regülasyon, transkripsiyonal regülasyonda rol oynadığı gösterilmiştir. HIF-1 α geninde DNA sekans değişimlerinin de tümör anjiogenezi, ilerlemesi ve metastazı, iskemik kardiovasküler bozukluk ve diabetik retinopati gibi hipoksi ile karakterize olan insan hastalıkları ile ilişkili olduğu belirlenmiştir.Biz çalışmamızda, hemoglobin düzeyi 17 g/dl ve üzerinde olan hastalarda HIF-1 α geni üzerinde 12. ezonda bulunan C1772T ve G1790A tek nükeotit polimorfizmlerini ve 2. ekzonda bulunan C111A polimorfizmini PCR-RFLP yöntemini kullanarak araştırdık. Bu üç polimorfizm sırası ile 28. kodonda serinden tirozine, 582. kodonda proliden serine ve 588. kodonda alaninden proline değişim ile sonuçlanmaktadır.Çalışmamız sonucunda, HIF-1 α genine ait C1772T ve G1790A nükleotitlerinde polimorfizm görülürken, C111A nükleotitine ait polimorfizme rastlayamadık. HIF-1 has firstly defined as a transcriptional complex which increases EPO levels as a response to hypoxia. Subsequent studies have shown that, in hypoxic conditions, HIF-1 activity also occurs in the cells which don’t produce EPO. This situation proved that HIF-1 has a general role on the gene activity as aresult of hypoxia. HIF- 1 has a heterodimer structure. It includes the HIF-1 beta subunit which is expressed continuously and the HIF-1 alfa subunit which is related with hypoxia. In several studies, it has shown that protein pruducts of the genes which HIF-1 targets, have roles on angiogenesis/vascular shape change, glucose metabolism, erythropoesis, cell proliferation, viability, cell migration, hormonal regulation and transcriptional regulation. It has determined that DNA sequence changes on HIF-1 alfa gene is also related with tumor angiogenesis, progress and methastasis, ischemic cardiovasculary disorder and diabetic rethinopathy.In our study, we have researched the C1772T and G179A one nucleotide polimorfisms on the exon 12 and C111A polimorphism on the exon 2 of the HIF-1 alfa gene using PCR-RFLP method, on the patiens whom they have equal or greater than 17 g/dl hemoglobin levels. This three polimorfism respectively result changes from serine to tyrosine on codon 28, from proline to serine on codon 582, and from alanine to proline on codon 588.As a result, we have found the polimorfisms on C11772T and G1790A, but not on c111A.
