Amaç: Bu çalışmanın amacı ligatür indüklü periodontitis modelinde sistemik rosuvastatin kullanımının alveoler kemik kaybı ve oksidatif durum üzerine etkisinin değerlendirilmesidir.Metodlar: 39 erkek Wistar Albino rat rastgele 4 gruba ayrıldı: kontrol grubu (K); Rosuvastatin grubu (R); Periodontitis grubu (P) ve Periodontitis+Rosuvastatin grubu (PR). Ligatürler her iki kadranda maksiller ikinci molar dişlerin servikslerine yerleştirildi ve ratlar oral gavaj yardımı ile su ya da rosuvastatin ile 14 gün süre ile tedavi edildi. Sakrifikasyon sonrası, maksillada histomorfometrik and histolojik analizler yapıldı. Serum malondialdehit (MDA), nitrik oksit (NO), superoksit dismutaz, glutatyon (GSH), glutatyon peroksidaz ve yüksek duyarlı C reaktif protein (hs-CRP) konsantrasyonları enzim bağlı immunosorbent assay ile değerlendirildi.Sonuçlar: Rosuvastatin, P grubu ile karşılaştırıldığında PR grubunda alveoler kemik kaybını istatistiksel anlamlı azaltmıştır (p<0,05). MDA konsantrasyonununda K grubu ile P grubu arasında istatistiksel anlamlı fark belirlendi (p<0,05) ve K ve PR grupları arasında istatistiksel fark bulunmadı (p>0,05). NO seviyesi PR grubunda P grubuna, R grubunda göre K grubuna göre yüksekti (p<0,05). GSH konsantrasyonu P ve PR gruplarında K grubuna göre istatistiksel olarak anlamlı yüksek tespit edildi (p<0,05). Alveoler kemik kaybı ve MDA (r=0,523, p=0,001), NO (r=0,370, p=0,01) ve GSH (r=0,371, p=0,01) arasında ve SOD-GSH-Px arasında (r=0,363, p=0,011) pozitif korelasyonlar tespit edildi.Tartışma: Çalışmanın sonucuna göre rosuvastatin, ligatür indüklü deneysel periodontitis modelinde alveoler kemik kaybını azalttı ve oksidatif durumu iyileştirdi. Background: The purpose of this study was to evaluate the effects of systemically administered rosuvastatin on alveoler bone loss (ABL) and oxidative status in ligature induced periodontitis in rats.Methods: Thirty nine male Wistar Albino rats randomly divided into 4 groups: Control group (C); Rosuvastatin group (R); Periodontitis group (P); and Periodontitis+Rosuvastatin group (PR). Ligatures were placed at the cervix of the maxillary second molars in both quadrants and all rats were treated with water or rosuvastatin by oral gavage, during 14 days. After sacrification, histomorphometric and histological analysis were performed on the maxillae. Serum malondialdehyde (MDA), nitric oxide (NO), superoxide dismutase, glutathione (GSH), glutathione peroxidase, and high sensitive C reactive protein (hs-CRP) concentrations were evaluted by enzyme-linked immunosorbent assay.Results: Rosuvastatin significantly decreased ABL in the PR group compared to the P group (p<0.05). MDA concentrations were statistically higher in the P group than those of the C group (p<0.05), there was not statistically significiant difference between the C and PR groups regarding MDA concentrations (p>0.05).NO levels were higher in the PR group than the P group and in the R group than the C group (p<0.05). GSH levels were significantly higher in the P and PR groups compared to the C group (p<0,05). Positive correlations were found between ABL and MDA (r=0.523, p=0.001), NO (r=0.370, p=0.01), and GSH (r=0.371, p=0.01) and between SOD-GSH-Px (r=0.363, p=0.011).Conclusion: The present data suggest that rosuvastatin decreases ABL and improve oxidative status in ligature-induced periodontitis model.
