Silibinin, Silybum marianum (Deve dikeni) adı verilen bitkiden elde edilen bir flavanoiddir. Bu deneysel çalışmada; hücre koruyucu, antiinflamatuar, antikanserojen, antioksidan etkileri olan ve proteüniriyi azalttığı bilinen Silibinini STZ ile diyabet olulturulan sıçanların böbreklerinde HIF-1α ve TLR2 genlerinin ekspresyon seviyelerine etkisini tedavi öncesi ve sonrası karşılaştırıp, histokimyasal olarak da inceledik.Çalışmamızda 5 grup bulunmakta olup: Kontrol Grubu, Diyabet Grubu (STZ 65 mg/kg, i.p. tek doz), Diyabetik Grup+100mg/kg Silibinin Tedavi Grubu, Diyabetik Grup+200mg/kg Silibinin Tedavi Grubu, Silibinin grubu (5 sıçana 100 mg/kg silibinin, 5 sıçana 200 mg/kg silibinin). Böbreğin biri HIF-1α ve TLR2 genlerinin ekspresyon seviyeleri için değerlendirilirken, diğer böbrek histopatolojik olarak değerlendirildi. Histolojik olarak ilaç alan gruplarda, diyabetik gruplara kıyasla anlamlı bir değişiklik gözlenmedi.Alınan doku örneklerinden elde edilen total RNA'lar kullanılarak RT-PCR metodu ile 2 adet hedef gen ve bir adet kalibratör genin ifadesindeki değişiklikler incelendi. Elde edilen bulgular kontrol grubu ve diğer gruplardaki örnekler ile karşılaştırılmıştır. 2-ΔΔCT yöntemi ile kat değişimleri hesaplandığında ve eşik -2/2 olarak alındığında TLR2 geninde değişimler tespit edilmiştir ancak istatistiksel olarak anlamlı bir fark bulunamamıştır (p>0,05).Sonuç olarak Silibinin’in diyabetli sıçanların böbrek dokusunda oluşan hasarda pek etkili olmadığı görüldü. Silibinin is a flavonoid derived from a plant called Silybum Marianum. In this experimental study we investigate the effects of silibinin which has anti-inflamatory, anticarcinogenic, antioxidant and cyto protective effects; on the expression levels of HIF-1α and TLR2 genes in the kidneys of STZ induced diabetic Rats before and after the treatment. We also investigate these effects by histochemically.In our study, there were 5 groups: Control Group, Diabetic Group (STZ, 65 mg / kg, ip single dose), Diabetic Group + 100mg / kg Treatment Group of Silibinin, Diabetic Group + 200mg / kg treatment group of silibinin, Group of Silibinin (100 mg / kg and 200 mg / kg silibinin). One of the kidney was used to evaluate the expression levels of HIF-1α and TLR2 genes, the other kidney was used for histopathological study. Histologically, there was not obseved any significant change in the Drug Groups, compared to the diabetic group.Total RNAs obtained from the tissue samples were investigated for changes of expression levels of 2 target and 1 calibrator gene. The findings were compared with samples from the control group and the other group. When fold change calculated by 2-ΔΔCT method and when the threshold value taken as -2/2; some changes for expression levels of TLR-2 was identified, but no statistically significant change was found (p>0,05).As a result, Silibinin was not so effective in the kidney tissue damage of diabetic rats.
