Amaç: Farklı içerik ve üretim şekillerine sahip, üç farklı kök ucu dolgu materyalinin (MTA, BioAggregate ve Biodentine) apikal örtücülüklerinin ve bağlanma dayanımlarının ex-vivo olarak incelenmesidir. Kapsam: Günümüzde, standart kök ucu dolgu maddesi olarak kullanılan MTA birçok materyalden (amalgam, Super-EBA vb.) üstün bulunmuştur. Bu çalışmada, yeni geliştirilen BioAggregate ve Biodentine materyallerinin sızıntı ve bağlanma özellikleri glikoz penetrasyon, push-out ve SEM analizi yöntemleriyle, MTA ile kıyaslanarak incelenmiştir. Ayrıca ultrasonik uçlarla kavite hazırlamanın ve dental operasyon mikroskobu (DOM) altında yapılan uygulamaların anlamlı bir fark yaratıp yaratmayacağı değerlendirilmiştir. Gereç ve yöntem: Anatomik özellikleri benzer 338 adet maksiller keser diş seçildi. Apikal boyut #40-0.06 olarak hazırlandı, kanal dolguları tamamlandı. Kök uçlarının 3 mm'si 90 derecelik açı ile uzaklaştırıldı. Glikoz penetrasyon testi için 170 diş, 2 kontrol ve 12 deney grubuna ayrıldı (MTA, BioAggregate, Biodentine materyalleri DOM+Ultrasonik, DOM+Frez, ultrasonik veya frez ile açılan kavitelerde uygulandı). Örnekler glikoz penetrasyon düzeneğine yerleştirilerek 60 dakika boyunca 103kPa basınç altında bırakıldı, sızan glikoz spektrofotometrik olarak belirlendi. Bağlanma dayanımı için 156 örnek de aynı şekilde hazırlanıp 12 alt gruba ayrıldı; 0,5 mm/dk hızla, 0,75 mm çapındaki bir uç ile push-out testi uygulandı. Taramalı elektron mikroskobu ile 2000x büyütme altında, her grubu temsil eden birer diş (toplam 12), dolgu-dentin arayüzü ilişkisi açısından değerlendirildi. Elde edilen bulgular Shapiro-Wilk, Kruskal-Wallis, Mann-Whitney U, Tek yönlü varyans, post hoc Tukey HSD ve Kendall's tau-b testleri ile istatistiksel olarak değerlendirildi. Bulgular: Glikoz penetrasyon (1,26-1,41 g/L) ve push-out (6,65-8,72 MPa) testinde materyallere bağlı anlamlı bir fark bulunmamıştır. DOM kullanılmayan gruplarda, kullanılan gruplara göre anlamlı derecede fazla sızıntı meydana geldiği (p<0,05) ve bağlanma dayanımlarının anlamlı derecede düşük olduğu görülmüştür (p<0,05). En az sızıntı ve en yüksek bağlanma dayanımı BioAggregate+DOM+Ultrasonik grubundadır. SEM görüntüleri test bulgularını desteklemiştir. Sonuç: Bu çalışma sınırları içerisinde, Biodentine, BioAggregate ve MTA materyallerinin DOM altında ultrasonik uçlarla hazırlanan kavitelerde kök ucu dolgusu olarak kullanımı önerilebilir. Aim: The aim of this study is to evaluate the apical sealing and bonding abilities of the different retrograde filling materials (MTA, BioAggregate and Biodentine) with different manufacturing forms and ingredients ex vivo. Scope: Today, MTA become a standard material in surgical endodontics, owing to superior properties in comparison to various materials such as amalgam, Super EBA, IRM etc. In this study, new materials Biodentine and BioAggregate were evaluated using glucose filtration, push-out and SEM methods by comparing with MTA. Besides, it is also investigated, if the use of ultrasonic tips for cavity preparation and use of dental operating Microscope (DOM) for the retrograde fillings make a significant difference. Materials and methods: Maxillary incisors (n=338) with similar anatomic features were selected. The roots were prepared to an apical size #40-0.06 and were obturated. Root apexes were cut 3mm off with 90 degree angle. For the glucose filtration test, 170 samples were divided into 2 control and 12 experimental groups. (MTA, BioAggregate, Biodentine materials were applied to the cavities prepared using DOM+ultrasonic tip, DOM+bur, Ultrasonic tip or bur).They were placed into a glucose filtration apparatus and were exposed to 103kPa pressure for 60 minutes. The amount of leaked glucose was measured spectrophotometrically. To measure the bond strength, 156 samples prepared with same methods were divided into 12 experimental groups and push-out test was performed at a speed of 0.5 mm/min using a 0.75 mm diameter plunger. The correlation between filling materials and dentin walls were examined in 12 samples using scanning electron microscope at 2000x magnification. Data were analyzed statistically by using Shapiro-Wilk, Kruskal-Wallis, Mann-Whitney U, One-way Anova, post hoc Tukey HSD and Kendall's tau-b tests. Results: Retrograde filling materials showed no significant difference in terms of glucose filtration (1,26-1,41 g/L) and push-out values (6,65-8,72 MPa). In the groups without DOM, significantly more leakage (p<0.05) and significantly less bonding strengths (p<0.05) occurred than that of the DOM-used. The least leakage and the highest bond strength was determined in group BioAggregate+DOM+Ultrasonic. These findings were correlated with SEM micrographs. Conclusions: Under the limitations of this study, the use of Biodentine, BioAggregate and MTA can be recommended as retrograde fillings in the cavities prepared with ultrasonic tips under DOM.
