Anne Sütüyle Maruz Kalınan Sentetik Gıda Boyalarının Sıçanlarda Öğrenme ve Nörodavranış Üzerine Etkisi Gıda boyaları gıdaların albenilerini artırmak veya üretimleri sırasında oluşan istenmeyen renk farklılıklarını gidermek amacıyla kullanılan gıda katkı maddeleridir. Gebelik dönemi ve çocukluk döneminde sentetik gıda boyalarına maruziyetin öğrenme ve davranış üzerine etkileri ortaya konmuştur. Ancak literatür taramasında, bu maddelere laktasyon döneminde maruziyetin erişkin döneme etkilerini gösteren araştırmalara rastlanmamıştır. NMDAR (N-metil- D-aspartat reseptörleri)'nin öğrenme ve hafızanın oluşma sürecinde etkili olduğu düşünülmektedir. Bu amaçla, çalışmamızda laktasyon dönemi boyunca ADI ve NOAEL dozlarında maruz kalınan sentetik gıda renklendiricilerinin (Eritrosin, Ponceau 4R, Allura Red AC, Sunset Yellow FCF, Tartrazin, Amarant, Brilliant Blue, Azorubin, Indigotine) erişkin döneme geldiklerinde sıçanlarda NMDA reseptörü alt birimlerinden NR1, NR2A, NR2B ekspresyonu ve öğrenme deneyleriyle öğrenme ve davranış üzerine etkilerini araştırdık. Çalışmamızda 18 adet dişi gebe sıçan kontrol, ADI ve NOAEL grubu olmak üzere üç eşit gruba bölündü. Gıda boyası grubuna, emzirme dönemi boyunca (21 gün) 9 sentetik renklendirici karışım halinde ADI ve NOAEL dozlarında günlük olarak verildi. Ardından yavrular 3 aylık olunca her grupta 12 dişi 12 erkek olacak şekilde rastgele seçildi bir hafta boyunca öğrenme deneylerine tabi tutuldu. Test sonunda hipokampüsleri çıkarılarak homojenatlarında NR1, NR2A ve NR2B reseptörleri çalışıldı. NR2A ve NR1 reseptör ekspresyonu açısından her iki cinsiyette de kontrol grubuna oranla istatistiksel olarak anlamlı bir fark saptanmadı (p>0,05). NR2B ekspresyonunun deney grubu dişilerde kontrol dişiye göre anlamlı oranda azaldığı saptandı ve öğrenme deneylerinde de dişilerin hafıza deneylerinde performanslarının düştüğü tespit edildi. Bu veri reseptör ekspresyonundaki düşüşü destekler bir bulgudur. Anne karnında gıda boyalarına maruziyet erişkin dönemde öğrenmede rol alan reseptörlerin bazılarının düzeylerini etkilemekte ve bu etki cinsiyete göre farklılık göstermektedir. Food colorings are the additives used to increase the allure of foods or eliminate unwanted color variations that occur during production. Exposure to food additives during the critical development period, which extends from the sixth month of gestation to several years after birth in human, has been implicated in the induction and severity of some childhood behavioral and development disorders and learning disabilities. However, the effects of synthetic food colors exposed via breastfeeding on learning and behavior in adult rats haven't been reported yet. N-methyl- D-aspartate receptors (NMDARs) are thought to be effective in the learning and memory generating process. In this study, we aim to investigate the effects of exposure to synthetic food colors (Erythrosine, Ponceau 4R, Allura Red AC, Sunset Yellow FCF, Tartrazine, Amaranth, Brilliant Blue FCF, Azorubine and Indigotine) in ADI and NOAEL doses via breastfeeding on subunit concentration of NMDARs (NR1, NR2A and NR2B isoforms) in rats by Western-Blotting and on learning and neurobehaviour by learning trials in offspring when they became adults. In our study, 18 female pregnant rat were divided into three groups as control, ADI and NOAEL groups. A mixture of 9 food colors was given daily to the food colour group during twenty-one day. When they became 3 month-old, 12 male and 12 female offspring were selected from the each group randomly. Selected rats were subjected to learning experiments for one week. At the end of learning experiments, hippocampus were removed, NR1, NR2A and NR2B receptors were studied in homogenates. There was no statistically significant difference in NR2A and NR1 receptor expression in both genders as compared to the control group (p> 0.05). NR2B expression was found to decrease significantly in the experimental group compared to the control group. Also in female experiment group memory performance was significantly decreased which supports the findings in receptor expression. The results indicate that exposure to synthetic food colors during reast feeding in offspring may lead to alterations of expression of nmdar su units which is related to learning process. these alterations differ according to gender.
