Osteoblastlar kemik yapımından sorumlu hücrelerdir. Ca+2 iyonu hücre içi ikincil haberci sisteminin önemli bir parçası olup fizyolojik birçok fonksiyonun yerine getirilmesinde önemli bir rol üstlenmektedir. [Ca+2]c miktarındaki artış, apoptozis ve serbest radikal düzeylerini de artırmaktadır. Fakat bu konu; osteonekroz gelişmesine sebebiyet veren zoledronik asit (ZA), bevasizumab (Bev), deksametazon (Deks) gibi ilaçların etki ettiği osteoblast hücrelerinde yeterince bilinmemektedir. Bu çalışmada ZA, Bev, Deks gibi ilaçların, kemik oluşumunda önemli görevi olan osteoblast hücrelerindeki kalsiyum sinyali, apoptoz ve oksidatif stres üzerine etkileri araştırılarak ve bu oluşturulan etkiye karşılık bir antioksidan olan selenyumun (Se) hücresel düzeyde oluşturduğu değişiklikler in vitro model üzerinde incelendi. Bu çalışmada insan osteoblast benzeri hücre serisi olan Saos-2 hücrelerini kullandık ve hücreleri pasajlanıp çoğaltılmalarının ardından; Kontrol, ZA, Bev, Deks, Se, ZA + Se, Bev + Se, Deks + Se, ZA + Deks, ZA + Bev, ZA + Deks + Se, ZA + Bev+ Se grupları olmak üzere 12 gruba ayırarak ilaçlar ile (ZA, Bev, Deks ve Se) 24 saat inkübe ettik. Sonuçlara bakıldığında; ZA, Bev, Deks, ZA+Deks ve ZA+Bev gruplarınının [Ca+2]c 'yi anlamlı olarak artırdığı görülürken, Se uygulamasının ise azalttığı tespit edildi. Benzer şekilde hücrelerdeki apoptoz, Kaspaz 3 ve Kaspaz 9, PARP-1 ve ROS üretim değerlerindeki ZA, Bev ve Deks kaynaklı artışların Se uygulaması ile azaltıldığı görüldü. Sonuç olarak, Selenyum tedavisi ile Zoledronik asit, bevasizumab ve deksametazondan etkilenmiş hücrelerde Ca+2 girişi, oksidatif stres ve apoptoz artışı azalmaktadır. Bu yüzden Zol, Bev ve Deks kaynaklı osteonekrozun etyolojisi ve tedavisinde Se tedavisinin etkinliğinin araştırılması gereken bir konu olduğu düşünülmektedir. Osteoblasts are the cells responsible for bone formation. Ca2+ is well known for its role as crucial second messenger in modulating many cellular physiological functions, Ca2+ overload is detrimental to cellular function and may present as an important cause of cellular ROS generation and apoptosis. But this topic is not well known in the osteoblast cells that have the effect of drugs such as zoledronic acid(ZA) ,bevacizumab(Bev), dexamethasone(Dex). In this study, effects of drugs such as zoledronic acid,bevacizumab,dexamethasone in the important task of osteoblast cells in bone formation on calcium signal , apoptosis and oxidative stres will be investigated and this corresponds to the effect generated selenium , an antioxidant formed at the cellular level changes will be examined in vitro models. In this study, we used the human osteoblast-like cell line as Saos-2 and after cells were passaged and multiplied, they were divided into 12 groups as Control, ZA, Bev, Dex, Se, ZA + Se, Bev + Se, Dex+ Se, ZA+ Dex, ZA + Bev, ZA + Bev +Se, ZA+Dex+ Se which were incubated with drugs(ZA, Bev, Dex and Se) for 24 h. According to the results; ZA, Bev, Dex, ZA + Dex and ZA + Bev groups significantly increased [Ca2+]c while Se application was found to decrease. . However, ZA, Bev and Dex-induced increase in apoptosis, caspase 3, caspase 9, poly (ADP-ribose) Polymerase 1 and intracellular reactive oxygen species production values in the cells were decreased by Se treatments. In conclusion, increase of oxidative stress, apoptosis and Ca2+ entry in the ZA, Bev and Dex-induced cells were decreased by Se treatment. Therefore, the efficacy of Se treatment in ZA, Bev and Dex-induced osteonecrosis etiology and treatment is thought to be a subject to be investigated.
