Kolorektal kanser dünyada üçüncü en yaygın teşhis edilen kanser ve kansere bağlı ölüme neden olan dördüncü kanser türü olması nedeniyle önemli bir halk sağlığı sorunudur. Son yıllarda, kemoterapötik maddeler ve alternatif bitkisel tedavi yöntemleri kolon kanseri tedavisinde önem kazanmıştır. Kurkumin, Curcuma longa bitkisinin etkili bir özütüdür ve birçok önemli etkisinin yanında kemoterapatik fonksiyona sahiptir. Kurkumin apoptozun indüklenmesi yoluyla anti-kanser etkilerini göstermektedir. İncelediğimiz literatürler ışığında, insan kolon kanseri hücre hattı üzerinde, kurkuminin uygun doz kullanımı ve zaman aralığını belirleyip immünohistokimyasal olarak ve gen ekspresyonu seviyesindeki apoptotik etkisini araştırmayı amaçladık. Kurkumin'in spesifik hedefi olarak bilinen 21 apoptotik geni seçtik ve kültür ortamında kolon kanseri üzerindeki aktivitesini belirledik. Çalışmamızda, CCL-228-SW 480 insan kolon adenokarsinoma hücre dizisi, uygun üretim koşullarında çoğaltılmıştır. Etkili kurkumin konsantrasyonu, deney grupları için 24 saat 50 uM, 48 saat 75 uM ve 72 saat 100 uM olarak belirlendi. Uygulanan dozların proliferasyon ve apoptoz için etkinliği, hücre canlılık testi, Brdu, TUNEL ve ayrıca Quantitive Real Time Polimeraz Zincir Reaksiyonu ile kurkumin uygulanmayan kontrol grubu ile kıyaslanarak analiz edildi. Sonuç olarak, kurkumin gruplarında kontrol grubuna kıyasla hücre canlılığı ve çoğalmasında belirgin bir azalma olduğunu ve doz ve zamana bağlı olarak apoptotik oranının arttığını bulduk (p˂0,05). Çalışmamızın, kolon kanseri tedavisinde kurkuminin klinik kullanımının geliştirilmesi adına yapılacak olan diğer çalışmaları destekleyeceğine inanıyoruz. Colorectal cancer is an important public health issue because of it is the third most commonly diagnosed cancer in the world and the fourth cancer type that causes cancer-related deaths. In recent yaers, chemotherapeutic agents and alternative herbal treatment methods have gained importance for colon cancer treatment. Curcumin is an effective extract of the plant Curcuma longa and has a chemoterapatic function as well as many other important effects. Curcumin shows anticancer effects mainly through induction of apoptosis. In the light of the literatures, we aimed to investigate the apoptotic effect of curcumin immunohistochemically and at the level of gene expression, by determining the appropriate dose and time interval, on SW-480 colon adenocarsinoma cell line. We choose to study 21 apoptotic genes known as the specific target of curcumin and determine its activity on colon cancer in culture medium. In our study, CCL-228-SW 480 human colon adenocarcinoma cell line was replicated in appropriate production conditions. Effective curcumin concentration was determined as 24 h 50 µM, 48 h 75 µM and 72 h 100 µM for experimental groups. The efficacy of the administered doses for proliferation and apoptosis, were analyzed by cell viability test, Brdu, TUNEL and also Quantitive Real Time Polymerase Chain Reaction with comparison to nontreated control group. As a result, we found that in the curcumin groups, there was a significant decrease in cell viability and proliferation compared to the control groups and there was an increase in apoptotic ratio depending on dose and time (p˂0,05). We believe that our study will support the other study for the development of clinical use of curcumin in the treatment of colon cancer. Keywords : Colon adenocarsinoma, curcumin, in-vitro.
