Prepubertal Dönem Süresince Yavru Sıçanlarda Monosodyum Glutamat'a Maruziyetin Öğrenme ve Nörodavranış Üzerine Etkileri MSG işlenmiş gıdalarda lezzet arttırıcı olarak kullanılan bir gıda katkı maddesidir. Yapılan birçok çalışmada MSG'nin MSS üzerine olan olumsuz etkileri gösterilmiştir. Biz de çalışmamızda çocukluk döneminde MSG tüketiminin öğrenme, hafıza ve bazı nörodavranışsal parametreler üzerine etkilerini araştırdık. Çalışmamızda anne sütünden ayrılmış 4 haftalık 66 adet sıçan kullanılmıştır. Sıçanlar eşit sayıda erkek ve dişi içeren 3 gruba ayrılmıştır (K, D1, D2). Oral gavaj yoluyla 6 hafta boyunca D1 grubundaki sıçanlara 25 mg/kg/gün, D2 grubundakilere 2,5 g/kg/gün dozunda MSG uygulanırken, K grubundakilere aynı hacimde içme suyu uygulanmıştır. Daha sonra sıçanların Morris Su Labirenti Testi, Açık Alan Testi ve Zorlanmış Yüzme Testindeki performansları değerlendirilmiştir. Bu testlerden sonra sakrifiye edilen sıçanların hipokampüslerinden Western Blot yöntemiyle NR2A, NR2B, 5-HT2A ve nAChR α7 reseptörlerinin konsantrasyonları ölçülmüştür. Morris Su Labirentinde D2 grubunda öğrenme hızının kontrol grubuna göre yavaş olduğu, 'Açık Alan testi'nde şahlanma sayısı ve merkez kareye giriş sayısının D2 grubunda K grubuna göre yine anlamlı düzeyde düşük olduğu saptanmıştır. Zorlanmış yüzme testinde ise D1 ve D2 grubunda hareketsiz geçirilen sürelerin anlamlı düzeyde arttığı bulunmuştur. Bu veriler MSG'nin nörodavranışsal çaresizlik ve anksiyete yarattığını desteklemektedir. Çalışılan reseptörlerden NR2A konsantrasyonunda her iki cinsiyette D2 gruplarında K grubuna göre anlamlı bir artış gözlenmiştir. Çalışmamızda yüksek dozda MSG'nin öğrenme hızı ve nörodavranışsal çaresizlik üzerine etkilerinin olduğu ve NR2A reseptöründeki artışın eksitotoksisite ile ilişkili olarak bu verilerle ilişkilendirilebileceğini düşünmekteyiz. Diğer taraftan nAChR α7 ve 5-HT2A düzeylerinde cinsiyetler arasında anlamlı farklılıklar olması MSG'nin dişi ve erkek cinsiyette farklı etkiler yaratabileceğini ortaya koymuştur. Effects of Monosodium Glutamate Exposure During Prepubertal Term on Learning and Neurobehaviour in Young Rats MSG is a food additive used as a flavor enhancer in processed foods. Many studies have shown the negative effects of MSG on the central nervous system. We have investigated the effects of MSG consumption in childhood period on learning, memory and some neurobehavioral parameters. Sixty-six post-weaning 4 weeks old rats were used in our study. The rats were divided into 3 groups including equal numbers of male and female. MSG was administered at 25 mg/kg/day for the rats in group E1 and 2.5 g/kg/day for the group E2 for 6 weeks by oral gavage, while the same volume of tap water was applied to the rats in the C group. Later, the performances of the animals in the Morris Water Maze Test, Open Field Test and Forced Swim Test were evaluated. After these tests, concentrations of NR2A, NR2B, 5-HT2A and nAChR α7 receptor concentrations were analyzed by Western Blot method from the hippocampi of the rats sacrificed. In the Morris Water Maze Task, it was found that the learning speed was slower in the D2 group compared to the control group, and the number of rearings in the open field test and the number of the central square entries were significantly lower than the C group in the E2 group. In the Forced Swim Test, it was found that immobility times in E1 and E2 were significantly increased compared to C group. These data support that MSG creates neurobehavioral despair and anxiety. There was also a significant increase in the concentration of NR2A receptors in the E2 groups of both sexes compared to the C groups. In our study, we suggest that high doses of the MSG affect learning and neurobehavior, and this may be associated with excitotoxicity, through the increase of the NR2A receptors. On the other hand, significant differences between sexes in nAChR α7 and 5-HT2A levels have shown that MSG can produce different effects in female and male genders.
