Epilepsi oluşumumunda hücre içi Ca+2 artışı ve aşırı oksidatif stresin rolü iyi bilinmektedir. TRPM2 kanalı oksidatif stres ile aktive olan Ca+2 geçirgen bir kanaldır. Epilepsi de reaktif oksijen türleri (ROS) ile aktive olan TRPM2 kanalı hastalığın oluşumunda rol oynayabilir. Bu çalışmada, TRPM2 blokeri N-(p-amylcinnamoyl) antranilik asit (ACA)’ın pentilen tetrazol (PTZ) ile epilepsi oluşturulan sıçanların hipokampus TRPM2 aktivasyonu, oksidatif stres ve apopitozis değerleri üzerinde etkileri araştırıldı. Sıçanlar kontrol, PTZ, ACA ve ACA+PTZ olarak dört gruba ayrıldı. Periton içi PTZ (60 mg/kg) verilerek, PTZ ve PTZ+ACA grupları epilepsi oluşturuldu ve 30 dakika süre ile epilepsi atak geçirme kayıtları alındı. ACA gruplarında PTZ veya plasebo verilmeden 2 saat önce periton içi ACA enjeksiyonları (25 mg/kg) yapıldı. PTZ enjeksiyonundan 30 dakika sonra tüm gruplardan hipokampus ve beyin korteksi örnekleri alındı. Hücre içi serbest Ca+2 yoğunluğu, TRPM2 akım yoğunlukları, hücre içi ROS üretimi, mitokondriyal zar depolarizasyonu, apopitozis, kaspaz 3, kaspaz 9 ile PARP1 ekspresyon düzeyleri PTZ grubunda, kontrol grubuna kıyasla belirgin bir şekilde yüksekti. Hipokampus ve beyin hücre canlılığı, lipit peroksidasyon, indirgenmiş glutatyon, glutatyon peroksidaz, vitamin A, vitamin E ve beta karoten düzeyleri ile epilepsi atak geçirme süreleri PTZ grubunda, kontrol grubuna kıyasla belirgin bir şekilde düşüktü. Bütün bu değerler, ACA tedavisi ile düzeldi. ACA tedavisi ile atak geçirme süreleri de uzadı. Sonuç olarak, literatürde ilk kez epilepsi oluşumunda TRPM2 kanalından Ca+2 girişinin önemli olduğu ve hipokampus mitokondriyal oksidatif stres ile apopitozise neden olduğu gözlenirken, TRPM2 inhibisyonun bu değerler üzerinde düzenleyici rolü gözlendi. Calcium ion (Ca2+) accumulation into the cytosol of hippocampus through oxidative stress seems to be a main reason in the etiology of epilepsy. TRPM2 channel is a cation-permeable Ca2+ channel and high expression level of the channel was found in the hippocampus. Although previous studies implicate reactive oxygen species (ROS) dependent channel activation in the generation of epilepsy, suppression of ongoing seizures by TRPM2 antagonist (N-(p-amylcinnamoyl) antranilik asit (ACA) has not yet been clarified. We tested the effects of TRPM2-specific antagonist (ACA) on the modulation of Ca2+ accumulation, apoptosis and anticonvulsant properties in hippocampus of pentylenetetrazole (PTZ) and ACA administrated rats. All rats were divided into four groups as control, PTZ, ACA and ACA+PTZ. Epilepsy was induced in the PTZ and ACA+PTZ groups by intraperitoneal injection of PTZ (60 mg/kg) and then epilepsy records were taken within 30 minutes. ACA (25 mg/kg) was administrated to ACA and ACA+PTZ groups 2 hours before the injections of PTZ. Brain cortex and hippocampus samples were taken. PTZ administration increased intracellular free Ca2+ concentrations, TRPM2 current densities, mitochondrial membrane depolarization, intracellular ROS production, apoptosis, PARP1, caspase 3 and 9 expression values, although the values were reduced in the ACA+PTZ group by ACA treatment. In the hippocampal neuron and brain, cell viability, lipid peroxidation, reduced glutathione, glutathione peroxidase, vitamin A, vitamin E and beta carotene values were decreased by the PTZ administration. However, the values were modulated by the ACA treatment. Latency time was also extended by application ACA. In conclusion, these results support a role for TRPM2 channels in inhibition of apoptosis, epileptic seizures, mitochondrial oxidative stress, and calcium accumulation, indicating that TRPM2 inhibition may possibly be a novel target in the hippocampus for prevention of epileptic seizures.
