Çeşitli Fiziksel ve Çevresel Faktörlerin Erwinia carotovora subsp. carotovora'daPektat Liyaz, Selülaz ve Proteaz Ekzoenzimlerinin Sentezine EtkilerininİncelenmesiFitopatojen bir bakteri olan Erwinia carotovora subsp. carotovora (Ecc)'nın GBIsuşu pektat liyaz'ın izoformları, sellülaz ve proteaz gibi çeşitli ekzoenzimlerüretebilme yeteneğine sahiptir. Ecc'de ekzoenzim üretimi bir global regülatörmekanizma tarafından kontrol edilir.Bu çalışmada, sıcaklık, oksijen limitasyonu ve farklı karbon kaynakları gibi çeşitliçevresel faktörlerin ekzoenzim üretimi üzerindeki etkileri araştırılmıştır. 30ºC'deekzoenzim üretimi oldukça yüksek değerlerde ölçülürken, sıcaklık 34°'yeyükseltildiğinde ekzoenzim üretiminde düşüş olduğu, 37ºC derecede ise ekzoenzimüretiminin oldukça azaldığı tespit edilmiştir. Kullanılan karbon kaynaklarının farklıolması da ekzoenzim üretiminde farklılıklara yol açmıştır. Örneğin glikozluortamlarda ekzoenzim üretimi gliserol bulunan ortamlara oranla daha fazladır.Aynı şekilde oksijen-limitasyon farklılıklarının ekzoenzim üretimi üzerindeki etkileride bu çalışmada gösterilmiştir. Oksijenli ortamlarda ekzoenzim üretimi yüksekdeğerlerde ölçülmüştür. Bunun aksine oksijen ortamı kısıtlanmış Erwinia suşlarının ekzoenzim üretiminin çok az olduğu tespit edilmiştir.Bu çalışma ile ekzoenzim üretiminin sadece genetik faktörlerin kontrolü altındaolmadığı, aynı zamanda çevresel faktörlerin de ekzoenzim üretiminde rol oynadığıgösterilmiştir. The analysis of effects various physiological and environmental factors onpectate lyase, cellulase and protease exoenzymes synthesis in Erwinia carotovorasubsp. carotovoraStrain GBI of a phytopathogen bacteria, Erwinia carotovora subsp. carotovora(Ecc), has to ability to produce various exoenzymes such as isoforms of pectatelyase, cellulase, protease. Exoenzyme production in Ecc is controlled by a globalregulator system.In this study effect of various environmental factors, such as temperature, oxygenlimitation and different carbon sources on exoenzymes production were investigated.While production of exoenzyme at 30ºC is measured at quite high levels, it wasdetermined that there was a decrease in the production of exoenzyme when thetemperature was brought up to 34ºC, that the production of exoenzyme highlydiminished at 37ºC. The fact that carbon sources used are different gave rise to somedifferences in the production of exoenzyme. For example, the production ofexoenzyme in glucose media is more than in the glycerol media is.In the same way, the effect of oxygen-limitation differences on the production ofexoenzyme was indicated in this study. The production of exoenzyme in oxygenmedia is measured at high levels. Contrary to this, it was determined that strains of Erwinia whose oxygen media is limited, production exoenyme rates is ratherinsufficient.By this study, it was shown that the production of exoenymes is not only undercontrol of genetic factors, but environmental factors affect a role in the production ofexoenzyme as well.
