Özet: Antalya ve civarından 2003 yılında toplanmış olan 2, 21 ve 24 numaralı Tetranychus urticae Koch. populasyonlarının sırasıyla clofentezine, fenpyroximate ve amitraz ilaçlarıyla ilaçlama kulesi-petri kabı yöntemi ile LC değerleri belirlenmiştir. 2, 21 ve 24 numaralı populasyonlara sırasıyla clofentezine, fenpyroximate ve amitraz ile her seleksiyon populasyonu için belirlenen LC60 değerleriyle 12, 14 ve 15 kez seleksiyon yapılmıştır. Elde edilen dirençli populasyonlar sırasıyla CLOF 12, FPY 14 ve AMT 15 olarak isimlendirilmiştir. Seleksiyon sonrası bu populasyonların hassas (GSS) populasyona göre direnç oranları sırasıyla 105,275 kat, 64,4574 ve 79,183 kat olarak belirlenmiştir. Dirençli populasyonlara 3 farklı sinerjist (Piperonyl butoxide (PBO), Triphenyl Phosphate (TPP) ve Iprobenfos (IBP) ) + ilaç uygulanmıştır. CLOF 12 populasyonunda clofentezine'e IBP ve TPP sinerjistlerinin etkisi görülmemiştir. PBO 1.267 oranında sinerjistik etki göstermiştir. FPY 14 ve AMT 15 populasyonlarında her üç sinerjist de fenpyroximate ve amitraz'ın etkisini artırmıştır. Direnç kazandırılan bu populasyonların diğer bazı ilaçlara direnç geliştirip geliştirmediği de incelenmiştir. Her 3 populasyonda da en yüksek çoklu direnç oranı Abamectin'e karşı belirlenmiştir. CLOF 12 populasyonunda Bifenthrin'e hassasiyet belirlenmiştir. FPY 14 ve AMT 15 populasyonlarında ise yanlıca Bifenthrin'e karşı hassasiyet belirlenmiştir. Resiprokal çaprazlama çalışmalarında CLOF 12, FPY 14 ve AMT 15 populasyonlarının direnç kalıtımının dominant karakterli olduğu belirlenmiştir. Esteraz (EST) enziminin poliakrilamid jel elektroforez (PAGE) yöntemiyle incelenmesi sonucu elde edilen verilere göre her üç dirençli populasyonun da selekte edildiği ana populasyona göre EST bantlarında yoğunluk tespit edilmiştir. Mikroplate reader ile yapılan kinetik okumalarda ise üç populasyonun da selekte edildikleri populasyonlara göre EST ve gluthation - S - transferase (GST) aktivitelerinde artış belirlenmiştir. Abstract: Populations of Tetranychus urticae Koch numbered 2, 21 and 24 were collected from Antalya province in 2003. LC degrees of the populations were detected respectively against clofentezine, fenpyroximate and amitraz by spray tower - petri dish method. These populations selected by LC60 of clofentezine, fenpyroximate and amitraz, 12, 14, 15 times respectively, were named respectively as CLOF 12, FPY 14 and AMT 15. After selections, resistant populations were respectively obtained to have 105,275, 64,4574 and 79,183 fold resistance. Different synergists (Piperonyl butoxide (PBO), triphenyl phosphate (TPP) and iprobenfos (IBP) ) + acaricide were used against resistant populations. No sinergistic effect was detected against clofentezine in CLOF 12 population with IBP and TPP. However, PBO was determined to have 1,267 fold sinergistic effect on this population. Effects of fenpyroximate and amitraz on populations were increased by all three synergists on FPY 14 and AMT 15 populations. In this study, multiple resistance of other some pesticide against these three populations was also detected. Higher multiple resistance was detected in three populations against abamectin. CLOF 12 population was provided to have susceptibility against bifenthrin. FPY 14 and AMT 15 populations were susceptible only against bifenthrin. Resiprocal crosses studies showed that resistance inheritance of CLOF 12, FPY 14 and AMT 15 populations was dominant character. Esterase (EST) enzyme activity was detected by polyacrylamide gel electrophoresis method (PAGE). After this study all three resistant populations showed dark EST bants, when compared to first selected populations. Kinetic read data obtained by microplate reader showed that EST and gluthation - S - transferase (GST) enzyme activities of three resistant populations increased when compared to first selected populations.
