Bu çalışmada gerek normal gerekse tuzlu koşullar altında çimlendirilen arpa (Hordeum vulgare L. cv. Tokak 157/37) tohumlarının kök uçlarında mitotik indeks, kromozom davranışları ve hücre döngüsü üzerine hidrojen peroksit (H2O2)'in etkileri araştırılmıştır. A. Mitotik indeks üzerine H2O2'in etkileri; 1. 20 C'de saf su ortamında elde edilen sonuçlar Normal koşullar altında çimlendirilen arpa tohumlarının kök uçlarındaki mitotik indeks üzerinde H2O2 ön uygulaması dikkate değer bir etkiye neden olmamıştır. 2. 20 C'de tuzlu koşullar altında elde edilen sonuçlar a. Tuz, arpa tohumlarının kök ucu mitotik indeksini konsantrasyon artışına paralel olarak engellemiştir. b. Çalışılan tüm tuz seviyelerinde H2O2 ön uygulaması, mitotik indeks üzerinde, kendi kontrolleriyle kıyaslandığında olumlu ya da olumsuz bir etkiye neden olmamıştır. B. Kromozom davranışları üzerine H2O2'in etkileri; 1. 20 C'de saf su ortamında elde edilen sonuçlar Normal koşullar altında çimlendirilen arpa tohumlarının kök uçlarındaki kromozom davranışları üzerinde H2O2 ön uygulamasının, kontrol grubuna göre çeşitli tiplerde kromozom anormalliklerine sebep olduğu tespit edilmiştir. H2O2 ön uygulaması, kromozom anormallik yüzdesini de kontrole göre arttırmıştır. 2. 20 C'de tuzlu koşullar altında elde edilen sonuçlar a. Tuz, konsantrasyon artışına paralel olarak kromozom anormallik yüzdesinde artışa neden olmuştur. b. Çalışılan H2O2 ön uygulaması, 0.35 ve 0.40 m tuzlulukta kromozom anormallik yüzdesini artırıcı bir etki göstermiştir. Tüm tuz seviyeleri dikkate alındığında en olumsuz etkiyi, 0.40 m NaCl'de ortaya çıkarmıştır. 0.45 m tuzlulukta ise hücre bölünmesi tamamen engellendiğinden dolayı kromozom anormallikleri incelenememiştir. C. Hücre döngüsü üzerine H2O2'in etkileri; 1. 20 C'de saf su ortamında elde edilen sonuçlar H2O2 ön uygulamalı arpa tohumlarındaki hücre döngüsünde G0- G1 safhasının yüzdesi, saf su ön uygulamalı tohumlara göre önemli derecede bir artış gösterirken, S safhasının yüzdesi dikkate değer bir azalma göstermiştir. 2. 20 C'de tuzlu koşullar altında elde edilen sonuçlar a. Saf su ön uygulamasından sonra NaCl'lü ortamda çimlendirilen arpa tohumlarındaki hücre döngüsünün G0- G1 ve S safhalarının yüzdelerinde, konsantrasyon artışına paralel olarak bir artış olmuştur. G2- M safhasına ait hücre populasyonunu bulunmamıştır. b. H2O2 ön uygulamalı arpa tohumlarında ise, tüm tuz konsantrasyonları kendi kontrol gruplarına göre G0- G1 safhalarının yüzdesini arttırırken, S safhalarının yüzdesini düşürmüştür. Tuz konsantrasyonlarında, G2- M safhasına ait hücre populasyonunu bulunmamıştır. Ayrıca H2O2 ön uygulamalı tohumlarda hücre boyutunu ifade eden CV değerinin, saf su ön uygulamalı tohumlara göre konsantrasyon artışına paralel olarak tüm tuz seviyelerinde arttığı gözlenmiştir. In the comparison to their self- controls, the pre- treatment of H2O2 had no negative or positive effect on mitotic index in all of the salt levels studies. B. Effects of H2O2 on chromosome behaviours 1. The results obtained at 20 C and in the background of distilled water. It was determined that H2O2 pre- treatment caused chromosome abnormalities in various types according to control groups on chromosome behaviours in root tips of barley seeds germinated under normal conditions. Also, H2O2 pre-treatment were increased chromosome abnormality percentage according to control. 2. The results obtained at 20 C and in salinity conditions a. In parallel with concentration rise, salt were increased on chromosome abnormality percentage. b. It was determined that H2O2 pre- treatment studied were increased chromosome abnormality percentage in 0.35 and 0.40 m salinity. When all salt levels were considired, the most positive effect were exhibited by 0.40 m NaCl. Chromosome abnormality could not be investigated because of 0.45 m salinity completely inhibited cell division. C. Effect of H2O2 on cell cycle 1. The results obtained at 20 C and in the background of distilled water It was determined that G0- G1 phase percentage of the cell cycle of barley seeds with H2O2 pre- treatment had an important rise according to control group while S phase percentage were decreased. 2. The results obtained at 20 C and in salinity conditions a. In parallel with concentration rise, G0- G1 and S phase percentage in cell cycle of barley seeds germinated in NaCl after distilled water pre- treatment were increased. Cell population belonging to G2- M phase weren't discovered. b. G0- G1 phase percentage in barley seeds with H2O2 pre- treatment were increased according to own control groups of all salt concentration while S phase percentage were decreased. In salt concentrations, cell population belonging to G2- M phase weren't obtained. In addition, CV value in seeds with H2O2 pre- treatment were increased according to seeds with distilled water pre- treatment in parallel with concentration rise all of salt levels.
