Tüm dünyada turunçgil yetiştirilen alanlarda yaygın olarak görülen Turunçgil tristeza virüsü (Turunçgil tristeza virüs=CTV) çok önemli ekonomik kayıplara neden olmaktadır. Kullanılan anaç ve kaleme bağlı olarak konukçusunda yaprak damar açılmasından bitkinin ölümüne kadar farklı belirtiler meydana getirmektedir. Son yıllarda dünyada CTV nün molekül er karakterizasy onuna yönelik çok sayıda araştırma yapılmıştır. Ancak ülkemizde CTV'nün varlığı 1980'li yıllardan bu yana bilinmekle birlikte moleküler karakterizasyonuna yönelik çalışmalar henüz yapılmamıştır. Bu çalışmada Türkiye'de turunçgil yetiştiriciliğinin yaygın olarak yapıldığı Doğu ve Batı Akdeniz, Ege sahil şeridi, Edremit Körfezi ve Doğu Karadeniz bölgelerindeki turunçgil üretim alanlarında bulunan ve serolojik ve biyolojik özellikleri belirlenen CTV izolatlarının 15 tanesi seçilerek moleküler özellikleri belirlenmiştir. Bu amaçla öncelikle farklı turunçgil üretim bölgelerinden seçilen 15 CTV izolatını içeren doku örneklerinden total RNA izolasyonu yapılmıştır. Daha sonra bu izolatların kılıf protein genleri iki aşamalı ters transkripsiyon polimeraz zincir reaksiyonu (RT-PCR) yöntemiyle çoğaltılarak tüm izolatlardan 672 bp büyüklüğünde bir bant elde edilmiştir. RT-PCR yöntemiyle çoğaltılan CTV kılıf protein genleri T-A klonlama yöntemiyle pGEM-T Easy plazmit vektörüne klonlanıp Escherichia coli bakterisine aktarılarak çok sayıda koloni elde edilmiştir. Her bir izolat için elde edilen beyaz kolonilerden en az beş tanesi seçilerek koloni PCR yöntemiyle taranmış ve her bir izolatın kılıf protein genini içeren en az iki beyaz koloni belirlenmiştir. Bu kolonilerden plazmid izolasyonu yapıldıktan sonra plazmidler EcoRI enzimiyle kesilip CTV kılıf protein geni içerdikleri teyit edilmiştir. Daha sonra plazmitlere klonlan CTV kılıf protein genlerinin dizilimi yapılarak bunların nükleotid dizilimi ve kodladığı amino asit dizilimi belirlenmiştir. Kılıf protein genlerinin nükleotid ve amino asit dizilimleri analiz edilerek ülkemizin farklı bölgelerinden elde edilen CTV izolatların birbirleriyle ve dünyadaki diğer izolatlarla benzerlik oranları ve fılogenetik ilişkileri ortya konmuştur. Citrus tristeza virus (CTV), widespread in all citrus growing areas of the world, causes significant economical losses. CTV induces different symptoms ranging from mild vein clearing to the death of citrus trees depending on the combination of the rootstock and scion used. Recently a number of studies have been conducted about molecular characterization of CTV in the world. However, even though the existence of CTV in Turkey has been known since 1980s, the molecular characteristics of CTV isolates have not been studied. In this study, 15 CTV isolates collected from different citrus growing region of Turkey including Eastern and Western Mediterranean, Aegean Cost, Edremit Gulf, Eastern Black Sea regions and previously characterized by serological and biological assays were selected and their molecular characteristics were determined. First, total RNA was isolated from citrus bark tissue collected from CTV infected trees from different regions. Then the coat protein (CP) gene of these isolates were amplified by a two-step reverse transcription-polymerase chain reaction (RT-PCR) method and a DNA fragment of 672 bp corresponding to the CP gene of CTV was obtained from all isolates. The CP genes amplified by RT-PCR were cloned into the pGEM-T Easy plasmid vector using T-A cloning method. After the transformation of then the Escherichia coli a number of colonies potentially carrying the CTV CP genes were obtained. For each isolate at least five white colonies were screened by colony PCR and at least two colonies carrying the pGEM-T Easy plasmid with the CTV CP gene were identified. Plasmids were isolated from these colonies and of the presence of CTV CP gene in the plasmids was confirmed by EcoRI digestion. Then the CTV CP genes cloned in tho the pGEM-T Easy plasmid were sequence and their nucleotide and deduced amino acid sequences were determined. Similarity and phylogenetic relationship of the CP gene of Turkish CTV isolates with each other and with the CP genes of CTV isolates from different regions of the world were determined using the nucleotide and amino acid sequences.
