Bu tez çalışmasında, ülkemizin dış ticaretinde önemli yeri olan fındık ve büyük bir potansiyele sahip kırmızı pul biberdeki aflatoksin sorununun UV-A ışını ile tahribatsız ve hızlı görüntüleme yöntemleri ile tespiti ve UV-C ışınının Aspergillus flavus ve Aspergillus parasiticus'un aflatoksijenik suşları üzerine antifungal etkisi incelenmiştir. Kırmızı pul biber görüntüleme sisteminde iki yöntem kullanılmış ve aflatoksin bulaşması bölgesel özellikler kullanılarak % 92,8 ve genel özellikler kullanılarak %7 2,7 başarı oranıyla tespit edilebilmiştir. Fındık örneklerinde ise görüntüleme sonuçları bize doğrudan aflatoksin varlığını işaret edememiş, ancak aflatoksijenik küf suşlarının varlığı % 97,5'lik oranla görüntülerde tespit edilebilmiştir. Yine bu tez çalışmasında yapılan aşamalı UV-C uygulamaları aflatoksijenik küf suşlarında kontrol grubuna kıyasla 5,48 log'luk azalma meydana getirmiş ve UV-C ışınının bu suşlar üzerinde de antifungal özelliğe sahip olduğu kanıtlanmıştır. Bu etkinin sebebinin küflerin protein yapılarının bozulması olduğu proteinlerinin SDS-PAGE ile incelenmesi sonucu ortaya konulmuştur. In this study, there have been two major objectives. First, to develop rapid and non-invasive aflatoxin detection method based on machine vision technology for red peppers and hazelnut both of which have important value for Turkey's export. Secondly, inhibition effect on aflatoxigenic Aspergillus flavus and Aspergillus parasiticus by use UV-C treatment. In red peppers' imaging system was two methods used, and aflatoxin contamination was detected at rate of 92.8 % successfully by using local feature and at a rate of 72.7 % successfully by using global feature. With hazelnut samples, aflatoxin was not detected directly. However, aflatoxigenic molds were detected at rate of 97.5 %. Consequently, hazelnuts which contained mold were seperated from uncontaminated. Thus, can be reduced to Turkey's aflatoxin problem by this study. A 5.48 log reduction was observed when hazelnuts were exposed to UV-C. Thus, we concluded that UV-C has antifungal effect over aflatoxigenic molds.
