Dünyada en sık görülen enfeksiyon kaynaklarından biri olan metisiline dirençli S. aureus susu (MRSA), antimikrobiyallere karşı geliştirdiği direnç mekanizmaları ile her geçen gün daha büyük bir sorun haline gelmektedir. Bu tezin amacı S. aureus susunda metisiline dirençte rol oynadığı düşünülen ve fem faktörleri olarak adlandırılan genlerden femA, femB, femX genlerinin varlığının PZR metodu ile araştırılmasıdır. Çalışmamızda Süleyman Demirel Üniversitesi Araştırma ve Uygulama Hastanesi'nin çeşitli kliniklerinden izole edilen 34 adet S. aureus susuna koagülaz testi uygulandı. Koagülaz testi sonucu S. aureus oldukları bir kez daha doğrulanan örneklerde PZR metodu ile femA, femB vefemX genlerinin varlığı araştırıldı. Çalışmanın sonunda 34 adet susun 30'unda femX geni pozitif olarak gözlenirken, suşların 4 tanesinde bu gene rastlanmadı. Yine bu 34 adet susun 27'sinde/e/nA geni gözlenirken, 7 tanesinde bu gen gözlenmedi. femB geni İse 34 Örnekten 14'ünde pozitif olarak tespit edilirken, 20'sinde negatif olarak gözlenmiştir. Methicillin resistant Staphylococcus aureus strain (MRSA), which is one of the most frequent cause of infections in the world, because of its improved resistance mechanism, has become a growing nuisance. The principal objective of this thesis is the investigation of the femA, femB, femX genes, which are called/em factors and, are thought to have an effect on methicillin resistance in S. aureus. in the study, priraarily coagulase test was applied to a total of 34 S. aureus strains, vvhich are obtained from various clinics of Süleyman Demirel University Hospital, so as to confirnı that they are S. aureus. After coagulase test, by applying PCR method, the existence of femA, femB, femX genes were investigated in these strains that are confirmed to be S. aureus . As a result of this study; 30 femX strains out of 34 were found to be positive while the other 4 were negative. 27 femA out of 34 were proved to be positive unlike the other 7. 14 femB out of 34 were found to be positive while 20 strains were negative.
