Kefir danesi, kendine özgü bir mikrofloraya sahiptir. Bu mikroflora laktik asit bakterilerini (LAB), asetik asit bakterilerini ve mayaları belli oranlarda bulundurmaktadır. Kefir, kefir danesinin süt içinde fermantasyonu sonucunda elde edilen fermente bir süt ürünüdür. Probiyotik bakteri ve maya zenginliği itibariyle kefir, sağlık yönünden yararları bilinen doğal fermente bir süt ürünü olarak dikkat çekmeye başlamıştır. Kefirin, antikanserojenik özelliğinin varlığı, bağışıklık ve sindirim sistemini olumlu etkilemesi, laktoz intoleransına karşı etkinliği, kolesterol düşürücü ve rahatlatıcı etkileri yapılan araştırmalarla desteklenmiş ve bilinçli tüketicilerin, fonksiyonel ürünlere olan talebi kefire olan ilgiyi de arttırmıştır.Bu tez çalışmasının amaçları, ülkemizde bulunan kefir dane bakteri florasını moleküler teknikle tespit etmek, kefir fermantasyonu kontrollü atmosfer ortamında gerçekleştirilerek kefirin fonksiyonel özelliğini geliştirmek, kefir danesi kullanılarak üretilen kefir ile ana kültürden üretilen kefirin özelliklerini karşılaştırmak ve doğal kültür olarak kefir danelerinin endüstriyel kullanımı amacıyla optimizasyonunu sağlamaktır. Polimeraz Zincir Reaksiyonuna (PZR) dayalı 16S rRNA tekniği kullanılarak bakteri florası tespit edilmiştir. Örneklerin kimyasal özellikleri (pH, % laktik asit, % KM, % protein), tat-aroma bileşikleri (GC-tepe boşluğu yöntemi), ekzopolisakkarit miktarı (Dubois yöntemi), mikrobiyolojik (laktobasil, laktokok, Lactobacillus acidophilus, Bifidobacterium spp. ve mayalar), duyusal ve fiziksel (Brookfield reometre) özellikleri 21 gün depolama süresince tespit edilmiştir.Kullanılan spesifik primerler ile PZR ürünlerinin dizi analizi sonuçlarına göre probiyotik özelliğe sahip Lactobacillus acidophilus, Lactobacillus johnsonii, Lactobacillus kefiranofaciens, Lactobacillus casei, Lactobacillus helveticus, Lactobacillus jensenii, Lactobacillus delbrueckii, Lactobacillus reuteri, Lactobacillus brevis, Streptococcus thermophilus ve Bifidobacterium bifidum bakterilerinin ülkemizde üretilen kefir danesinin bakteri florasında doğal olarak bulunduğu PZR yöntemi ile tespit edilmiştir. Kefir danesinde Bifidobacterium bifidum uluslar arası düzeyde ilk olarak bu çalışmayla tespit edilmiştir. Araştırmada normal atmosferde kefir danesinden üretilmiş kefir örneği (KG), % 10 CO2 atmosferde kefir danesinden üretilmiş kefir örneği (KG-C), normal atmosferde kefir ana kültürü kullanılarak üretilen kefir örneği (K), % 10 CO2 atmosferde kefir ana kültürü kullanılarak üretilen kefir örneğinden (K-C) oluşan dört farklı uygulama bulunmaktadır. Kefir örneklerinin 1. gün laktobasil içerikleri 9,21-9,28 log kob/ml; laktokok içerikleri 9,23-9,27 log kob/ml değişim aralığında tespit edilmiştir. KG ve KG-C kefir örneklerinde 1. gün maya içerikleri 5,50 ve 5,53 log kob/ml; K ve K-C kefir örneklerinin maya içerikleri 4,77 ve 4,71 log kob/ml olarak tespit edilmiştir. KG, KG-C, K ve K-C kefir örneklerinin 1. gün L. acidophilus içerikleri sırasıyla 5,78; 6,43; 6,10 ve 6,42 log kob/ml olarak tespit edilmiştir. Kefir örneklerinin depolama süresince Bifidobacterium spp. içerikleri 3,19-6,14 log kob/ml değişim aralığında tespit edilmiştir. Dane veya kültür kullanımı ile farklı atmosfer uygulamasının kefir örneklerinin pH ve titrasyon asitliği üzerine önemli bir etkisi olmadığı bulunmuştur. Kefir örneklerinin depolama süresince pH değeri 4,29-4,53; laktik asit değerleri %0,81-0,95; %KM değerleri %7,86-8,21; protein değerleri %3,09-3,48 olarak bulunmuştur. Dane ve ana kültürden üretilmiş kefir örneklerinin 1. gün EPS içerikleri 79-130,15 mg/L aralığında değişmektedir. Kefir örneklerinde depolama süresince asetaldehit değişimi 3,81-23,58 mg/L; etanol içeriği 76,46-5147,70 mg/L değişim aralığındadır. Kefir örnekleri 1. gün viskozite değerleri 225-315 mPas değişim aralığında olup, farklı uygulamalarla üretilen kefir örneklerinin Newtoniyen olmayan akış gösterdiği ve pseudoplastik akış tipi olduğu saptanmıştır. Duyusal analiz bulgularında kefir örneklerinin 1. gün genel değerlendirme sonuçları 7,7-9,7 puan değişim aralığında ve en çok beğeni alan K-C örneği olarak tespit edilmiştir.Bu doktora tezi kapsamında özgün bulgular tespit edilmiştir. Öncelikle ülkemize ait kefir danelerinin ve bunlardan üretilen kefirin bakteri florası tespiti yapılmıştır. Kefir üretiminde farklı atmosfer uygulaması ilk olarak bu tez çalışmasında yapılmış, bu uygulamanın kefirde bulunan probiyotik bakterilerin gelişimini teşvik ettiği bulunmuştur. Kefir danesinden elde edilen ana kültürden üretilen kefir örnekleri genel olarak kimyasal, mikrobiyal, reolojik ve duyusal özellikleri olumlu olarak kefir danesinden üretilenlerle benzerlik göstermiştir; kefir danesine göre ana kültürün işletmeler tarafından kullanımı daha kolay olabilecektir. Endüstriyel üretimde sorun olabilecek bu probleme çözüm olarak, bu tez kapsamında ilk olarak kefir danesinin üretiminin yapılabileceği fermentörün tasarımı yapılmıştır.Bu doktora tez çalışmasıyla, kefir üretimine katkı sağlayacak, ulusal ve uluslar arası düzeyde literatürdeki boşlukları giderecek, fonksiyonel etkinliğinin en önemli kriteri olan mikroflorasının ve özellikle probiyotik içeriğinin belirlenmesiyle önemli bulgular tespit edilmiş ve değerlendirilmiştir. Kefir grain has a unique microflora. This microflora includes lactic acid bacteria, acetic acid bacteria and yeasts. Kefir is a fermented dairy product, which is a result of the fermentation of the kefir grains in milk. Kefir has started to draw more attention as a natural product with health benefits, mostly due to the presence of probiotic bacteria and yeast. Research has shown that kefir has anticarcinogenic and antimutagenic effects, improves the immune and digestive systems and lactose intolerance, and reduces cholesterol. Increased attention to the functional foods by conscious customers also increases the great interest in kefir.The aims of the thesis were to detect the bacterial flora of kefir grain using molecular techniques, to compare the properties of kefir samples produced using kefir grains and those produced from the mother culture (kefir produced from kefir grains was used as mother culture), and to provide an optimisation of kefir production with natural cultures obtained from kefir grains for the industrial scale processing. In addition, another significant purpose was to develop the functional properties of kefir using controlled atmosphere fermentation. Polymerase Chain Reaction (PCR)-based 16S rRNA was used to determine bacterial flora. The chemical properties (pH, lactic acid %, total solids %, protein %), taste-aroma compounds (with GC-headspace method), amount of exopollysaccharides (according to Dubois method), microbiological (Lactobacilli, Lactococci, Lactobacillus acidophilus, Bifidobacterium spp. and yeasts), sensory and textural (Brookfield Rheometer) analysis were carried out during 21 day storage period.According to the results of PCR products with the specific primers it was identified that Lactobacillus acidophilus, Lactobacillus johnsonii, Lactobacillus kefiranofaciens, Lactobacillus casei, Lactobacillus helveticus, Lactobacillus jensenii, Lactobacillus delbrueckii, Lactobacillus reuteri, Lactobacillus brevis, Streptococcus thermophilus and Bifidobacterium bifidum were the natural inhabitants of the kefir grains used in our country. The identification of Bifidobacterium bifidum in kefir grain was the first report at international level. Four different applications were considered: kefir produced from kefir grains in ambient atmosphere (KG), kefir produced from kefir grains in 10% CO2 amosphere (KG-C), kefir produced from mother culture in ambient atmosphere (K), kefir produced from mother culture in 10% CO2 amosphere (K-C). Lactobacillus and Lactococcus contents of kefir samples varied between 9,21-9,28 log cfu/ml and 9,23-9,27 log cfu/ml respectively, at the first day of storage. Yeast contents of KG and KG-C samples were 5,50 and 5,53 log cfu/ml, respectively, whereas K and K-C kefir samples had 4,77 and 4,71 log cfu/ml, respectively, at the first day of storage. Content of L. acidophilus were 5,78; 6,43; 6,10 and 6,42 log cfu/ml, respectively. Bifidobacterium spp. content of kefir samples were between 3,19-6,14 log cfu/ml during storage. It was determined that the use of different atmosferic conditions or the choice of grain/mother culture did not have a significant effect on pH and titratable acidity. During storage period pH, lactic acid %, total solids % and protein % contents of kefir samples were 4,29-4,53; 0,81-0,95 %; 7,86-8,21 % and 3,09-3,48 %, respectively. Kefir samples produced from the mother culture and from the grains had 79-130,15 mg/L EPS levels at the first day of storage. Acetaldehyde and ethanol contents of kefir samples were 3,81-23,58 mg/L; 76,46-5147,70 mg/L, respectively during storage. Viscosity values of kefir samples were between 225-315 mPas at first day. The flow type of all kefir samples were pseudoplastic, which is a non-Newtonian flow type. Based on the sensory analysis of kefir samples, overall evaluation score at first day were between 7,7-9,7 and K?C had the highest score.Unique findings were reported in this dissertation; the bacterial flora of kefir grains obtained from different locations of Turkey has been identified with using molecular techniques. For the first time, it was found that the application of 10% CO2 atmosphere during kefir fermentation supports the growth of probiotic bacteria. Kefir samples produced from mother culture affirmatively had similar chemical, microbiological, rheological and sensory properties with kefir samples produced from kefir grains; therefore, kefir mother culture obtained from kefir grains could be used in dairy plants, which is easier and more applicable than the maintenance of kefir grains in large scale. Based on the microbial and sensory results shelf life of kefir is suggested for 14 days. Kefir grains are propagated using traditional methods, which is not easy to replicate in industrial production. To address this issue, a design of a fermenter for kefir grain propagation is also provided for the first time in the literature.This doctoral thesis presents significant findings with regard to the evaluation of probiotic content and microflora of kefir, which are arguably the most important criteria for its therapeutic efficacy. These findings will provide significant contributions not only to the literature at international level also to the dairy indusrty.
