Bu tez çalışmasında Vibrio anguillarum ile deneysel olarak infekte edilerek oluşturulan vibriozis infeksiyonunun gökkuşağı alabalıklarında (Oncorhynchus mykiss) meydana getirdiği hematolojik değişimlerin incelenmesi amaçlanmıştır. Araştırmada 208.7ł8.11 gr. ağırlığında 100 adet gökkuşağı alabalığı kullanılmıştır. Bunlardan 50 adet balığa, mililitrede 100 adet/ml konsantrasyonunda Vibrio anguillarum bakterisi içeren inokulum periton içine 0.2 ml/balık dozunda enjekte edilerek infekte grup oluşturulmuştur. İnfeksiyonun 1, 2, 3, 4, 5, 7, 9, 11, 13 ve 15. günlerinde 5.er adet olmak üzere infekte ve kontrol grubu balıkların kuyruk venasından enjektörle alınan kan örnekleri eritrosit, trombosit ve lökosit sayıları, lökosit tipleri, hemoglobin, hematokrit, eritrosit indeksleri (MCV, MCH, MCHC), total plazma proteini, albümin ve kortizol miktarları yönünden incelenmiştir. İnceleme neticesinde infekte grup balıklarda eritrosit, lökosit, hemoglobin, hematokrit, albümin ve total plazma proteini değerlerinde istatistiki olarak önemli azalma görülürken, kortizol seviyesi ve trombosit sayısında ise istatistiksel olarak önemli artış tespit edilmiştir. Lökosit tiplerinden nötrofil ve monosit hücreler infekte grup balıklarda hastalığın seyrine bağlı olarak denemenin ilk günlerinde artış gösterirken, denemenin ilerleyen günlerinde kademeli olarak azalarak kontrol grubuna ait değerlere yaklaşmış; buna karşılık lenfosit hücreler ise denemenin başında azalırken, ilerleyen dönemde artış göstermiştir. Hastalığın ilk günlerinde lenfosit sayısında azalma, nötrofil sayısında artış olduğunun görülmesi hastalığın akut seyir gösterdiğini ortaya koymuştur. The aim of this study was to investigate haematological changes which caused vibriosis occured by experimentally infected with Vibrio anguillarum on rainbow trouts (Oncorhynchus mykiss). 100 rainbow trouts weighing 208.7ł8.11 gr were used for this research. Vibrio anguillarum at a concentration of 100 bacteria/ml was injected by intraperitoneally (i.p.) amount 0.2 mL to 50 fishes and infected group was occurred. The blood samples of five fishes each infected and control groups were collected from caudal vein in 1, 2, 3, 4, 5, 7, 9, 11, 13 and 15 days after the infection by injector and examined in terms of erythrocyte, platelet and leukocyte counts, leukocyte types, haemoglobin, haematocrit, erythrocyte indices (mean cell volume (MCV), mean cell haemoglobin (MCH), mean cell haemoglobin concentrations (MCHC)), total plasma protein, albumin, and cortisol levels. As a result of investigation, erythrocytes, leukocytes, haemoglobin, haematocrit, albumin and total plasma protein levels were decreased, however cortisol levels and platelet counts were increased in the infected groups. Neutrophil and monocyte cells were increased at the beginning of the experiment and were decreased in days following; on the other hand, lymphocytes were decreased at the beginning of the experiment and were increased in in days following depend on the disease process in the infected groups. Reduction in the number of lymphocytes and increase number of neutrophils in the early days of the disease was revealed that the disease was acute clinical course.
