Bu çalışmada, Süleyman Demirel Üniversitesi Anorganik Kimya Araştırma laboratuvarında yeni olarak sentezlenmiş bir grup Schiff Bazı Kompleksinin prostat kanserli hücre hatları olan PC-3 ve DU-145 üzerinde sitotoksik etkisi MTS canlılık testiyle belirlenmiştir. Kontrol grubu olarak kök hücre potansiyeli olan dental folikül hücreleri kullanılmıştır. Toksisite testlerinin sonucu 4 gün süreyle 24 saat aralıklarla monitörize edilmiş ve hücrelerin canlılık oranı spektrofotometrik olarak (490 nm) belirlenmiştir. Doxorubisin ve paklitaxel standart kemoterapik ajan olarak bu çalışmada kullanılmıştır. Bakır ve mangan metali içeren kompleks grubun diğer üyelerine göre daha etkindir. Test edilen Schiff baz türünün prostat kanserli hücre hatlarında zamana bağlı artış gösteren bir toksisiteye neden olduğu gözlenmiş ve bu hücre hatlarında 4 günlük inkübasyon sonucu %100 e varan hücre ölümleri saptanmıştır. Canlılık testi sonuçlarına göre en etkili konsantrasyon belirlenmiş ve bu doz kullanılarak hücrenin apoptoza gidip gitmediğini saptamak amacıyla apoptotik markerlar olan Anneksin-V ve p53 flow sitometri ile, p21 ve caspase 3 ise gercek zamanli PCR yöntemi ile belirlenmeye çalışılmıştır. Elde edilen sonuçlar bu çalışmada kullanılan Schiff bazının apoptozu indükleyerek hücre ölümüne sebep olduğunu göstermektedir. Devam eden çalışmalar ile test molekülünün diğer bazı kanser hatları ve in-vivo modeller üzerindeki tedavi potansiyelleri araştırılabilir. In this study, one group of Schiff Base complexes, newly synthesized in Süleyman Demirel Inorganic Chemistry Laboratory, were tested on PC-3 and DU-145 Prostate cancer cell lines for its cytotoxicity by using MTS proliferation assay. Dental follicle cells having stem cell potantial were used as control group in the tests. The results of cytotoxicity tests were monitored in 24 hours intervals for 4 days and the ratio of the viable cells was determined by spectrophotometry. Doxorubisin and paclitaxel was used as standard chemotherapic agents. The complex including copper and mangan was found more effective on these cancer cell lines than the other complexes. This Schiff base complex was seen to cause increase in cytotoxicity by time. In all cell lines, after 4 days incubation, nearly 100% of the cell were found to be dead. According to MTS results, the most effective dose was determined. Applyıng the Schiff base on the cells at this dose it was check if the cells are undergoing apoptosis. The results showed that Schiff base is inducing apoptosis on cancer cells which was confirmed by detection of Annexin-V and p53 by using Flow cyotmetry and caspase3 by using Real Time PCR. As a future remark, the Schiff base is planned to be test on other cancer cell lines and in-vivo models. Keywords: Schiff Base, prostate cancer, MTS, chemotherapic agent, cytotoxicity apoptosis.
