Bu tez çalışmasında, NSAID grubu bazı bileşikler ile klofibrik asit ve bezafibrik asidin, Gemini Phenyl C6 kolonda alıkonma özellikleri üzerine, pH ve organik modifiyer derişiminin etkisi incelenmiştir. Farklı çözelti karışımlarında, farklı pH.larda yapılan enjeksiyonlardan elde edilen alıkonma zamanları kullanılarak bileşiklere ait pKa değerleri ve bu çözelti karışımında asit-baz türlerinin alıkonma parametreleri elde hesaplanmıştır. Bu çözelti bileşimi ve özellikleri kullanılarak farklı modeller test edilmiş, mobil faz pH.sı ve bileşiminin alıkonma üzerine etkisi konusunda çeşitli yaklaşımlar yapılmıştır. Mobil faz pH.sının ve organik modifiyer oranının bileşiklerin kromatografik davranışı üzerine olan etkisi incelenmiştir. Suda çözünürlüğü düşük olan NSAID grubu bileşiklerin pKa değerleri, farklı pH.ya sahip mobil fazlardan elde edilen alıkonma değerleri kullanılarak hesaplanmıştır. Mobil faz optimizasyonu için alıkonma değerleri ile mobil faza ait N T E değerleri arasındaki ilişkiden de yararlanılmıştır. Ayrıca . ve Rs değerleri hesaplanmış ve bu veriler dikkate alınarak mobil faz derişimi belirlenmiştir. Bu bileşiklerin ayrımı, 25 Mm fosforik asit içeren, pH.sı 4,75 olan % 40 asetonitril-su içeren mobil fazda sağlanmıştır. Gaz kromatografisinde ise, bu bileşiklerin türevlendirilmesi için 5 farklı türevlendirici, farklı sıcaklık ve farklı reaksiyon süreleri kullanılarak optimum şart belirlenmiştir. Türevlendirmede, metillendirici olarak metanolik BF3 ve metanolik HCl; TMS türevlerinin hazırlanması için MBSTFA, MSTFA ve BSTFA kullanılmıştır. The influence of pH and solvent composition of acetonitrile.water mobile phase on the retention of some NSAIDs, clofibric acid and bezafibric acid on a Gemini Phenyl C6 column was studied. The fit of retention to pH for a particular solvent composition provides the pKa values of the equilibria between the acid-base species and the retention parameters of these species at this solvent composition. Several models were tested that relate this parameters to solvent composition and properties in order to propose a general model to predict retention for any mobile phase pH and composition. Correlation between retention and the mobile phase pH, measured in the acetonitrile-water mixture has been used to investigate the pH effect on chromatographic behavior of these compounds. pKa values of water insoluble NSAIDs in acetonitrile-water binary mixture were determined by using the variation of capacity factors of these drug candidates as a function of the mobile phase pH. Mobile phase was optimised by establishing relationships between retention parameters and N T E of mobil phase. In addition, . and RS values were used in order to establish a general model relating elution behaviour of substances with composition of mobile phase.The optimal mobile phase for separation of some NSAIDs, clofibric acid and bezafibric acid was a mixture of acetonitril water with a percentage of ACN-water of 40% (v/v). The pH of the mobile phase containing 25 mM phosphoric acid was adjusted 4,75. For gas chromatographic analysis, five different derivatizing agents were tested on a standard solution of NSAIDs, clofibric acid and bezafibric acid with various reaction times and temperatures. NSAIDs, clofibric acid and bezafibric acid were converted into their methyl derivatives with methanolic BF3 and methanolic HCl. Also, TMS derivatives were prepared with MSTFA, MBSTFA and BSTFA.
