Bu tez çalışmasında hindi (Meleagris gallopavo) konjunktivasının tüm bölgelerinde (marginal, tarsal, orbital, forniks ve bulbar) bulunan mast hücrelerinin farklı fiksatiflerin uygulanması ile alt tiplerinin (bağ dokusu ve mukozal mastositler) ve konjunktiva bölgelerindeki dağılımlarının belirlenmesi amaçlandı.Konjunktivanın göz kapaklarının iç yüzeyini örten ince saydam bir membran yapısında olduğu tespit edildi. Konjunktivanın temel olarak üç ana bölgeye (Palpebral konjunktiva, Forniks konjunktivası ve Bulbar konjunktiva) ayrıldığı belirlendi.Palpebral konjunktivanın kan damarlarının en fazla bulunduğu bölge olduğu, tarsal konjunktivadan itibaren ise kadeh hücrelerinin başladığı tespit edildi. Forniks konjunktivanın en fazla kadeh hücresi ve lenf folikülü içeren bölge olduğu saptandı. Bulbar konjuktivada kadeh hücresine rastlanmadı.Çalışmada kullanılan fiksatifler arasında mast hücrelerinin şeklini ve granüllerini en iyi tespit eden fiksatifin Mota’nın kurşun asetat solüsyonu (BLA) olduğu belirlendi. BLA ve Carnoy solüsyonlarında mast hücrelerinin metakromazi özelliklerinin daha iyi gözlendiği tespit edildi. Formaldehit tespit solüsyonunda toluidine blue boyamasında mast hücrelerinin tamamının mavi renkte boyandığı ancak metakromazi özelliğinin çok zayıf olduğu saptandı. Alcian Blue- Safranin O uygulaması sonucunda safranin (+) ve karışım halinde Alcian Blue- Safranin O (+) mast hücrelerinin çok nadir olduğu tespit edildi. Konjunktiva bölgelerinde bu kombine boyama sonucunda Alcian Blue (+) mast hücre sayısının fazla olduğu yani mast hücrelerinin büyük birt kısmının mukozal mast hücresi karakterinde olduğu belirlendi. Carnoy ve %10’luk formalin fiksatiflerinde tek başına safranin pozitif mast hücresine rastlanmadı.Mast hücrelerinin genel dağılımlarının belirlenmesi amacıyla uygulanan Toluidine blue ve Thionin boyama metodları ile verilerin istatistiksel değerlendirilmesi sonucunda mast hücre sayısının en fazla olduğu bölgenin palpebral konjunktiva olduğu bunu sırasıyla forniks ve bulbar konjunktivanın izlediği belirlendi.Berberin sülfat uygulaması ile mast hücrelerinin zayıf sarı-yeşil floresan verdikleri tespit edildi. Bölgeler arasında bir farka rastlanmadı.Sonuç olarak hindi mast hücrelerinin boyanma özelliklerine göre farklılıklar gösteren heterojen özellikte olduğu belirlenmiştir. Bu özellikleri ile konjunktiva hastalıklarının patogenezinin belirlenmesinde kullanılabileceğini düşünmekteyiz. In this thesis, it was aimed to determine that subtypes (connective tissue and mucosal mastocytes) and distributions of mast cells in all regions (marginal, tarsal, orbital, fornix and bulbar) of turkey (Meleagris gallopavo) conjunctiva by the implementation of the different fixatives.It was found to be that conjunctiva was a thin transparent membrane covering the internal surface of eyelids. There were basically three main regions (Palpebral conjunctiva, Fornix conjunctiva and Bulbar conjunctiva) of the conjunctiva were separated. It was detected that palpebral conjunctiva was the region the blood vessels where the maximum, and the goblet cells started from the tarsal conjunctiva. Fornix conjunctiva was found to be that the region goblet cell and lymph follicle where the maximum. The goblet cell was not encountered in the bulbar conjunctiva.Mota’s lead acetate solution (BLA) was the best fixative among the used in this study for shape and granules of mast cells. It was detected that metachromasia feature of mast cells was observed better with BLA and Carnoy solutions. In the formaldehyde fixative and Toluidine Blue staining, all of tha mast cells stained blue but the metachromasia was very weak. As a result of Alcian Blue-Safranin O application, the safranin (+) and the mixed Alcian Blue-Safranin O (+) mast cells were very rare. In the same combined staining, it was detected that Alcian Blue (+) mast cells were excessive so a large number of mast cells were property of mucosal mast cell in the conjunctival regions. Only safranin positive mast cell was not found in the Carnoy and 10% formalin fixatives.As a result of statistical evaluation of data with the Toluidine Blue and Thionin staining methods applicated for determination of the general distributions of mast cells, it was detected that the palpebral conjunctiva was the region with the highest number of mast cell, and fornix and bulbar conjunctiva respectively followed by it.Mast cells gave weak yellow-green fluorescence through the Berberine sulfate application. There was no difference among the regions.Consequently, it was determined that turkey mast cells were heterogeneous showed differences according to the staining properties. We think that they can be effectively used to determine pathogenesis of conjunctival diseases with these characteristics.
