Bu araştırmada kurşun [Pb(NO3)2], çinko [Zn(NO3)2.4H2O] ve kadmiyum [Cd(NO3)2.4H2O] ortamlarındaki Sorghum bicolor L. tohumlarının çimlenmesi ve çimlenme sonrası oluşan fideciklerin 25±1 °C sabit sıcaklık ve devamlı 8000 lux ışık şiddeti, %60±5 nem koşullarındaki büyüme odasına transferinden 21 gün sonraki büyüme parametreleri üzerindeki ağır metal etkilerinde gibberellik asit (GA3/G), kinetin (K) ve bu iki bitki büyüme maddesinin (BBM) kombinasyonlarının (K+G) rolü incelenmiştir.Kurşun (Pb), çinko (Zn) ve kadmiyum (Cd) konsantrasyon artışına paralel olarak tohumların çimlenmesini hem engellemiş hem de geciktirmiştir.Çimlenme üzerinde en zararlı etkiyi Cd yapmış bunu sırası ile Pb ve Zn izlemiştir. BBM uygulamaları Pb ile Cd’un çimlenmeyi engelleyici ve geciktirici etkisini çoğunlukla hafifletemezken Zn’nun yol açtığı bu olumsuzlukları değişik derecede ve önemli ölçüde hafifletmişlerdir. Her üç ağır metal de hem çimlenme hem de fide büyümesi aşamasında radikula/kök uzaması başta olmak üzere koleoptil/gövde uzamasını belirgin şekilde engellemiştir. Her iki aşamada da bu ağır metallere karşı radikula/kök uzaması koleoptil/gövde uzamasından daha duyarlı bulunmuştur. BBM hem çimlenme hem de fide büyümesi aşamasında bu ağır metallerin radikula/kök uzaması üzerindeki baskısını hafifletmede başarılı olamamışlardır. Çimlenme aşamasında Pb, Zn ve Cd’un koleoptil uzaması üzerindeki olumsuz etkilerini hafifletmede K+G başta olmak üzere uygulanan tüm bitki büyüme maddeleri (BBM) oldukça başarı sağlamışlardır. Fide büyümesi aşamasında Pb ve Zn’nun gövde uzaması üzerindeki engelleyici etkileri GA3 uygulaması ile belirgin derecede hafifletilebilinirken Cd'un engelleyici etkileri hiçbir uygulama ile hafifletilememiştir. Çimlenme aşamasında çalışılan ağır metallerin taze ağırlık ve fidecik su içeriği yüzdesinde yaptıkları olumsuz etkiler K ve K+G başta olmak üzere BBM uygulamaları ile çoğunlukla hafifletilebilmiştir. Fidelerin taze ağırlık, kuru ağırlık ve fide su içeriği yüzdelerinde de her üç ağır metalin olumsuz etkileri görülürken bu engellemelerden en fazla taze ağırlığın etkilendiği belirlenmiş ancak BBM ile bu etkilerin hafifletilemediği tespit edilmiştir. In this study, the roles of gibberelic acid (GA3/G), kinetin (K) and a combination of these two plant growth regulators (PGR) on the effects of heavy metals on some growth parameters of the germination of the seeds and subsequently, of the seedlings from the seeds grown for 21 days in a growth chamber under conditions of 25±1 °C constant temperature, continuous fluorescent light (8000 lux) and 60%±5 moisture were studied.Lead (Pb), zinc (Zn) and cadmium (Cd) both delayed and prevented the germination of the seeds with the increases of their concentrations. The most adverse effect on the germination was done by Cd, and followed by Pb and Zn respectively. PGR pretreatments could not mostly reduce the preventing and delaying effects of Pb and Cd on germination, while they could reduce these negative effects caused by Zn. All three heavy metals prevented coleoptile/shoot growth, especially radicle/root growth significantly both in germination and seedling growth stages. In the both stages radicle/root growth was more sensitive than coleoptile/shoot growth against those three heavy metals. PGR failed to reduce the restraining effects of these heavy metals on radicle/root elongation during both germination and seedling growth stages. All the used PGR, especially K+G were considerably successful at the reducing of the adverse effects of Pb, Zn and Cd on coleoptile growth in germination stage.While the inhibitory effects of Pb and Zn on shoot elongation could be reduced with GA3 pretreatment, the ones of Cd could not be alleviated with any treatment. Adverse effects of studied heavy metals on fresh weight and water content percentage of the seedlings during germination could mostly be reduced with PGR treatments, mainly K and K+G. All three heavy metals were determined to have negative effects mainly on fresh weight, dry weight and water content percentage of the seedlings and that PGR pretreatment could not reduce these effects in the stage of seedling growth.
