Sefalosporinler β-laktam antibiyotiklerinin ikinci büyük grubudur ve dört kuşakta sınıflandırılırlar. Bu antibiyotikler, uygun antibakteriyel aktivite, β-laktamaz direnci ve farmakokinetik özelliklerinden dolayı ağır enfeksiyonların tedavisinde yaygın olarak kullanılmaktadır.Bu çalışmada sefalosporin grubunda bulunan sefadroksil vesefoperazonun elektrokimyasal davranışları incelenip bu etken maddelerin çeşitli ortamlarda tayini için voltametrik yöntem geliştirilmiştir. Elektrokimyasal davranışları, karbon grafit elektrot kullanılarak diferansiyel puls voltametri yöntemiyle incelenmiştir. Destek elektrolit ve pH’nın, bileşiklerin elektrokimyasal davranışları üzerine etkisi incelenmiştir. Gözlenebilme sınırı (S/N=3) sefadroksil ve sefoperazon için sırasıyla 5,50×10-7M ve 7,608×10-6 M olarak bulunmuştur. Aynı şekilde alt tayin sınırı (S/N=10) sefadroksil için1,835×10-5M ve sefoperazon için 2,536×10-5 M'dır.Optimize edilen yöntem ile farmasötik ve kanörneklerinde sefoperazonun tayini gerçekleştirilmiştir. Bu uygulamalarda %100’e yakın geri kazanım değerlerielde edilmiştir. Elektrot yüzeyine kütle transferini karakterize etmek için pik akımları üzerine tarama hızının etkisi dönüşümlü voltametri yöntemi ile incelenmiş ve elektrokimyasal davranışın difüzyon kontrollü olduğu belirlenmiştir. Cephalosporins are the second major group of β-lactam antibiotics, they are classified into four generations. These antibiotics are widely used in clinical therapy for the treatment of severe infections, because of their convenient antibacterial activity, β-lactamases resistance and pharmacokinetic properties.In this study, the electrochemical behaviours of cefadroxil and cefoperazone that are located in the cephalosporin group were examined and voltammetric method was developed for determining those substances in the different mediums. The electrochemical behaviours of the substances in the different mediums. The electrochemical behaviours of the substances were examined at a carbon graphite electrode by using differential pulse voltammetry. Effect on electrochemical behaviour of supporting electrolyte and pH were determined. The limit of detection (S/N=3) was found to be 5,50×10-7 M and 7,608×10-6 M, respectively for cefadroxil and cefoperazone. Similarly limit of quantification (S/N=10) was 1,835×10-5M for cefadroxil and 2,536×10-5 M for cefoperazone. The cefoperazone analysis with these optimized method was carried out in pharmaceutical and biological sample. In such applications, recovery values around 100% were reconised. To characterize the mass transfer to the electrode surface, effect of the scan rate on the peak currents was examined by the cyclic voltammetry method.
