Bu tez çalışmasında, tuz stresi altında yetişen mısır bitkisine topraktan uygulanan nitrik oksidin mısır bitkisinin gelişimi, mineral beslenmesi ve bazı fizyolojik özellikleri üzerine etkisi araştırılmıştır. Bitkilere tuz stresi oluşturmak amacıyla kontrol grubu hariç tüm saksılara 40 mM NaCl uygulanmıştır. Nitrik oksit (NO)’ in tuz stresi altında yetiştirilen mısır bitkisindeki fizyolojik değişimlere ve gelişime etkisini belirlemek amacıyla nitrik oksit vericisi olarak sodyum nitroprussid (SNP) ve NO yakalayıcısı olarak 0. 5 ve 1 mM dozlarında metilen mavisi (MM) uygulanmıştır.Araştırma sonuçlarına göre, NaCl ve SNP’ in artan dozları bitkinin yaş ve kuru ağırlığını kontrole göre önemli oranda azaltmıştır. Membran geçirgenliği, nisbi nem içeriği, yaprak su tutma kapasitesi, total antioksidan aktivitesi, prolin içeriği, lipid peroksidasyonu ve stoma direnci NaCl ve SNP uygulamalarından istatistik olarak önemli derecede etkilenmemiştir. Bitkinin K ve Ca içerikleri uygulamalardan önemli olarak etkilenmezken, N, Cl, Na, Mg, Fe, Zn ve Mn içeriklerinde önemli değişimler belirlenmiştir. Sonuç olarak, tuz stresi altında yetiştirilen mısır bitkisinin strese karşı tolerans geliştirmesinde, NO vericisi olan SNP nin dışarıdan 0. 5 mM ve 1. 0 mM gibi yüksek dozlarda uygulanmasının olumlu etki oluşturmadığı tespit edilmiştir. In the present thesis work, the effect of nitric oxide (NO) on growth, mineral nutrients contentrations and some physiological properties of maize under salt stress condition was investigated. For this propose, 40 mM NaCl (except control), exogenously applied sodium nitroprussid (SNP) as a NO donor, and methylene blue (MB), as a NO scavenger, (0, 0.5 and 1.0 mM) were supplied.Results of this study showed that, NaCl and SNP applications were significantly decreased plant fresh and dry weight compared control. Membrane permeability, lipid peroxidation, measured in terms of malondialdehyde (MDA) content, excised-leaf water loss, and relative water content, stomatal resistance and proline accumulation were not affected by NaCl and SNP treatments.Nitrogen, P, Cl, Na, Mg, Fe, Zn, Cu and Mn concentrations of maize plant were significantly affected, however no significant effect on the K and Ca concentrations of maize plant was observed. Nitrogen, Na, Cl, Na, K, Ca, Mg, Fe, Zn and Mn concentrations were increased by increasing levels of SNP. The results of this study indicated that high doses (0.5 mM and 1.0 mM) of exogenously applied sodium nitroprusside (SNP), as a NO donor, have not positive effect to improve plant performance aganist salinity stress.
