Amaç: Bu tez çalışmasının amacı, Pseudomonas aeruginosa'da üretimi çevreyi algılama (QS) sistemi tarafından düzenlenen virülens faktörleri üzerine boronik asit türevi moleküllerin potansiyel inhibitör etkisinin araştırılmasıdır.Yöntem ve Bulgular: 2-Nitrofenilboronik asit (2-NFBA), 4-Bromofenilboronik asit (4-BFBA), 4-Klorofenilboronik asit (4-KFBA), 4-Fluorofenilboronik asit (4-FLFBA), 4-Formilfenilboronik asit (4-FFBA), 4-Metoksifenilboronik asit (4-MFBA), Metilboronik asit (MBA) ve Fenilboronik asit (FBA) moleküllerinin, potansiyel QS inhibitör etkileri QS inhibitor seçici suş 1 (QSIS1) incelenmiştir. Potansiyel inhibitör olduğu tespit edilen moleküllerin QS üzerine etkileri, Chromobacterium violaceum 026 ve C. violaceum VIR07 biyosensör suşları kullanılarak test edilmiştir. Ayrıca bu moleküllerin P. aeruginosa PA01 suşu, P. aeruginosa PAK01, P. aeruginosa PAK02, P. aeruginosa PAK04 izolatlarında elastaz ve piyosiyanin üretimi, biyofilm oluşumu ve kayma hareketi üzerine inhibitör etkileri araştırılmıştır. Çalışma sonucunda QS inhibitörü olarak en aktif molekülün 4-BFBA olduğu tespit edilmiştir.Sonuç: 2-NFBA, 4-BFBA, 4-KFBA ve 4-FLFBA moleküllerinin viyolasin, elastaz ve piyosiyanin üretimini, kayma hareketi ve biyofilm oluşumunu önemli derecede inhibe ettiği tespit edilmiştir. İnhibitör etkisi en yüksek olan 4-BFBA molekülünün, P. aeruginosa PA01 ve P. aeruginosa PAK01, PAK02, PAK04 klinik izolatlarında sırasıyla elastaz üretimini % 44-73, piyosiyanin üretimini %54-74, biyofilm oluşumunu %18-39 kayma hareketini %34-88 oranında inhibe ettiği belirlenmiştir.Çalışmanın Önemi: Bu çalışmanın sonuçları P. aeruginosa virülensini azaltmak için boronik asit moleküllerinin potansiyel etkinliklerini ortaya koymuştur. Objective: The aim of this study was to investigate the potential inhibitory effect of the boronic acid molecules on the production of virulence factors in Pseudomonas aeruginosa regulated by the quorum sensing (QS) system. Method and Findings: QS inhibitor selector strain1 (QSIS1) was used as the first screen to evaluate the potential QS inhibitory effect of boronic acid molecules 2-Nitrophenylboronic acid (2-NPBA), 4-Bromophenylboronic acid (4-BPBA), 4-Chlorophenylboronic acid (4-CPBA), 4-Fluorophenylboronic acid (4-FLPBA), 4-Formylphenylboronic acid (4-FPBA), 4-Methoxyphenylboronic acid (4-MPBA), Methylboronic acid (MBA) and Phenylboronic acid (PBA). We have tested boronic acid molecules as quorum sensing inhibitor by using Chromobacterium violaceum CV026 and C. violaceum VIR07 as biosensor strains. And QS inhibitory activity of the chosen substances further investigated on elastase, pyocyanin production, biofilm formation and swarming motility in P. aeruginosa PA01 strain, P. aeruginosa PAK01, P. aeruginosa, PAK02, P. aeruginosa PAK04 isolates. Our results have demonstrated that the most active molecule was 4-BPBA as a QS inhitor. Conclusion: 2-NPBA, 4-BPBA, 4-CPBA and 4-FLPBA caused a significant inhibition of violacein, elastase and pyocyanin production, swarming motility and biofilm formation. The most active molecule was 4-BPBA caused a substantial decrease in elastase production (44-73% decrease), in pyocyanin production (54-74% decrease), in biofilm formation (18-39% decrease) and in swarming motility (34-88% decrease) in P. aeruginosa.Significance of the Study: The results of this study highlight the potential effectiveness of boronic acid molecules in attenuating the virulence of P. aeruginosa.
