Bu çalışmada, kuraklık ve tuz stresleri ile bu stresler altında silisyum uygulamasının, zengin bir protein ve yağ kaynağı olan yerfıstığının anatomik özellikleri, antioksidatif sistem bileşenleri ve iki farklı gen ifadesine etkisi karşılaştırmalı olarak incelenmiştir. Elde edilen veriler ışığında, literatürde çok kısıtlı bilgi bulunan tuz ve kuraklık stresinin yerfıstığı dokularındaki etkisinin incelenen parametreler dahilinde aydınlatılması ve stres toleransı için yerfıstığı ıslahında kullanım potansiyeli olabilecek çeşitli moleküler mekanizmaların belirlenmesi bu çalışmanın hedefleri arasındadır. Ayrıca silisyumun kuraklık ve tuz stresine etki mekanizmalarının bir derece daha aydınlatılması ve bu elementin, abiyotik stresle mücadele için tarımdaki potansiyel kullanımının değerlendirilmesi de çalışmanın bir diğer hedefidir.Bu hedeflerle, kuraklık ve tuz stresleri altında değişen gövde/kök uzunluğu, kuru/yaş ağırlık ve bağıl su içeriği, prolin, malondialdehit, hidrojen peroksit ve klorofil miktarları, iyon sızıntısı ve SOD enzim aktiviteleri gibi antioksidan sistem göstergeleri ile LEA7-1 ve rezveratrol sentaz (STS) gen aktiviteleri, silisyum varlığında kontrol bitkilerle karşılaştırmalı olarak kök ve gövde dokularında incelenmiştir.İstatistiksel olarak anlamlı olmamakla birlikte uygulanan streslerin yerfıstığı anatomisi üzerinde olumsuz etkileri gözlenmiş, kuraklık stresinin yaş ağırlık üzerindeki etkileri tuz stresine kıyasla daha belirgin bulunmuş, her iki stresin de gövde uzunluğu ve su tutma kapasitesi üzerindeki etkileri benzer şiddette olmuştur. Silisyum uygulaması normal şartlar altında gerçekleştirildiğinde incelenen tüm anatomik parametreler üzerinde olumlu etkiler göstermiş, her iki stres uygulaması altında da genellikle doku kuru ve yaş ağırlıkları ile uzunluklarının artmasına neden olmuştur. Stres altındaki dokularda su tutma kapasitesi de silisyum varlığında belirgin biçimde artmıştır.Tuz stresi olumsuz etkisini incelenen parametrelerden özellikle malondialdehit (MDA) ve hidrojen peroksit (H2O2) miktarları ile kök dokularında süperoksit dismutaz (SOD) enzim aktivitesini artırarak göstermiştir. Tuz stresi altında rezveratrol sentezi yolundaki en önemli enzimlerden birini kodlayan rezveratrol sentaz (STS) gen ifadesi her iki dokuda da azalmış, LEA7-1 gen ifadesi ise kökte belirgin bir düşüş gösterirken gövdede artmıştır.Kuraklığın dokulardaki olumsuz etkileri ise çarpıcı biçimde gövde MDA, H2O2 ve prolin seviyeleri ile SOD enzim aktivitesi üzerinde gözlenmiş, kök dokularının gövdeye kıyasla daha az stres belirtisi gösterdiği de belirlenmiştir. Kuraklık stresi STS gen ifadesini belirgin biçimde arttırmazken, hem gövde hem de kök dokularında LEA7-1 aktivitesi belirgin artışlar göstermiş, bu genin yerfıstığı kuraklık stres toleransında önemli işlevleri olduğu ve yerfıstığı abiyotik stres toleransı için moleküler ıslah çalışmalarında kullanım potansiyeli olabilecek bir gen olduğu belirlenmiştir.Elde edilen veriler silisyumun yerfıstığında, anatomik özellikler, MDA ve prolin seviyeleri ile SOD enzim ve LEA gen aktiviteleri gibi çeşitli parametreler dahilinde, ancak belirli stres uygulamalarında ve belirli dokularda hafifletici etkisi olduğunu göstermiş, bununla beraber gerçekleştirilen birçok uygulama altında silisyumun toksik etkilerine de rastlanmıştır. Yapılan çalışmalar, uygulama süresi ve miktarının detaylı biçimde optimize edilmediği sürece tarımsal silisyum uygulamalarının bitki stres direncine olumsuz etkiler de yapabileceğini ortaya koymuştur. Bu nedenle yerfıstığı yetiştiriciliğinde toprak silisyum miktarının artırılması pratikte kullanışlı bulunmamıştır. In this study, particular antioxidative system components and the expression levels for the two genes under drought and salt stress and potential effects of silicon on the defense system of treated peanut plants, which are rich sources of protein and lipids, were investigated by comparing the interactions of the analyzed components. One of the objectives of the study is the investigation of the effects of salt and drought stresses on peanut tissues, on which very limited information exists in the literature and investigation of molecular mechanisms taking part in abiotic stress tolerance, which also have a potential to be used in peanut breeding. Another objective of the study is to understand the mechanisms taking part in drougt and salt stress tolerance of plants with silicon application and help evaluation of the potential use of this element in agricultural production.With these objectives, some antioxidative system indicators including, shoot/root lenghts, dry/wet weights, relative water content, prolin, malonedialdehyde, hydrogen peroxide and chlorophyll contents, ion leakage and SOD enzyme activity were determined and the expression of the genes coding LEA7-1 and resveratrol synthase (STS) enzymes was analysed comparatively in both root and shoot tissues under drought and salt stress in the presence of silicon.Although statisticially insignificant, applied stresses negatively affected peanut anatomical features, effects of drought stress appeared to be more prominent when compared to salt stress and the levels of both stresses on shoot lenght and water retention capacity occured at similar intensities. When applied under normal conditions, silicon positively affected all investigated parameters and under both stress applications, presence of silicon usually increased tissue wet/dry weights and lenghts. Water retention capacity in stressed tissues also increased prominently under silicon application.Salt stress showed its negative effects mainly by increasing tissue malonedialdehyde (MDA) and hydrogen peroxide (H2O2) levels and also by increasing superoxide dismutase enzyme activity (SOD). Under salt stress, one of the most important enzymes in the resveratrol synthesis pathway namely resveratrol synthase (STS) gene expression decreased in both tissues, while LEA7-1 gene expression decreased in root tissues and increased in shoots.Negative efects of drought stress were observed significantly in shoot tissues in terms of MDA, H2O2, prolin levels and SOD enzyme activities and it was also determined that root tissues exhibit less stress symptoms. Drought stress did not cause any important enhancement in STS gene expression, while LEA7-1 activities showed prominent increase in both shoot and root tissues. Therefore, the gene was determined to have the potential to be used in molecular breeding of peanut for abiotic stress tolerance.Obtained data showed that silicon application in peanut has some roles in stress alleviation, only in terms of specific parameters including MDA and prolin levels in addition to SOD enzyme and LEA gene activities and only under limited stress applications and in specific tissues. While under various applications toxic effects of silicon were also observed. Conducted studies showed that silicon application may have negative effects on plant stress tolerance unless application durations and amounts were strictly optimized. Therefore, increasing the soil silicon levels for peanut cultivation was not found useful in practice.
