Bu tez çalışmasında sıralı anaerobik-aerobik döngü modunda işletilen laboratuar ölçekli bir ardışık kesikli reaktörde karbon kaynağının ve katyon dengesinin biyolojik aşırı fosfor giderimi (BAFG) prosesi ve aktif çamurun çökme performansı üzerine etkisi incelenmiştir. İlk deneysel çalışma aşamasında katyon dengesinin aktif çamurun çökme performansı ve fosfor giderim verimi üzerine etkisinin belirlenmesi amacıyla karbon kaynağı olarak sodyum asetatın kullanıldığı sentetik atıksu çözeltisi bileşimindeki toplam monovalent (M) katyon (NH4+, K+ ve Na+) miktarının (meq/L) toplam divalent (D) katyon (Ca2+ ve Mg2+) miktarına (meq/L) oranının (M:D) 7,6 ve 1,5 olarak ayarlandığı durumlarda her biri otuz günlük iki periyot halinde reaktör işletimi yapılmıştır. Sonuçlar, aktif çamurun çökme performansını ifade eden çamur hacim indeksi (ÇHİ) değerinin her bir işletme periyodu boyunca M:D oranı 7,6 olduğunda 125 mL/g değerinden 440 mL/g değerine arttığını ve 1,5 olduğunda ise 130 mL/g değerinden 70 mL/g değerine azaldığını ve aerobik faz boyunca fosfor giderim veriminin M:D oranı 7,6 için %38-50 aralığında değiştiğini ve M:D oranı 1,5 için %80 değerine kadar arttığını göstermiştir. İkinci deneysel çalışma aşamasında karbon kaynağının BAFG prosesi üzerine etkisinin belirlenmesi amacıyla karbon kaynağı olarak yalnızca asetat ve yalnızca glikoz ile beslenen reaktörlerde her biri altmış günlük iki periyot halinde reaktör işletimi yapılmıştır. Karbon kaynağı olarak 500 mg KOİ/L asetat beslenen reaktörde tüm işletme süresi boyunca kimyasal oksijen ihtiyacı (KOİ) giderimi bazında anaerobik substrat tüketimi %85 değerine kadar artarken anaerobik fazdaki fosfat salım verimi 34. gün itibariyle başlangıç değeri olan %50'den %350'ye artmış ve bu süreden sonra fosfat salımı stabil hale gelmiştir. Aerobik fazdaki fosfat giderim verimi ise 38. güne kadar hızla artmış ve bu aşamadan sonra yavaşça artmıştır. Bu süreçte en yüksek fosfat giderim verimi %93,2 olarak elde edilmiştir. Aktif çamurun toplam fosfor miktarı (askıda katı madde (AKM) konsantrasyonunun yüzdesi olarak, %AKM), giderilen KOİ başına salınan fosfor (Psal/KOİgid) oranı ve fosfor salımının fosfor giderimine (Psal/Pgid) oranı için elde edilen deneysel değerler de zamanla belirli bir seviyeye kadar artmıştır. Asetatın beslendiği reaktörde kullanılan aktif çamur değiştirilmeden karbon kaynağı olarak 500 mg KOİ/L glikozun beslendiği reaktörde ise tüm işletme süresi boyunca KOİ gideriminde bariz bir değişim gözlenmezken anaerobik fosfor salım veriminde ve aerobik fosfor giderim veriminde zamanla bir azalma meydana gelmiştir. Bu süreçte anaerobik fosfor salım verimi %346 değerinden %62 değerine ve aerobik fosfor alım verimi %92 değerinden %62 değerine azalmıştır. Öte yandan tüm işletme süresi boyunca Psal/KOİgid ve Psal/Pgid değerlerinde ve ayrıca aktif çamurun toplam fosfor içeriğinde de kademeli olarak azalma meydana gelmiştir. Bu sonuçlar, glikoz ile beslenen sistemde anaerobik substrat kullanımı gerçekleşmesine rağmen asetat ile beslenen sistemdekinden farklı olarak mikroorganizmaların fosfor kullanım metabolizmasında değişiklik meydana geldiğini ortaya koymaktadır. Glikoz ile beslenen sistemde anaerobik fazda glikojen tüketiminin asetat ile beslenen sistemdekine göre artması, glikojen parçalanmasının asetat ile beslenen sistemde polifosfat parçalanması ile birlikte gerçekleştiğini fakat glikoz ile beslenen sistemde polifosfat mekanizmasına daha az gereksinim duyularak gerçekleştiğini açıkça ortaya koymaktadır. Bu sonuçlar, asetat ile beslenen sistemde polifosfat depolayan organizmaların baskın mikroorganizmalar olduğu ve katyon dengesinin ayarlanması için besleme çözeltisinde Mg ve Ca iyonlarının miktarlarının azaltılması haricinde işletme şartları değiştirilmemiş olmasına rağmen glikoz ile beslenen sistemde glikojen depolayan organizmaların baskın hale geldiği fikrini desteklemektedir. Sonuç olarak, karbon kaynağının asetattan glikoza değiştirilmesinin BAFG prosesinde fosfor salımının ve alımının tamamen inhibe olmadan bozulmasına yol açan olumsuz bir etki gösterdiği ve polifosfat depolayan organiamaların (PAO) metabolizmasında polifosfat metabolizmasını kullanmaktan ziyade glikojen metabolizmasını kullanmalarını teşvik eden bir metabolik davranış değişikliğine sebep olduğu düşünülmektedir. Karbon kaynağı olarak hem asetatın hem de glikozun kullanıldığı her iki sistemde de tüm işletme süresi boyunca amonyum azotunun konsantrasyonunda anaerobik ve aerobik fazlarda başlangıç konsantrasyonuna (41,6 mg/L) göre bariz bir değişim meydana gelmemesi, aerobik fazlarda fosfor gideriminin kimyasal giderim gerçekleşmeden yalnızca biyolojik şekilde gerçekleştiğini göstermektedir. Öte yandan nitrit azotu ve nitrat azotu konsantrasyonlarında da anaerobik ve aerobik fazlarda başlangıç konsantrasyonuna göre bariz bir değişim olmaması, substratın yalnızca BAFG prosesi için kullanıldığına işaret etmektedir. Karbon kaynağının asetattan glikoza değiştirilmesinden önce katyon dengesinin sabit kalması için sentetik atıksudaki Mg ve Ca iyonlarının miktarlarının ayarlanması, glikoz ile beslenen BAFG sisteminin katı-sıvı faz ayrımı bakımından başarılı şekilde işletilmesi ile sonuçlanmıştır. Asetat ve glikoz ile beslenen sistemler için elde edilen ÇHİ sonuçları işletme süreleri sonunda sırası ile 36 mL/g ve 40 mL/g olarak elde edilmiştir. Bu sonuçlar, BAFG sistemlerinin karbon kaynağının türünden bağımsız olarak başarılı şekilde işletilebilmesi için katyon dengesinin kritik bir faktör olduğunu ortaya koymaktadır. In this study, the effect of carbon source and cation balance on the enhanced biological phosphorus removal (EBPR) process and the settling performance of the activated sludge was investigated in a laboratory-scale sequencing batch reactor operating under alternating anaerobic-aerobic cycling mode. In the first experimental study stage, a two period-reactor operation each of thirty days was carried out in order to determine the effect of cation balance on the settling performance of the activated sludge and the phosphorus removal efficiency by adjusting the ratio of total monovalent (M) cation (NH4+, K+ and Na+) content (meq/L) to total divalent (D) cation (Ca2+ ve Mg2+) content (meq/L) in the composition of synthetic wastewater solution, where sodium acetate was used as sole carbon source, as 7.6 and 1.5. The results showed that sludge volume index (SVI) which explains settling performance of the activated sludge increased to 440 mL/g from 125 mL/g when M:D ratio was 7.6 and decreased to 70 mL/g from 130 mL/g when M:D ratio was 1.5 during each operation period, and phosphorus removal efficiency during aerobic phases changed in the range of 38-50% for M:D ratio 7.6 and increased up to 80% for M:D ratio 1.5. In the second experimental study stage, a two period-reactor operation each of sixty days was carried out in order to determine the effect of carbon source on the EBPR process in reactors feeding with only acetate and only glucose as sole carbon source. In the reactor fed with 500 mg COD/L of acetate, phosphate release efficiency during the anaerobic phase increased to 350% from 50%, which was the start-up efficiency, on day 34 and then it became stabil while anaerobic substrate consumption as chemical oxygen demand (COD) removal increased up to 85% during the whole operation period. Aerobic phosphate removal efficiency increased rapidly during the first 38 days of the operation and then increased slowly during the remaining days. The highest phosphate removal efficiency was obtained as 93.2% in this experimental period. The experimental values obtained for total phosphorus content of the activated sludge (as percent value of concentration of mixed liquor suspended solids (MLSS), MLSS%), phosphate release per COD removal (Prel/CODup) and ratio of phosphate release to phosphate removal (Prel/Pup) were also increased up to a certain level by the time. In the reactor fed with glucose of 500 mg COD/L without changing the activated sludge used in acetate-feeding reactor, anaerobic phosphorus release efficiency and aerobic phosphorus uptake efficiency decreased while COD removal did not change significantly during the experimental study. In this study, anaerobic phosphorus release efficiency decreased from 346% to 62% and aerobic phosphorus uptake efficiency decreased from 92% to 62%. Also, values for Prel/CODup and Prel/Pup and total phosphorus content of activated sludge decreased gradually in this study. These results show that unlike the phosphorus utilization mechanism of microorganisms in the acetate-feeding system, the mechanism was changed although substrate was consumed anaerobically in the glucose-feeding system. Increase in glycogen consumption during anaerobic phases in the glucose-feeding reactor than that of acetate-feeding reactor clearly show that glycogen degredation took place along with polyphosphate degredation in the acetate-feeding reactor, however it took place with less requirement for polyphosphate mechanism in the glucose-feeding reactor. These results supports that polyphosphate accumulating organisms were dominant in the acetate-feeding system and that glycogen accumulating organisms became dominant in the glucose-feeding system although operating conditions of the system were not except of decreasing the amount of Mg and Ca ions in the feeding solution in order to adjust cation balance. Consequently, it is thought that altering carbon source from acetate to glucose showed an adverse effect leading to deterioration of EBPR process without complete inhibition of phosphorus release and uptake and resulted in a change in metabolic behaviour of polyphosphate accumulating organisms (PAOs) favoring utilization of glycogen metabolism rather than polyphospate metabolism. In the both system where acetate or glucose was used as sole carbon source, no remarkable change in the concentration of ammonium-nitrogen during anaerobic and aerobic phases compared to the initial concentration (41.6 mg/L) implies that phosphorus uptake in aerobic phases occured only biologically without any chemically-induced removal. Also, no remarkable change in the concentration of nitrite-nitrogen and nitrate-nitrogen during anaerobic and aerobic phases implies that substrate was consumed only for EBPR process. Adjusting of the amount of Mg and Ca ions in the feeding solution in order to set a constant cation balance before altering the carbon source from acetate to glucose resulted in successful operation of glucose-feeding EBPR system in terms of separation of solids and liquid phases. SVI value of acetate- and glucose-feeding system was obtained as 36 mL/g and 40 mL/g, respectively, at the end of operation time. These results show that cation balance is a critical factor to successfully operate EBPR systems with independency of the type of carbon source.
