Sefalasporinler, β-Laktam antibiyotiklerin alt grubudur. Sefalasporinlerin temel çekirdeği dihidrotiazin çekirdeği ve β -Laktam halkasından oluşan 7-aminosefalasporanik asittir. Farklı R1 ve R2 sübstitientlerinin bağlanması ile farklı farmokolojik ve farmokokinetik özelliklere sahip sefalasporinler oluşur. Aktivitelerine göre tanınmış dört kuşak sefalosporin vardır. Sefalosporinler yaygın olarak bakteriyel enfeksiyonların tedavi edilmesi için kullanılmaktadırlar. Bunlar yüksek etkinliğe ve düşük toksisiteye sahip antibiyotiklerdir. β- laktam antibiyotikleri bakteriyel organizmalarda hücre duvarı biyosentezini inhibe ederek çalışırlar ve bunlar en yaygın kullanılan antibiyotiklerdendir. Bunlar genellikle bakteriostatik seviyelerine yakın konsantrasyonlarda bakterisit etkiye sahiptirler. Kimyasal yapının fizikokimyasal parametreleri içinde, fonksiyonel grup sabitleri en önemlilerinden biridir. Bu çalışmada dört sefalasporinin (Seftibuten, sefaleksin, sefdinir ve sefadroksilin) pKa değerleri, ACN-su karışımında bu bileşiklerin asit baz denkliklerine ilişkin bilgi eksikliğini gidermek için UV/pH titrasyon metoduyla ve sıvı kromatografik metotla tayin edilmiştir. Spektral veriler, titrasyon süresince sürekli olarak kaydedilip nötral ve iyonlaşmış türlerin varlığını yansıtmıştır. Yarı yükseklik metodu, farklı pH'larda kaydedilmiş UV absorbans spektral verilerinden pKa değerlerinin belirlenmesinde kullanılmıştır. pKa tahmini için seçilen metot izokratik ters faz sıvı kromatografidir. Sefalasporinlerin iyonlaşma sabitleri, kapasite faktörünün mobil fazın pH'sına bağlılığı kullanılarak tayin edilmiştir. The cephalosporins, a subgroup of β-lactam antibiotics. The basic nucleus of the cephalosporins is 7-aminocephalosporanic acid which comprises a dihydrothiazine nucleus and a β-lactam ring. Substitution of the various R1 and R2 groups results in cephalosporins with different pharmacological and pharmacokinetic properties. There are four recognized class generations of cephalosporins based on spectra of activity. Cephalosporins are widely used for treating bacterial infections. They are highly effective antibiotics and have low toxicity. β-lactam antibiotics work by inhibiting cell wall biosynthesis in the bacterial organism and are the most widely used group of antibiotics. They are usually bactericidal at concentrations close to their bacteriostatic levels. Among all of the pyhsicochemical parameters of a chemical entity, the functional group constant (pKa) is one of the most important. In this study, pKa values of four cephalosporins (ceftibuten, cephalexin, cefadroxil and cefdinir) was determined in ACN - water mixtures, in order to overcome the lack of information regarding the acid base equilibria of this compounds by means of a UV/pH titration method and liquid chromatographic method. Spectral data was recorded continuously during the course of titration to reflect the concentration of neutral and ionised species present. Halving method was applied to deduce pKa values from the multivawelength UV absorption data recorded at different pH values. The method of choice is pKa estimation by isocratic reverse phase LC. Dissociation constants of cephalosporins was determined by using the dependence of the capacity factor on the pH of the mobile phase.
