Çalışmamızda ratların testis dokusunda demir (Fe) ve çinko (Zn) ağır metallerinin oluşturduğu hasarı ve bu hasarı önlemede antioksidan olan juglon (5-hidroksi-1,4-naftokinon)'nın koruyucu etkisinin histolojik ve immunohistokimyasal yöntemlerle incelemesi amaçlandı. Bu çalışmada Süleyman Demirel Üniversitesi Deney Hayvanları Üretimi ve Deneysel Araştırma Laboratuvarında (HADYEK) yetiştirilen ergin erkek Wistar-Albino türü ratlar kullanıldı. Denekler rastgele 5 gruba ayrıldı. Grup I; demir (Fe) (0.3 ml. stok çözeltiden demir/600 ppm. + 0.7ml.su), Grup II; çinko (Zn) (0.2 ml. stok çözeltiden çinko/400 ppm + 0.8ml.su), Grup III; demir (0.3 ml. stok çözeltiden demir/600 ppm.) + Antioksidan (juglon (5-hidroksi-1,4-naftokinon)/200 ppm- 0.7 ml.), Grup IV; çinko (0.2 ml. stok çözeltiden çinko/ 400 ppm.) + Antoksidan (juglon (5-hidroksi-1,4-naftokinon)/200 ppm-0.7ml.+ 0.1ml. su), Grup V; kontrol (1 ml. Su); her gruba 30 gün boyunca günlük tek seferde gavaj yöntemi uygulandı. Deney sonunda ratlar dekapite edilerek testis dokuları çıkartıldı ve rutin histolojik takipler yapıldıktan sonra alınan kesitlere histolojik boyamalar ve TUNEL metodu uygulandı. Histopatolojik değerlendirmelerde Fe ve nispeten Fe+Juglon uygulanan gruplarda testis dokusunda anlamlı derecede hasar saptandı. En belirgin bulgular, seminifer tübül epitelinde yer yer kopmalar ve epitel kalınlığında azalmalar, seminifer tübüllerin bazal laminasında ayrılmalar, germ hücrelerinde azalma, interstisyel alanda yoğun ödem ve konjesyon, seminifer tübül epitelinde vakuolizasyonlar şeklinde olduğu saptandı. Zn ve Zn+Juglon uygulanan gruplarda ise testis dokularında ki hasarların Fe ve Fe+Juglon grubu kadar olmadığı tespit edildi. TUNEL yöntemine göre ise Fe ve Fe+Juglon ve Zn gruplarının kontrol grubuna göre apoptotik hücre sayısında anlamlı derecede artış gözlendi (p˂0.05). Zn+Juglon grubu ile kontrol grubu arasında ise anlamlı bir farklılık saptanmadı.
The purpose of the study is to investigate the damage caused by iron (Fe) and zinc (Zn) heavy metals in the testis tissue of rats and the protective impact of juglone –an antioxidant in preventing this damage- with the help of histological and immunohistochemical methods. Adult, male Wistar-Albino rats raised in Suleyman Demirel University Experimental Animals Production and Experimental Research Laboratory (HADYEK) were used in this study. Subjects were randomly grouped into 5. Group I; iron (Fe) (iron from 0.3 ml stock solution/600 ppm. + 0.7 ml.water), Group II; zinc (Zn) (zinc from 0.2 ml stock solution/400 ppm + 0.8 ml.water), Group III; iron (iron from 0.3 ml stock solution /600 ppm.) + Antioxidant (juglone (5-hidroxy-1,4- naphthoquinone)/200 ppm- 0.7 ml.), Group IV; zinc (zinc from 0.2 ml stock solution/400 ppm.)+Antioxidant (juglone (5-hidroxy-1,4- naphthoquinone)/200 ppm-0.7 ml.+ 0.1 ml. water), Group V; control (1 ml. Water); gavage technique was applied to each group once a day throughout 30 days. At the end of the experiment, rats were decapitated and their testis tissues removed, then histological staining and TUNEL method were applied following the routine histological follow-ups. Based on the histopathological examinations, significant damage was observed in the testis tissue of the groups administered with Fe and relatively Fe+Juglone The most explicit findings were; partly ruptures in Seminiferous tubules epithelium and decrease in thickness of epithelium, dissociation in basal lamina of the seminiferous tubules, decrease in the number of germ cells, intense edema and congestion in the interstitial area, and vacuolization in the Seminiferous tubules epithelium. The damage associated with the Zn and Zn+Juglone was determined to be less significant than the damage by Fe and Fe+Juglone groups. Based on the TUNEL method, a significant increase was observed in the number of apoptotic cells in Fe, Zn and Fe+Juglone groups compared to the control group (p˂0.05). No significant difference was observed between the Zn+Juglone group and control group.
