Bu tez çalışması Brahma (Gallus gallus) horoz testisinin histolojik yapısının ve bu dokudaki bazı glikokonjugatların (α-D-Man, α-D-Glc, N-asetil-D-Galaktosamin, Fucα1-2Gal-R, Galactose b1,3-N-acetylgalactosamine a1-, GlcNAcβ1-4GlcNAcβ1-4GlcNAc,Neu5Ac) lektin (Con A, SBA, UEA-I, PNA, WGA) histokimyasal yöntemle analizinin yapılması amaçlandı. Çalışmada araştırma materyali olarak Süleyman Demirel Üniversitesi Ziraat Fakültesi Çiftçi Eğitim ve Tarımsal Uygulama kümesinden temin edilen sağlıklı, erişkin 6 adet horoza ait testisler materyal olarak kullanıldı. Çalışma, Süleyman Demirel Üniversitesi Hayvan Deneyleri Etik Kurulu (HADYEK) onayı (karar tarihi: 07.01.2014/ karar no: 01) kapsamında gerçekleştirildi. Alınan testis dokuları, Bouin solusyonunda 16 saat süreyle tespit edildi. Tespit işleminden sonra rutin histolojik doku takibinden geçirilen örnekler parafinde bloklandı. Parafin bloklardan 6-7 μm kalınlığında alınan kesitlere genel histolojik yapının belirlenmesi için Masson'un üçlü boyama metodu uygulandı. Glikokonjugat içeriğinin belirlenmesi amacıyla alınan doku kesitleri horseradish peroxidase (HRP) bağlı Canavalia ensiformis aglutinin (Con A), Glycine max aglutinin (SBA), Ulex europaeus aglutinin (UEA-I), Arachis hypogaea aglutinin (PNA) ve Triticum vulgaris aglutinin (WGA) lektinleri ile inkübe edildi. Uygulanan lektin histokimyasal yöntemler sonucunda primer spermatositlerde Con A lektininde çok güçlü bağlanma gözlenirken Leydig hücrelerinde zayıf bağlanma gözlendi. Sekonder spermatositlerin zayıftan güçlüye kadar tüm glikokonjugatlar içerdiği belirlendi. WGA lektinine karşı erken ve geç dönem spermatidlerde çok güçlü reaksiyon gözlenirken Leydig hücrelerinde reaksiyon gözlenmedi. Spermatogonyumlarda ve lamina prorianın bazal laminasında SBA ve PNA lektinlerine karşı çok güçlü reaksiyon saptandı. UEA-I ve SBA lektin işaretlenmesi Leydig hücrelerinde saptanmadı. Peritubuler hücrelerde zayıf (ConA, UEA-I, SBA) ve orta yoğunlukta ( WGA ve PNA) doğru bağlanma saptandı. Sertoli hücrelerinin çok yoğun şekilde tüm glikokonjugatları içerdiği belirlendi. Sonuç olarak Leydig hücreleri hariç farklı gelişme aşamasındaki spermatojenik hücrelerin tamamının ilgili tüm glikokonjugatları içerdiği sonucuna varılabilir. Sonuçlar kanatlı türlerin testislerinde meydana gelen spermatogenezisin hangi aşamasında ne tür şekerin karıştığını ortaya çıkarmıştır. Kanatlı üreme biyolojisi araştırmalarında, spermatogenezisin düzenlenmesinde bu şekerlerin ne tür roller oynayacağına ilişkin çalışmalara katkı sağlayacağı düşünülmektedir. This thesis study was aimed to determine the histological structure of Brahma (Gallus gallus) cock's testes and the distribution of some glycoconjugates (α-D-Man, α-D-Glc, N-acetyl-D-Galactosamine, Fucα1-2Gal-R, Galactose ß1,3-N-acetylgalactosamine, GlcNAcβ1-4GlcNAcβ1-4GlcNAc, Neu5Ac) in this tissue by using some lectins (Con A, SBA, UEA-I, PNA, and WGA) through lectin histochemical method. Testes of 6 healthy, adult cocks provided from Farmers' Training and Agricultural Practices Center (Süleyman Demirel University, Faculty of Agriculture) were used as study material. The study protocol was approved by Animal Experiments Local Ethics Committee (HAYDEK) of SüleymanDemirel University (Approval date: 07.01.2014, Decision No: 01) in accordance with the guidelines of the International Association for the Study of Pain. After fixation, the samples subjected to routine histological tissue processing were embedded in paraffin. Serial 6-7 µm thick sections were cut and collected on albumin-coated slides. Masson trichrome staining was carried out to demonstrate the general histological structure. The sections were incubated with the horseradish peroxidase (HRP)-conjugated Canavaliaensiformis agglutinin (Con A), Glycine max agglutinin (SBA), Ulex europaeus agglutinin (UEA-I), Arachis hypogaea agglutinin (PNA) and Triticum vulgaris agglutinin (WGA) lectins in order to determine the glycoconjugate content and distribution. Lectin histochemistry results indicated that primary spermatocytes reacted very strongly with Con A lectin, but Leydig cells reacted weakly with this lectin. A weak to strong reaction to all glycoconjugates were observed in secondary spermatocytes. A very strong reaction to WGA was noticed in early and late spermatids as WGA lectin did not bind to Leydig cells. A very strong reaction to SBA and PNA was detected in spermatagonium and the basal lamina of lamina propria. Any UEA-I and SBA lectin labelling was not observed in Leydig cells. Sertoli cells were found to contain plenty of all glycoconjugates intensely. In conclusion, it can be inferred that except for Leydig cells, all of spermatogenic cells at different stages of development contain all the glycoconjugates. Results reveal that what sort of sugar units are involved in which stages of spermatogenesis that occurs in the poultry testes. It has been expected that present study will contribute to the studies such as what kind of roles these sugars will play in the regulation of spermatogenesis in the researches into reproductive biology of poultry.
