Bu tez çalışmasında, hem normal koşullar hem de tuz stresi altında çimlendirilen Allium cepa L.'nın tohum çimlenmesi, fide büyümesi (radikula uzunluğu, radikula sayısı ve taze ağırlık), mitotik aktivite ve kromozom anormallikleri üzerine Ginkgo biloba yaprak ekstraktı (GbYE), Aloe vera yaprak ekstraktı (AvYE) ve arı sütü (AS)'nün etkileri araştırılmıştır. Tek başına GbYE, AvYE ve AS'li ortamda çimlendirilen tohumların radikula uzunluğu saf su ortamında çimlendirilen kontrol tohumlarınınkilerle karşılaştırıldığında artarken, nihai çimlenme yüzdesi istatistiksel açıdan kontrol tohumlarınınkilerle aynı olmuştur. Tek başına AvYE'li ortamda çimlendirilen tohumların radikula sayısı kontrol tohumlarınınkilere göre azalırken, tek başına GbYE ve AS'li ortamda çimlendirilen tohumların radikula sayısı istatistiksel açıdan kontrol tohumlarınınkilerle aynı değerleri göstermiştir. Ayrıca, AS'li ortamda büyütülen fidelerin taze ağırlığı saf su ortamında büyütülen kontrol fidelerininkilerle karşılaştırıldığında artmıştır. Fakat tek başına GbYE ve AvYE'li ortamda büyütülen fidelerde bu parametre kontrole göre azalmıştır. Dahası, tek başına GbYE, AvYE ve AS'li ortamda çimlendirilen A. cepa tohumlarının kök ucu meristemlerinde mitotik indeks ve kromozom anormalliklerinin sıklığı saf su ortamında çimlendirilen kontrol tohumlarınınkilere göre önemli bir artış göstermiştir. Diğer yandan, tuz stresi A. cepa'nın tohum çimlenmesi ve fide büyümesini önemli ölçüde engellemiştir. Ayrıca, tohumların kök ucu meristemlerinde mitotik indeksi önemli ölçüde azaltmış, kromozom anormalliklerinin sayısını ise artırmıştır. Oysaki tuzun tohum çimlenmesi, fide büyümesi ve mitotik aktivite üzerindeki engelleyici etkisi GbYE, AvYE ve AS uygulamaları ile çeşitli derecelerde önemli ölçüde hafifletilmiştir. Fakat tuzun kromozom anormallikleri üzerindeki zararlı etkileri bu uygulamalar ile daha da artmıştır. In this thesis work, the effects of Ginkgo biloba leaf extract (GbLE), Aloe vera leaf extract (AvLE) and royal jelly (RJ) on the seed germination, seedling growth (radicle length, radicle number and fresh weight), mitotic activity and chromosomal aberrations of Allium cepa L. germinated under both normal conditions and salt stress were studied. The radicle length of seeds germinated in the medium with GbLE, AvLE and RJ alone increased in comparison with ones of the control seeds germinated in distilled water medium while their final germination percentage were statistically the same as ones of the control seeds. The radicle number of seeds germinated in the medium with AvLE alone reduced according to ones of the control seeds while the radical number of seeds germinated in the medium with GbLE and RJ alone statistically showed the same values as ones of the control seeds. In addition, the fresh weight of seedlings grown in the medium with RJ alone increased in comparison with ones of the control seedlings grown in distilled water medium. But, this parameter in the seedlings grown in the medium with GbLE and AvLE alone decreased according to the control. Furthermore, the mitotic index and the frequency of chromosomal aberrations in root tip meristems of A. cepa seeds germinated in the medium with GbLE, AvLE and RJ alone demonstrated a significant increase according to ones of the control seeds germinated in distilled water medium. On the other hand, salt stress considerably inhibited the seed germination and seedling growth of A. cepa. In addition, it markedly decreased the mitotic index in root tip meristems of the seeds and increased the number of chromosomal aberrations. Whereas, the inhibitive effect of salt on the seed germination, seedling growth and mitotic activity was dramatically alleviated in varying degrees by GbLE, AvLE and RJ applications. But, the detrimental effects of salt on the chromosomal aberrations greatly increased over with these treatments.
