Bu tez çalışmasında, koyun (Ovis aries), (alt ve üst) trake mukozasında GAL (Galanin), PHI (Peptit Histidin İzolösin) ve Leu-Enk (Leu-Enkefalin) peptitlerinin immunohistokimyasal lokalizasyonlarının belirlenmesi amaçlandı. Çalışmada 10 adet koyun (Ovis aries)’a ait trake (alt ve üst) örnekleri materyal olarak kullandı. Bouin solüsyonunda 16 saat süreyle tespit edilen dokular daha sonra rutin histolojik doku takibinden geçirilerek parafine gömüldü ve bloklandı. Parafin bloklardan alınan 5-6 μm kalınlığındaki kesitlere immunohistokimya yöntemi uygulandı. Üst trake lamina epitelyalisindeki GAL, PHI ve Leu-ENK immunoreaktif hücrelerin yoğunluğunun alt trakeye göre daha fazla olduğu saptandı. PHI immunoreaktif hücrelerin ise üst trake lamina epitelyalisinde diğer peptitlere göre daha fazla sayıda bulunduğu belirlendi. GAL, PHI ve Leu-ENK immunoreaktivitesine aynı zamanda düz kas, seromuköz bez hücreleri, bu bezler ile kan damarları çevresinde ve gangliyon hücrelerinde de rastlandı. Sonuç olarak GAL, Leu-ENK ve PHI'nın geniş mukozal dağılımının, memeli solunum sistemindeki vasküler ve sekretuvar fonksiyonların düzenlemesinde etkili olabileceği düşünülmektedir. Solunum yolu epitellerinde bulunan GAL, PHI ve ENK peptidleri, düzenleyici peptid hormonların ya da nörotransmitterlerin lokal olarak harekete geçirilmesinde de etkili olabilirler. This thesis study aimed to determine immunohistochemical localization of the peptides Galanin (GAL), Peptide Histidin Isoleucine (PHI) and Leu-Enkephalin (Leu-ENK) in tracheal mucosa (upper and lower) of sheep (Ovis aries). The tracheal samples taken from 10 sheep (Ovis aries) were used as the study material. The samples were fixed in Bouin’s solution during 16 hours. After fixation, the samples were passed through routine tissue processing and embedded in paraffin. The immunohistochemical method was applied to 5-6 μm thickness sections to determine immunohistochemical localization of relevant peptides. It was observed that GAL, PHI and Leu-ENK immunoreactive cells were more numerous in the upper part of trachea than in its lower part. It was found that PHI-immunoreactive cells were more than other peptides in lamina epithelialis in upper trachea. GAL, PHI and Leu-ENK immunoreactivity was detected in smooth muscles, seromucous gland cells, around blood vesels and in ganglion cells. In conclusion, it is thought that wide-ranging mucosal distribution of GAL, Leu-ENK and PHI may have effective in regulating the vascular and secretory functions in mammalian respiratory system. Existence of the peptides GAL, PHI and Leu-ENK in the airway epithelium suggests that they may also be effective in acting as regulatory peptides, local hormones or neurotransmitters.
