Bu tez çalışmasında yumurtlama dönemi (16 hafta ve sonrası) ve yumurtlama dönem öncesi (10-16 hafta) Denizli tavuğu (Gallus gallus domesticus) oviduktunun infundibulum, magnum, isthmus, uterus ve vagina bölümlerindeki glikokonjugatların histokimyasal olarak belirlenmesi amaçlandı. Çalışmada Süleyman Demirel Üniversitesine bağlı Ziraat Fakültesi Çiftçi Eğitim ve Tarımsal Uygulama kümesinden temin edilen sağlıklı 6'şar adet yumurtlama dönemi ve yumurtlama dönemi öncesi tavuklara ait ovidukt materyal olarak alındı. Yumurtlama dönemi ve yumurtlama dönemi öncesinde infundibulum ve vagina bölgelerinde propriyal bezlere rastlanmazken magnum, isthmus ve uterus bölgelerinde propriyal bezler gözlendi. Yumurtlama döneminde magnum ve vagina örtü epitelinde güçlü sülfat içeren hücrelere rastlandı. Her iki döneme uygulanan Best Carmine yöntemi sadece isthmus bölgesi örtü epiteli hücrelerinde rastlandı. Alizarin Kırmızısı uygulamasında sadece uterus bölgesi yumurtlama dönemi ve öncesinde reaksiyon gözlendi. Performik Asit/AB pH 2.5 uygulamasında yumurtlama döneminde infundibulum ve magnum bölgelerinde reaksiyon görülmesine karşın yumurtlama dönemi öncesi sadece isthmus bölgesinde reaksiyona rastlandı. Yumurtlama dönemi magnum ve isthmus bölgelerinde nötral mukosubtans gözlenirken uterus bölgesi ve yumurtlama dönemi öncesi tüm bölgelerde reaksiyon gözlenmedi. KOH/PAS uygulamasında yumurtlama dönemi infundibulum, magnum, isthmus, vagina örtü epiteli hücrelerinde ve yumurtlama dönemi öncesi infundibulum ve vagina örtü epiteli hücrelerinde güçlü reaksiyon gözlendi. Nötral ve asidik mukosubtansı birlikte içeren hücreler yumurtlama dönemi isthmus ve vagina bölgesinde rastlandı. Sülfatlı ve karboksilli asidik mukosubtansı birlikte içeren hücrelere yumurtlama dönemi magnum, uterus, vagina bölgelerinde ve yumurtlama dönemi öncesi isthmus ve vagina bölgelerinde gözlendi. Sonuç olarak uygulanan histokimyasal yöntemler sonucunda farklı gelişme aşamalarındaki tavukların yumurta kanalı yapısında bulunan çeşitli glikonjugatların karakterizasyonu belirlendi. Elde edilen yumurtlama dönemi ve yumurtlama dönemi öncesi histolojik ve histokimyasal bulguların diğer kanatlı türleri ile benzerlik ve farklılıklar gösterdiği, ortaya çıkan bu farklılıkların ise tür ve yaş gibi faktörlerden kaynaklanabileceği sonucuna varıldı. In this thesis, it was aimed to determine histochemically the glycoconjugates in infundibulum, magnum, isthmus, uterus and vagina regions of oviduct of Denizli chicken (Gallus gallus domesticus) during laying period (16 weeks and after) and pre-laying period (10 to 16 weeks). Oviduct of 6 healthy laying and pre-laying chicken each from Farmer Training and Agricultural Practice Poultry in Faculty of Agriculture, Süleyman Demirel University was used as the material. These glands were observed in magnum, isthmus and uterus regions while not existing proprial glands in infundibulum and vagina regions during laying and prelaying period. Cells containing strong sulfate were found in surface epithelium of magnum and vagina during laying period. A reaction was detected in only surface epithelium cells of isthmus region as a result of the Best Carmine method applied to both periods. Results of Alizarin Red application showed that the only reaction was observed in the uterus region during laying and pre-laying period. The results of Performic Acid / AB pH 2.5 application indicated that the reaction was determined in the infundibulum and magnum regions during laying period although only seen in the isthmus region during pre-laying period. Neutral mucosubstance was observed in the magnum and isthmus regions during laying period even though any reaction to neutral mucosubstances did not show in all regions during laying and pre-laying periods. The results of KOH/PAS application indicated that strong reaction was found in the surface epithelium cells of infundibulum, magnum, isthmus and vagina during laying period and in the surface epithelium cells of infundibulum and vagina during pre-laying period. Cells containing both neutral and acidic mucosubtances were found in the isthmus and vagina regions during the laying period. Cells containing both sulphate and carboxylic acidic mucosubtances were observed in magnum, uterus and vagina regions during the laying period and in the isthmus and vagina regions during the pre-laying period. In conclusion, the employed histochemical techniques revealed that various glycoconjugates were found throughout the oviduct of chicken in different developmental stages. It has been concluded that the histological and histochemical findings obtained from laying and pre-laying period showed differencesand similarities with other avian species, and that these emerging differences could result from factors such as species and age.
