Bu tez çalışmasında koyun (Ovis aries) abomazum (kardia, fundus ve pilorus) mukozasında bulunan glikokonjugatların, DBA (Dolichos biflous), PSA/PEA (Pisum sativum) ve AIA (Artocarpus integrifolia) lektinleri kullanılarak belirlenmesi amaçlandı. Bu çalışmada Isparta Et Entegre Tesisin'den temin edilen 10 adet koyun (Ovis aries)'a ait abomazumdan (kardiya, fundus vepilorus) alınan örnekler materyal olarak kullandı. Bouin solüsyonunda 16 saat süreyle tespit edilen dokular daha sonra rutin histolojik doku takibinden geçirilerek parafinde gömüldü ve bloklandı. Parafin bloklardan 5-6 μm kalınlığında alınan kesitlere lektin immünohistokimya yöntemi uygulandı. Kardiya bölgesinde yüzey epitel hücrelerinin yüzeyinde, apikal sitoplazmalarında, foveola gastrika epitel hücreleri ve bez epitel hücrelerinde DBA, PSA/PEA ve AIA reaksiyonu saptandı. Bez epitel hücrelerinin çekirdeklerinin üstündeki alanda DBA' ya karşı güçlü reaksiyon gözlendi. Fundus bölgesinde yüzey epitel hücreleri ve kollum hücrelerinin apikal sitoplazmalarında DBA, PSA/PEA ve AIA reaksiyonuna rastlandı. Foveola gastrika epitel hücrelerinin çekirdeklerinin üstündeki alanda çok güçlü AIA reaksiyonu saptandı. Fundus bezlerinde prensipal hücrelerin yüzeyinde DBA ve AIA uygulamasında çok güçlü reaksiyon gözlendi. Bezlerin hem yüzeye yakın kısımlarında hem de dip kısımlarda bulunan pariyetal hücrelerde uygulanan üç lektine karşı da reaksiyon belirlendi. Pilorus bölgesinde yüzey epitel hücrelerinin yüzeyinde ve apikal sitoplazmalarında PSA/PEA lektin uygulamasında reaksiyon saptanmadı. Bez epitel hücrelerinin çekirdeklerin üstündeki dar alanda DBA ve AIA'ya karşı çok güçlü reaksiyon gözlendi. Bez epitel hücrelerinin yüzeyinde ise DBA reaksiyonu gözlenmedi. Sonuç olarak, koyun (Ovis aries) abomazumunun kardiya ve fundus bölgelerindeki yüzey epitel hücreleri yüzeyinde α-mannoz veα-glukoz terminal uçlu glikokonjugatları bulunduğu tespit edildi. Ayrıca kardiya, fundus ve pilorus bölgelerinin yüzey epitel hücreleri ile fundustaki prensipal hücreler yüzeyinde α-Galaktoz uçlu glikokonjugatın çok yoğun miktarda olduğu saptandı. Bununla birlikte pilorus bez epitel hücrelerinin yüzeyi ile apikal sitoplazmalarında α-asetilgalaktozamina rastlanmadı. Anahtar Kelimeler: Koyun, abomazum, mukoza, lektin histokimyası.
In this thesis study it was aimed the determination of glycoconjugates in mucosa of sheep's (Ovis aries) abomasum (cardia, fundus ve pylorus) using DBA (Dolichos biflorus), PSA/PEA (pisum sativum) and AIA (Artocarpus intergrifolia) lectins. The samples taken from abomasum of 10 sheep (Ovis aries)obtained from Isparta Meat Integrated Plant were fixed in Bouin's solution during 16 hours. After fixation, the samples were passed through tissue following procedure and embedded in paraffin. The lectin histochemical method was performed to determine glycoconjugate character to the5-6 μm thickness sections taken from paraffin blocks. Results of lectin histochemistry revealed that a reaction to DBA, PSA/PEA and AIA lectins occurred in surface epithelial cells and their apical cytoplasm, gastric foveolar epithelial cells and glandular epithelial cells in the cardia region. A strong reaction to DBA was observed in an area above the nucleus of glandular epithelial cells. Similarly, a reaction to DBA, PSA/PEA and AIA lectins was seenin surface epithelial cells and the apical cytoplasm of column cells in the fundus region. A very strong reaction to AIA was determined in an area above the nucleus of glandular epithelial cells. A very strong reaction to DBA and AIA was detected on surface of glandular principal cells.It was established that parietal cells found in near-surface and bottom parts of the glands reacted with three lectins employed. Any reaction to PSA/PEA lectin was not found in surface epithelial cells and their apical cytoplasm in the pylorus region.A very strong reaction to AIA and DBAwas noticed in a narrow area above the nucleus of glandular epithelial cells. Any reaction to DBA was not detected on the surface of glandular epithelial cells. As a result, surface epithelial cells in the cardia and fundus regions of sheep's (Ovis aries) abomasum were found to contain α-mannose and α-glucoseterminal glycoconjugates. α-Galactose terminal glycoconjugates were also determined to abound in surface epithelial cells of fundus and pylorus regions and in the surface of principal cells in the fundus region.However, glycoconjugates containing α-D-acetylgalactosamine were not found in the surface of pyloric glandular epithelial cells and in their apical cytoplasm.
