Bu tez çalışmamızda; kemometrik kalibrasyon yöntemlerinden temel bileşen analiz yöntemi (PCA), temel bileşen regresyon yöntemi (PCR) ve kısmi en küçük kareler yöntemi (PLS), ilaç numunesinde bulunan amoksisilin (AMX), lansoprazol (LNZ) ve klaritromisin (CTM) etken maddelerinin aynı anda miktar tayinlerinde hiçbir ayırma işlemi uygulanmaksızın başarı ile uygulanmıştır. Yüksek Performanslı Sıvı Kromatografisi (HPLC) ile yapılan çalışma sonucu elde edilen veriler kemometrik olarak değerlendirilmiştir. Kemometrik yöntemlerin validasyonunda; AMX, LNZ ve CTM için sırasıyla 10-50 ppm, 5-25 ppm ve 120-600 ppm derişim aralıklarında her bileşen için 20 adet yapay karışım seti hazırlanmıştır. Optimize edilmiş HPLC metodu ile elde edilen veriler 6 farklı dalga boyunda DAD dedektör yardımıyla yapılmıştır (205 nm, 210 nm, 215 nm, 220 nm, 225 nm ve 230 nm). Kullanılan istatistik program ile kalibrasyon setinin pik alan oranları (etken madde /iç standart) ve derişim değerlerinin varyans-kovaryans matriksleri hesaplanmıştır. AMX, LNZ ve CTM karışımlarının analizi sonucunda elde edilen verilere PCR ve PLS yöntemleri uygulanarak, % geri kazanım sonuçları ile buna karşılık gelen bağıl standart sapma değerleri hesaplanmıştır. % geri kazanım sonuçları PCR yöntemine göre AMX, LNZ, CTM için sırasıyla %99,99; %100,05; ve %100,0; PLS yönteminde %99,99; %99,96 ve %99,94 olarak bulunmuştur. Bağıl standart sapma sonuçları AMX, LNZ, CTM için sırasıyla PCR yöntemine göre 0,0004; 0,02; ve 0,0002; PLS yönteminde 2,93; 2,39 ve 1,96 olarak bulunmuştur. Elde edilen veriler doğrultusunda PCA, PCR ve PLS yöntemleri; ilaç numunesindeki AMX, LNZ ve CTM ’nin eş zamanlı miktar tayinlerine uygulanmıştır. Bu çalışmada kullanılan kemometrik yötemler çok bileşenli ticari ilaç numunesinin kantitatif tayini ve çözünürlük testleri için etkili bir yöntem olduğu gözlemlenmiştir. In this thesis, three different chemometric calibration methods including principle component analysis (PCA), principal component regression (PCR) and partial least square (PLS) were successfully appllied to the simultaneous determination of amoxicillin, lansoprazole, and clarithromycin in drug sample without using any separation step. The results obtained by High Performance Liquid Chromatography (HPLC) were evaluated chemometrically. In the validation of chemometric methods; 20 artificial mixture sets were prepared for each component at 10-50 ppm for AMX, 5-25 ppm for LNZ and 120-600 ppm for CTM. The data obtained by optimized HPLC method were measured by DAD detector with 6 different wavelengths (205 nm, 210 nm, 215 nm, 220 nm, 225 nm ve 230 nm). The variance-couvarsian matrix of the peak area ratios (active ingredient/ internal standard) and concentration values in the calibration set were calculated by using the statistical program. % Recovery results and relative standard deviation values were calculated by applying PCR and PLS methods to the obtained data in the analysis result. The % recovery results for AMX, LNZ and CTM were found to be 99.99%, 100.05% and 100.0% in PCR method and 99.99%, 99.96% and 99.94% in PLS method, respectively. The relative standard deviation results for AMX, LNZ and CTM were found to be 0.0004, 0.02, ve 0.0002 in PCR method and 2.93, 2.39 ve 1.96 in PLS method, respectively. PCA, PCR and PLS methods in the direction of the obtained data were applied on the simultaneous analysis results of AMX, LNZ and CTM in the drug sample. Both of the twochemometric methods in this study can be satisfactorily used for the quantitative analysis and for dissolutions tests of multicomponent commercial drug.
