Lactarius deliciosus (Çıntar, Kanlıca), Agaricus bisporus (Kültür mantarı, Beyaz şapkalı mantar), Lentinus edodes (Shiitake), Pleurotus ostreatus (Yaprak mantarı, İstiridye mantarı), Ganoderma lucidum (Reishi), Hericium erinaceus (Aslan yelesi), Morchella spp. (Kuzugöbeği), Boletus edulis (Çörek mantarı, Ayı mantarı, Porçini), Cantharellus cibarius (Sarıkız mantarı) mantar türlerinin kuru madde, toplam kül, toplam fenolik madde içeriği, antioksidan kapasiteleri (ORAC ve CUPRAC), antimikrobiyal aktiviteleri ve HPLC ile fenolik asitlerini belirlemek amaçlanmıştır. Bu amaçla yapılan kül analizi sonucunda mantarların toplam kül miktarlarının 2,92-12,64 g/100 g k.a. aralığında değiştiği tespit edilmiştir. En düşük kül miktarı Ganoderma lucidum mantarında (2,92 g/100 g k.a.), en yüksek kül miktarı ise Morchella spp. mantarında (12,64 g/100 g k.a.) tespit edilmiştir. Mantarların su, metanol ve su-metanol (50:50 v/v) ekstraktlarının toplam fenolik madde içeriği Folin-Ciocalteu testi ile antioksidan kapasitesi ise oksijen radikal absorbans kapasite (ORAC) ve Bakır(II) indirgeyici antioksidan kapasite (CUPRAC) testi kullanılarak analiz edilmiştir. Kurutulmuş mantar örneklerinden fenolik maddelerin elde edilmesi için su (S), metanol (M) ve su-metanol (SM) (50:50 v/v) karışımı olarak 3 farklı çözücü sistem kullanılmıştır. Toplam fenolik madde içeriği açısından tüm mantar türlerinde en yüksek değerler su ekstraktlarında bulunmuştur. Boletus edulis (17,11 mg GAE/g k.a.), Morchella spp. (14,19 mg GAE/g k.a.) ve Pleurotus ostreatus (10,85 mg GAE/g k.a.) mantarlarının su ekstraktları en yüksek fenolik içerik göstermiştir. Mantar türlerinde en düşük fenolik madde içerikleri metanol ekstraktlarında gözlemlenmiştir. En düşük değerleri ise Cantharellus cibarius (0,65 mg GAE/g k.a.), Lentinus edodes (1,69 mg GAE/g k.a.) ve Hericium erinaceus (1,96 mg GAE/g k.a.) mantar türlerinin metanol ekstraktları göstermiştir. Antioksidatif özellikler açısından da mantarların çoğu toplam fenolik madde içeriği analizinde olduğu gibi su ekstraktlarında daha yüksek değerler göstermiştir. ORAC analizi sonucunda örneklerin antioksidan kapasiteleri 19,84-206,47 μmol TE/g k.a. aralığında değişim göstermiştir. En yüksek antioksidan kapasiteyi Boletus edulis su-metanol ekstraktı (206,47 μmol TE/g k.a.), Boletus edulis su ekstraktı (204,14 μmol TE/g k.a.) ve Morchella spp. su ekstraktı (199,31 μmol TE/g k.a.) göstermiştir. En düşük antioksidan kapasiteyi ise Cantharellus cibarius su-metanol ekstraktı (19,84 μmol TE/g k.a.), Cantharellus cibarius metanol ekstraktı (21,42 μmol TE/g k.a.) ve Cantharellus cibarius su ekstraktı (22,74 μmol TE/g k.a.) göstermiştir. CUPRAC analizi sonuçları da toplam fenolik madde içeriği ve ORAC analizi sonuçları ile korelasyon göstermiştir. CUPRAC analizi ile antioksidan kapasitenin 20,22-314,00 μmol TE/g k.a. aralığında değiştiği belirlenmiştir. CUPRAC analizi sonucunda en yüksek antioksidan kapasiteyi Boletus edulis su ekstraktı (314,00 μmol TE/g k.a.), Boletus edulis su-metanol ekstraktı (270,23 μmol TE/g k.a.) ve Morchella spp. su ekstraktı (233,81 μmol TE/g k.a.) göstermiştir. En düşük antioksidan kapasiteyi ise Cantharellus cibarius su- metanol ekstraktı (20,22 μmol TE/g k.a.), Lactarius deliciosus su-metanol ekstraktı (28,77 μmol TE/g k.a.) ve Pleurotus ostreatus metanol ekstraktı (31,61 μmol TE/g k.a.) göstermiştir. Örneklerin fenolik asitlerinin tespiti için yapılan HPLC analizleri sonucu mantar örneklerinin içerdikleri gallik asit, klorojenik asit, vanilik asit, kafeik asit, pkumarik asit ve ferulik asit değerleri hesaplanmıştır. Mantar türü ve kullanılan ekstraksiyon çözgeninin fenolik asit içerikleri üzerine etki ettiği belirlenmiştir. Tüm örnekler içerisinde en yüksek konsantrasyonda bulunan fenolik asidin gallik asit olduğu ve örneklerdeki gallik asit konsantrasyonlarının 0,18-4,89 mg/g k.a. aralığında değişim gösterdiği tespit edilmiştir. Gallik asit açısından tüm mantarlarda en düşük değerler metanol ekstraktlarında görülmüştür. Gallik asit ve klorojenik asidin mg/g k.a. oranında tüm mantar ekstraktlarında mevcut olduğu belirlenmiştir. Boletus edulis’in tüm ekstraktlarında 6 fenolik asidi de içeren tek mantar türü olduğu bulunmuştur. Bu mantar türünün en yüksek konsantrasyonda içerdiği fenolik asit olarak su-metanol ekstraktında gallik asit (3,58 mg/g k.a.) olduğu tespit edilmiştir. Boletus edulis ve Pleurotus ostreatus'un en yüksek fenolik asit konsantrasyonlarını içeren mantarlar olduğu tespit edilmiştir. Fenolik asit içerikleri yüksek olan bu mantarlar, toplam fenolik madde içeriği ve antioksidan kapasitelerinin de yüksek olması ile bu sonuçları desteklemektedir. Mantar örneklerinde E. coli, P. aeruginosa gibi Gram negatif bakterilere ve L. monocytogenes ve Staphylococcus aureus gibi Gram pozitif bakterilere karşı antimikrobiyal aktivite tespit edilmiştir. Tüm mantar türlerinde test organizmalarına karşı su veya su-metanol (50:50 v/v) ekstraktları metanol ekstraktlarına göre daha fazla inhibisyon alanı göstermiştir. Tüm mantar türleri en yüksek inhibisyon alanlarını bir Gram pozitif olan Listeria monocytogenes test mikroorganizmasına karşı göstermiştir. L. monocytogenes’e karşı inhibisyon alanları 9,00-22,33 mm arasında dağılım gösterirken bir Gram negatif olan E. Coli'ye karşı ise 7,00-10,00 mm arasında inhibisyon alanları ölçülmüştür. Tüm test organizmalarına karşı B. edulis ve C. cibarius türlerinin en az birer ekstraktı (su, metanol veya su-metanol) inhibisyon alanı oluşturmuş ve çalışmada mevcut mantar türleri arasında en güçlü antimikrobiyal aktiviteye sahip oldukları tespit edilmiştir. Bu çalışmada görüldüğü gibi ülkemizde kültürü yapılan ve doğal yenilebilir mantar türleri içerdikleri fenolik bileşenler ve antioksidan potansiyelleri açısından oksidatif zararları azaltacak koruyucu bir ajan olarak gıda endüstrisinde kullanılabilir. Total solids, ash content, total phenolic content, antioxidant capacity (ORAC and CUPRAC), antimicrobial activity and phenolic acids by HPLC of Lactarius deliciosus (Saffron milk cap mushroom), Agaricus bisporus (White button mushroom), Lentinus edodes (Shiitake), Pleurotus ostreatus (Oyster mushroom), Ganoderma lucidum (Reishi), Hericium erinaceus (Lion’s mane), Morchella spp. (Morel), Boletus edulis (Bolet, Porcini), and Cantharellus cibarius (Golden chantarella) mushrooms that are either harvested wild or cultivated in Turkey were determined in this study. Ash content of samples ranged between 2,92-12,64 g per 100 g of dry matter (d.w.). The lowest ash content was in Ganoderma lucidum (2,92 g/100 g d.w.) whereas the highest in Morchella spp. (12,64 g/100 g d.w.). Because phenolics are known to contribute largely to antioxidant potential, extraction methods were also studied. Total phenolic content and the antioxidant capacities of water, methanol, and water-methanol (50:50 v/v) extracts of these mushrooms were analyzed by the Folin-Ciocalteu, oxygen radical absorbance capacity (ORAC) and cupric reducing antioxidant capacity (CUPRAC) assays. Three different solvent system; including water (W), methanol (M) and water-methanol (WM) (50:50 v/v) mixture were used to obtain phenolic extracts from dried mushroom samples. Higher values of total phenolic contents were found in water extracts among all mushroom samples. Water extracts of Boletus edulis (17,11 mg GAE/g d.w.), Morchella spp. (14,19 mg GAE/g d.w.) and Pleurotus ostreatus (10,85 mg GAE/g d.w.) had the highest phenolic contents. The lowest phenolic contents were determined in methanol extracts of mushroom samples. Methanol extracts of Cantharellus cibarius (0,65 mg GAE/g d.w.), Lentinus edodes (1,69 mg GAE/g d.w.) and Hericium erinaceus (1,96 mg GAE/g d.w.) mushroom samples showed the lowest values. Water extracts of mushrooms showed also higher values for antioxidative properties similar to phenolic contents. Higher values of antioxidant capacities were observed in Boletus edulis (206,47 μmol TE/g d.w.) water-methanol extracts, Boletus edulis (204,14 μmol TE/g d.w.) water extracts, Morchella spp. (199,31 μmol TE/g d.w.) water extracts and Pleurotus ostreatus (169,11 μmol TE/g d.w.) water extracts. The lowest values of antioxidant capacities were observed in Cantharellus cibarius (19,84 μmol TE/g d.w.) water-methanol extracts, Cantharellus cibarius (21,42 μmol TE/g d.w.) methanol extracts and Cantharellus cibarius (22,74 μmol TE/g d.w.) water extracts. The results of CUPRAC and ORAC assays showed a significant correlation. Results of antioxidant capacity based on CUPRAC assay ranged from 20,22 to 314,00 μmol TE/g d.w.. The highest antioxidant capacities were determined in water extract of Boletus edulis and Morchella spp. (233,81 μmol TE/g d.w.) mushrooms whereas lowest antioxidant capacity values were found in water-methanol extracts of Cantharellus cibarius (20,22 μmol TE/g d.w.) and Lactarius deliciosus (28,77 μmol TE/g d.w.), and methanol extract of Pleurotus ostreatus (31,61 μmol TE/g d.w.) mushrooms. In addition to the determination of total phenolics and antioxidant capacity assays identification of phenolic acids; gallic acid, chlorogenic acid, caffeic acid, vanillic acid, p-coumaric acid and ferulic acid, by HPLC analysis were performed. Mushroom species and extraction solvent had a significant impact on phenolic acid concentration. Gallic acid had the highest concentration and ranged between 0,18 and 4,89 mg/g d.w. among all extracted samples. The lowest values for all mushroom samples in terms of gallic acid were found in methanol extracts. Gallic acid and chlorogenic acid were detected in all samples. All extracts of Boletus edulis were found to be the only species of mushroom containing 6 of the phenolic acids. Gallic acid (3,58 mg/g d.w.) was present in the water-methanol extract as the phenolic acid contained in the highest concentration of this mushroom species. Boletus edulis and Pleurotus ostreatus contained the highest phenolic acid concentrations, which supports our results because of the high content of total phenolic contents and antioxidant properties. Mushroom extracts showed a mild to strong antimicrobial activity against Gram negative Escherichia coli and Pseudomonas aeruginosa, and Gram positive Listeria monocytogenes and Staphylococcus aureus. Water or water-methanol extracts of all mushroom species showed greater inhibition zones than those of methanol extracts. Zones of inhibition against L. monocytogenes ranged from 9 to 22,33 mm; However, same extracts showed 7 to 10 mm inhibition zones against E. coli. Highest antimicrobial activity was observed in water and/or water-methanol extracts of Boletus edulis and Cantharellus cibarius. Natural edible mushrooms cultured in Turkey can be used in the food industry as a protective agent to reduce oxidative damage in terms of phenolic components and antioxidant potentials. Mushroom extracts showed a mild to strong antimicrobial activity against Gram negative Escherichia coli and Pseudomonas aeruginosa, and Gram positive Listeria monocytogenes and Staphylococcus aureus. Water or water-methanol extracts of all mushroom species showed greater inhibition zones than those of methanol extracts. Zones of inhibition against L. monocytogenes ranged from 9 to 22,33 mm; However, same extracts showed 7 to 10 mm inhibition zones against E. coli. Highest antimicrobial activity was observed in water and/or water-methanol extracts of Boletus edulis and Cantharellus cibarius. Natural edible mushrooms cultured in Turkey can be used in the food industry as a protective agent to reduce oxidative damage in terms of phenolic components and antioxidant potentials.
