Bu çalışmada, dişi ve erkek keçi kalbinin atriyum ve ventriküllerinde atrial natriüretik peptit (ANP), beyin natriüretik peptit (BNP), C-tip natriüretik peptit (CNP) ile natriüretik peptit reseptörlerinin (NPR-A, NPR-B ve NPR-C) immunohistokimyasal dağılımlarının belirlenmesi amaçlandı. Uygulanan peroksidaz-anti-peroksidaz (PAP) immunohistokimya yöntemi sonucunda çalışılan bütün kardiyak bölgelerde natriüretik peptitlerin ve reseptörlerinin zayıftan güçlüye doğru immunoreaktivite gösterdiği tespit edildi. Erkek ve dişi keçi sağ ve sol atriyal kardiyomiyositlerinde ve endotel hücrelerinde güçlü ANP immunoreaktivitesi saptandı. Erkek ve dişi keçi sağ ve sol ventriküler kardiyomiyositlerinde, arter ve vena mediya tabakasındaki düz kaslarda zayıf-orta; endotel hücrelerinde ise orta-güçlü yoğunlukta ANP immunoreaktivitesi gözlendi. Erkek keçi sol atriyal kardiyomiyositlerinde zayıf, dişi keçi sol atriyal kardiyomiyositlerinde, endotel hücrelerinde ve kapillerde orta-güçlü yoğunlukta BNP immunoreaktivitesi saptandı. Ayrıca damar mediya tabakasındaki ve endokardiyum tabakasındaki düz kas hücrelerinde de immunoreaktivite saptandı. Erkek keçi sağ atriyal kardiyomiyositlerinde sol tarafa göre daha güçlü immunoreaktivite gözlenirken, dişi keçi sağ atriyal kardiyomiyositlerinde sol tarafa göre daha zayıf immunoreaktivite saptandı. Hem dişi hem de erkek keçi endotel hücrelerinde güçlü immunoreaktivite saptandı. Dişi ve erkek keçi sol ve sağ ventriküler kardiyomiyositlerinde sırası ile orta ve orta-güçlü yoğunlukta BNP immunoreaktivitesi saptanırken; endotel hücrelerinde güçlü immunoreaktivite tespit edildi. Erkek keçi sol atriyal kardiyomiyositlerinde ve endotel hücrelerinde orta-güçlü yoğunlukta CNP immunoreaktivitesi gözlenirken, dişi keçi sol ve sağ, erkek keçi sağ atriyal kardiyomiyositlerinde zayıf CNP immunoreaktivitesi gözlendi. Erkek keçi sol ve sağ ventriküler kardiyomiyositlerinde güçlü immunoreaktivite gözlenirken, dişi keçi sol ve sağ ventriküler kardiyomiyositlerinde zayıf immunoreaktivite gözlendi. Sol ventriküldeki endotel hücrelerinde ise zayıf-orta yoğunlukta, sağ taraftaki endotel hücrelerinde güçlü immunoreaktivite saptandı. Erkek keçi sol atriyal kardiyomiyositlerinde zayıf NPR-A immunoreaktivitesi saptanırken, dişi keçi sol atriyal kardiyomiyositleri ile dişi ve erkek keçi sağ atriyal kardiyomiyositlerinde zayıf-orta yoğunlukta immunoreaktivite saptandı. Erkek keçi endotel hücrelerinde orta-güçlü yoğunlukta immunoreaktivite saptanırken, dişi keçi endotel hücrelerinde zayıf immunoreaktivite görüldü. Dişi ve erkek keçi sol ve sağ ventriküler kardiyomiyositlerinde ve endotel hücrelerinde zayıf immunoreaktivite saptandı. Erkek keçi sol ve sağ, dişi keçi sol atriyal kardiyomiyositlerinde ve endotel hücrelerinde orta yoğunlukta NPR-B immunoreaktivitesi saptanırken, dişi keçi sağ atriyal kardiyomiyositlerinde zayıf immunoreaktivite saptandı. Hem atriyum hem de ventriküllerdeki endotel hücrelerinde orta yoğunlukta immunoreaktivite saptandı. Dişi keçi sağ ventrikülündeki koroner arter mediya tabakası düz kas hücrelerinde güçlü immunoreaktivite saptandı. Ayrıca erkek keçi sol ventrikülündeki epikardiyal gangliyonlarda zayıf-orta yoğunlukta immunoreaktivite tespit edildi. Dişi ve erkek keçi sağ ve sol atriyal ve ventriküler kardiyomiyositlerinde ve endotel hücrelerinde orta-güçlü yoğunlukta NPR-C immunoreaktivitesi tespit edildi. Ayrıca erkek keçi sol ventrikülündeki epikardiyal gangliyonlarda güçlü yoğunlukta immunoreaktivite tespit edildi. Sonuç olarak, bu çalışmada keçi kalp dokusunda natriüretik peptitler ve reseptörlerinin immunohistokimyasal dağılımı araştırılmıştır ve bu yönü ile memeli kalp dokusunda hem natriüretik peptitleri hem de reseptörlerini birlikte inceleyen ilk çalışmadır. NPR-A ve NPR-B'ye göre daha güçlü NPR-C immunoreaktivitesi gözlenmesi, bu reseptörün özellikle endotel hücrelerindeki temizleme fonksiyonuna atfedilmektedir. Ayrıca, memeli kalp dokusunda ANP ve BNP’nin yanı sıra CNP, NPR-A, NPR-B ve NPR-C’nin immunohistokimyasal dağılımına yönelik sınırlı sayıdaki çalışmalara da önemli ölçüde katkıda bulunduğu düşünülmektedir. The aim of this study was to investigate the immunohistochemical distribution of atrial natriuretic peptide (ANP), brain natriuretic peptides (BNP), C-type natriuretic peptide (CNP) and their receptors (NPR-A, NPR-B and NPR-C) in cardiac atria and ventricles of male and female goats. Peroxidase-anti-peroxidase (PAP) immunohistochemistry method revealed that natriuretic peptides and their receptors show weak to strong immunoreactivity in all cardiac regions studied. ANP immunoreactivity was found to be strong in right and left atrial cardiomyocytes and endothelial cells of male and female goats. The immunoreactivity was determined to be weak to moderate in the right and left ventricular cardiomyocytes and smooth muscles of artery and vein media layer of the male and female goats. The moderate to strong immunoreactivity was established in endothelial cells. While weak BNP immunoreactivity was detected in the left atrial cardiomyocytes of the male goat, moderate to strong immunoreactivity was found in the left atrial cardiomyocytes of the female goat, in the endothelial cells and capillaries. Immunoreactivity was also detected in smooth muscle cells in the media layer of vessels and endocardial smooth muscle cells. As stronger immunoreactivity was observed in the right atrial cardiomyocytes of the male goat than the left side, weaker immunoreactivity was detected in the right atrial cardiomyocytes of the female goat than the left side. Strong immunoreactivity was detected in the endothelial cells of both female and male goat. Moderate and moderate to strong BNP immunoreactivity was detected in the left and right ventricular cardiomyocytes of the female and male goats, respectively, with observation of the strong immunoreactivity in the endothelial cells there. As moderate to strong CNP immunoreactivity was observed in the left atrial cardiomyocytes and endothelial cells of the male goat, weak immunoreactivity was detected in the left and right atrial cardiomyocytes of the female goat and in right atrial ones of the male goat. With the detection of weak immunoreactivity in the left and right ventricular cardiomyocytes of female goat, strong immunoreactivity was detected in the left and right ones of male goat. It has been found that the immunoreactivity was weak to moderate in endothelial cells of left ventricle, with the observation of strong immunoreactivity in endothelial cells at the right side. Weak NPR-A immunoreactivity was detected in the left atrial cardiomyocytes of the male goat when determining weak to moderate immunoreactivity in the left atrial cardiomyocytes of the female goat and in the right ones of female and male goats. Moderate to strong immunoreactivity was found in the endothelial cells of the male goat while weak immunoreactivity was observed in the endothelial cells of the female goat. Weak immunoreactivity was established in the left and right ventricular cardiomyocytes and endothelial cells of the female and male goats. When moderate NPR-B immunoreactivity was detected in the left and right atrial cardiomyocytes of male goat, in the left atrial ones of the female goat weak immunoreactivity was detected in the left atrial cardiomyocytes of the female goat. Moderate NPR-B immunoreactivity was detected in both atrial and ventricular endothelial cells, with being detected strong immunoreactivity in the smooth muscle cells of coronary artery media layer in the the female goat. In addition, weak to moderate immunoreactivity was detected in left ventricular epicardial ganglia of male goat. Moderate to strong NPR-C immunoreactivity was observed in the left and right atrial and ventricular cardiomyocytes and endothelial cells of male and female goats. Strong immunoreactivity was also determined in left ventricular epicardial ganglia of male goat. In conclusion, the immunohistochemical distributions of natriuretic peptides and their receptors in goat heart tissue have been examined in this study, and from this aspect, this is the first study that investigates both natriuretic peptides and receptors together in a mammalian heart tissue. The observation of stronger NPR-C immunoreactivity than NPR-A and NPR-B has been attributed to its clearance function in especially endothelial cells. Moreover, it has been thought to be contributed significantly to a limited number of studies on the immunohistochemical distributions of CNP, NPR-A, NPR-B and NPR-C, as well as of ANP and BNP, in mammalian heart tissue.
