Bu çalışmada, koyun (Ovis aries) alt solunum yolları organlarında (üst ve alt trake, primer bronş, akciğer) mast hücrelerinin dağılımı, yerleşimi, morfolojileri ve boyanma özelliklerinin belirlenmesi amaçlandı. Mast hücre sayımları ve demonstrasyonu için Toluidin Blue (TB)(pH 0.5), Thionin (pH 0.3), Alcian Blue(pH 0.3)/Safranin O(pH 1.0) (AB/SO) ve Berberin sülfat (pH 0.2) boya yöntemleri uygulandı. Tüm preparatlardaki mast hücrelerinin sayısal yoğunluğu oküler mikrometre yardımıyla belirlendi. Elde edilen veriler semikantitatif olarak değelendirildi. Çalışılan bölgelerin tümünde Mukozal Mast Hücrelerinin (MMH) genellikle Bağ Doku Mast Hücrelerinden (BDMH) daha fazla sayıda olduğu tespit edildi. TB, Thionin, AB/SO ve Berberin sülfat uygulaması sonucunda mast hücre yoğunluğunun en fazla akciğer ve primer bronşlarda olduğu belirlendi. Mast hücre Şekillerinin mekik, oval, yuvarlak yada Şekilsiz yapıda ve farklı büyüklüklerde oldukları saptandı. Thionin ve TB uygulamaları sonucunda mast hücrelerinin metakromazi gösterdikleri saptandı. AB/SO uygulama sonucu tüm bülgelerde mast hücrelerinin kırmızı (SO+), mavi (AB+) ve mor (AB/SO+) olarak reaksiyon vardikleri gözlendi. Berberin sülfat uygulamasında ise mast hücrelerin sarı fluoresans gösterdiği saptandı. Toluidine Blue (TB) (pH 0.5), Thionin (pH 0.3), Alcian Blue (pH 0.3) / Safranin O (pH 1.0) (AB / SO) and Berberine sulfate staining methods were applied to count and demonstrate the mast cells. The numerical density of mast cells in all the slides was determined by means of an ocular micrometer. The obtained data were evaluated in a semiquantitative manner. It has been found that mucosal mast cells (MMCs) generally outnumbered the connective tissue mast cells (CTMCs) in all the studied regions. TB, Thionin, and AB / SO and Berberine sulfate staining revealed that the mast cell density was the largest in lung and primary bronchus. Mast cells were found to be in the spindle, oval, round or amorphousshaped and in varying sized. Mast cells stained metachromatically with Thionin and TB. In the AB/SO staining, mast cells stained red with SO (+), blue with AB (+), and purple with AB/SO (+) in all the examined regions. Mast cells showed yellow fluorescence Berberine sulfate staining. In conclusion, it is thought that mast cells may contribute to distribution and heterogeneity, allergic diseases of respiratory tract, cancer diagnosis and treatment, pathological studies.
