Çalışmamızda Wistar-Albino cinsi erkek ratlara uygulanan 50 Hz (10 kV/s) elektrik alanın(EA) beyin dokusunda oluşturduğu etkilerin ve bu etkileri azaltmak için uygulanan antioksidan juglon (5-hidroksi-1,4-naftokinon)'un olumlu etkisinin immunohistokimyasal, kan dokusunda oluşturduğu etkilerin ise biyokimyasal, fizyolojik ve comet yöntemi ile değerlendirilmesi amaçlandı. Bu çalışmada Süleyman Demirel Üniversitesi Deney Hayvanları Üretimi ve Deneysel Araştırma Laboratuvarı'nda (HADYEK) yetiştirilen ergin erkek Wistar-Albino türü ratlar kullanıldı. Denekler rastgele 3 gruba ayrıldı (n=8). Grup I; kontrol, Grup II; 50 Hz elektrik alan, Grup III; 50 Hz elektrik alan + Antoksidan (juglon (5-hidroksi-1,4-naftokinon)/300 ppm), her gruba 30 gün boyunca günlük tek seferde gavaj yöntemi uygulandı. Deney sonunda ratların beyin dokuları çıkartıldı ve rutin histolojik takipler yapıldıktan sonra histolojik boyama ve immunohistokimyasal yöntem uygulandı. Histopatolojik değerlendirmelerde elektrik alan grubu beyin dokusunda anlamlı derecede hasar saptandı. İmmunohistokimyasal değerlendirmelerde ise gruplar arası anlamlı farklılığı belirlemek için non-parametrik testlerden Kruskul-Wallis yöntemi uygulandı. Buna göre gruplar arası farklılık anlamlı tespit edildi (p˂0.05). Bu anlamlı farklılık EA grubundan kaynaklanırken, EA+Juglon grubu ile Kontrol grubu arasında anlamlı bir farklılık olmadığı belirlendi. Kaspaz 3 ve tnf- α immun pozitif hücre yoğunluğu kıyaslandığında anlamlı bir farklılık gözlenmedi. Toplam comet skoru açısından gruplar arasında fark istatistiksel olarak anlamlı bulunmuştur (p=0.001). Gruplar arası yapılan karşılaştırmada; kontrol grubu ile elektrik alan grubu arasında fark istatistiksel olarak anlamlı ve elektrik alan grubunda comet skoru artmıştır (p=0.001), juglon grubunda comet skoru kontrol grubuna göre azalmış olsa da, fark istatistiksel olarak anlamlı değildir (p>0.05). Juglon grubu ile elektrik alan grubu arasında fark istatistiksel olarak anlamlı ve juglon grubunda comet skorunda azalma tespit edilmiştir (p=0.001). Kandaki antioksidan parametrelerini belirlemek için değerlendirilen MDA, CAT aktivitelerindeki değişiklik gruplar arasında anlamlı bulunmazken, elektrik alan grubundaki SOD aktivitesindeki artıştan kaynaklanan gruplar arası farklılıklar istatistiksel olarak anlamlı bulunmuştur. Kandaki Glukoz, ALT, Direkt Bilirübin, BUN biyokimyasal parametrelerinde gruplar arasında anlamlı bir farklılık olmadığı tespit edilmiştir (p˃0.05). Üre, AST ve İndirekt bilirubin biyokimyasal parametreleri ise gruplar arasında anlamlı bir farklılık olduğu, bu farklılığın EA grubundaki artıştan kaynaklandığı belirlenmiştir (p˂0.05). The aim of this study was to evaluate the possible effects of 50 Hz electric field (EF) on the brain tissue and positive effect of juglon (5-hidroksi-1,4-naftokinon) which has antioxidant property by immunohistochemical technique of male Wistar-Albino rats. The effects on blood tissue was also examined using biochemical, physiological and comet methods. Animals were randomly divided into three groups (8 in each group); group I: control, group II: electric field, group III: 50 Hz electric field+ juglon (5-hidroksi-1,4-naftokinon)/300 ppm. Each group was performed one juglone application per day by gavage during 30 days. At the end of the thirty day, the animals were sacrificed and brain tissue was subjected to routine histologic and immunohistochemical processes. As a result of histophatological examination; the brain tissue of 50 Hz electric field exposure rats showed severe histopathological changes. To evaluate of immunohistochemical process results Kruskul-Wallis was used and there was a significant (p˂0.05) differences were detected between groups. While this significant difference was due to the EA group, it was determined that there was no significant difference between the EA+Juglon group and the Control group. When compare to caspase 3 and tnf-α immunoreactive cell density; there wasn't observed any difference. The difference between groups was statistically significant according to total comet score (p=0.001). In comparison between the groups; the difference between the control group and the electric field group was statistically significant and the comet score in the electric field group was increased (p = 0.001), although the comet score in the juglone group was lower than the control group, the difference was not statistically significant (p>0.05). It was found that the comet scor was reduced in the juglone group and the difference between the juglone group and the electric field group was statistically significant (p=0.001). Assessed to determine antioxidant parameters on the blood, although increasing in SOD activity in electric field group was found significant among all of the groups, we did not find significant differences in MDA, CAT activity level. Also there was no significant differences in glucose, ALT, Direct Bilirubin and BUN levels in blood between groups (p˃0.05). Urea, AST, and indirect bilirubin biochemical parameters were found to be significantly different among the groups, and this difference was determined to be due to the increase in the EA group.
