Hücresel oksidatif stres düzeyinin normal sınırların üzerine çıkması Alzheimer hastalığı (AlzH)'nın ve amiloid beta (Aβ) senil plakların oluşmasında önemli bir role sahiptir. Gelişen teknolojiyle beraber nanomateryal kullanımı da giderek yaygınlaşmaktadır. Nörodejeneratif hastalıklardan AlzH yaklaşık %80'lik bir oranla en sık rastlanan demans tipi olarak bilinmektedir. AlzH tedavisinde şimdiye kadar klasik tıbbi yaklaşımlar benimsensede hastalığın tedavisinde fazla etkin olamadıkları bilinmektedir. AlzH görülen hastaların kan ve saç örneklerinde glutatyon peroksidaz ve selenyum (Se) yetersizliğinin olduğuna dair bildirimler mevcuttur. Se içeren nanopartiküllerin (SeNP), deneysel AlzH oluşturulan modellerde mitokondriyal oksidatif stres ve apoptozis düzeyleri üzerine henüz bir bildirim mevcut değildir. Bu nedenle bu çalışmada; DBTRG hücrelerinde in vitro AlzH modelinin tedavisinde Se ve SeNP'nin apoptozis düzeyleri, hücre canlılığı, kaspaz 3 ve 9 enzim aktiviteleri, mitokondriyal zar depolarizasyon ve hücre içi serbest oksijen radikalleri (SOR) üretimi seviyelerine etkileri araştırıldı. Hücreler; kontrol, Se (200 nM), Aβ (20 M), SeNP (50 nM), Aβ + Se ve Aβ + SeNP olmak üzere altı gruba ayrıldı. Tüm hücreler 24 saat boyunca rutin hücre kültürü ortamında bırakıldılar. İnkübasyonu tamamlanan gruplara yukarıda anılan analizler yapıldıktan sonra gerekli istatistik değerlendirmeleri tamamlandı. Kontrol grubuna kıyasla, Aβ grubunda SOR üretimi, mitokondriyal zar depolarizasyonu, apoptozis düzeyleri ile kaspaz 3 ve 9 aktivite düzeylerinin anlamlı bir şekilde arttığı gözlendi. Se, SeNP, Aβ + Se ve Aβ + SeNP gruplarında, bu düzeylerde anlamlı azalma gözlenirken, hücre canlılığı değerinin Aβ grubuna kıyasla önemli ölçüde arttığı gözlemlendi. Sonuç olarak; AlzH modeli oluşturulmuş DBTRG hücrelerinde mitokondriyal oksidatif stres ve apoptozis düzeyleri üzerinde Se ve SeNP'nin olumlu etkileriyle düzenlendiği gözlemlendi. Bu değerler üzerinde SeNP'ye kıyasla, Se'nin etkisi daha önemli seviyede olduğu gözlemlendi.
Alzheimer's disease (AlzH) is one of the neurodegenerative diseases, is known to the most common dementia type with a high rate of 80%. Although it is not an effective option for the treatment until now, the conventional methods are used to treatment of AlzH. Exceeding the normal limits of intracellular oxidative stress (SOR) levels has a trigger role of AlzH incidence and amyloid beta (Aβ) senyl plaque formation. The usage of nanomaterials with developing technology is becoming very common. In the present reports in the literature, the low level of selenium (Se) and glutathione peroxidase enzyme are observed in blood and hair of patientst with AlzH. There is no report on the effects of Se containing nanoparticles (SeNP) on apoptosis and SOR levels in the literature. Hence in this study, we investigated effects of selenium (200 nM and 24 h) SeNP (50 nM and 24 h) on apoptosis, cell viability, caspase 3 and 9 activity values, mitochondrial membrane depolarization and intracellular SOR production levels in in vitro AlzH model of DBTRG neuronal cells. The cells were divided into six groups as follows; control, Aβ, Se, SeNP, Aβ +Se and Aβ+SeNP. After all incubations (24 hours), the cells were experimented with aforementioned analyzes and statistical evaluations were completed. The SOR, mitochondrial membrane depolarization, apoptosis, caspase 3 and 9 values were lower in the Aβ group compared to control group, although cell viability level was higher in the group than in control. However, the values were decreased in Se, SeNP, Aβ +Se and Aβ+SeNP groups by the Se and SeNP treatments, although cell viability level in the group was increased by the treatments. In conclusion, treatment by Se and SeNP may help the cells keeping their survival though inhibition of Aβ-induced mitochondrial oxidative stress and apoptosis. It seems that protective effects of Se treatment on the values were more significant as compared to SeNP treatment. This is the first in vitro study and report that proves the amendatory effects of Se and SeNP treatments on cell viability parameters which impaired by oxidative stress in DBTRG cells as an AlzH model.
