Bu tez çalışmasında, sığır eritrositinden 2',5' ADP-Sefaroz afinite kromotografisi kullanılarak saflaştırılan glukoz-6-fosfat dehidrogenazın (G6PD) stabilitesini ve tekrar kullanılabilirliğini geliştirmek için glutaraldehit ile aktive edilmiş kitosan kaplı manyetik nanopartiküller üzerine kovalent olarak immobilizasyonu gerçekleştirilmiştir. Kitosan kaplı manyetik nanopartiküller, kitosan çözeltisi içinde Fe2+ ve Fe3+ iyonlarının amonyak ile kopresipitasyonuyla (in situ) sentezlenmiştir. Kitosan kaplı manyetik nanopartiküllerin glutaraldehit ile aktivasyonu sonrasında G6PD'nin immobilizasyonu gerçekleştirilmiştir. Kitosan kaplı manyetik nanopartiküller X-ışınları kırınımı (XRD), fourier transform infrared (Kızılötesi) spektroskopisi (FTIR), termogravimetrik analiz (TGA), titreşimli örnek manyetometresi (VSM) ve geçirimli elektron mikroskobu (TEM) teknikleri kullanılarak karakterize edilmiştir. İmmobilizasyon süresi, glutaraldehit ile aktivasyon süresi, glutaraldehit konsantrasyonu, glutaraldehit çözeltisinin pH'sı ve enzim/destek materyali oranının immobilizasyon üzerine etkisi belirlenmiştir. Serbest ve immobilize enzime ait optimum pH, sıcaklık ve kinetik özellikler belirlenmiştir. Ayrıca immobilize enzimin termal stabilitesi, saklama stabilitesi ve tekrar kullanılabilirliği çalışılmıştır. Sonuç olarak, immobilize G6PD, pH 8,0 K-fosfat tamponunda 50°C'de optimum aktivite göstermiştir. Serbest enzime ait Km değeri 66±0.5 μM ve Vmax değeri 1,42±0,03 μmoldk-1 iken, immobilize enzimin Km değeri 115±4 μM ve Vmax değeri 0,092±0,04 μmoldk-1'dir. Termal stabilite sonuçlarına göre, 5 saat 40°C'de inkübasyon sonucunda serbest G6PD aktivitesinin %32'sini, immobilize G6PD ise %39'unu sürdürmüştür. 50°C'de ise 5 saat sonunda serbest G6PD aktivitesinin %23.5'ini, immobilize G6PD %34'ünü korumuştur. 4ºC'de saklanan serbest ve immobilize G6PD'nin kalan aktivitesi 9 gün sonunda sırasıyla %33 ve %72'dir. 11 kez kullanım sonucunda immobilize G6PD aktivitesinin %29'unu sürdürmüştür. In this thesis study, G6PD purified from bovine erythrocytes by 2',5' ADP-Sepharose 4B affinity chromatography was covalently immobilized on the chitosan-coated magnetic nanoparticles activated with glutaraldehyde, to improve its stability and reusability. The chitosan-coated magnetic nanoparticles were synthesized by co-precipitating Fe2+ and Fe3+ with ammonia in chitosan solution. After activation of chitosan coated magnetic nanoparticles with glutaraldehyde, immobilization of G6PD was performed. The chitosan-coated magnetic nanoparticles were characterized by X-ray diffraction (XRD), fourier transform infrared spectroscopy (FTIR), thermogravimetric analysis (TGA), vibrating sample magnetometer (VSM), and transmission electron microscopy (TEM) techniques. The influences of immobilization time, activation time with glutaraldehyde, the enzyme/support ratio, glutaraldehyde concentration for activation, and pH of the glutaraldehyde solution on immobilization were determined. The optimum pH, temperature and kinetic parameters of free and immobilized enzyme were determined. Additionally, thermal stability, storage stability and reusability of the immobilized enzyme were studied. As a result, the immobilized G6PD showed the optimum activity at pH 8.0 with K-phosphate buffer at 50°C. While the value of Km was 66±0,5 μM and the value of Vmax was 1,42±0,03 μmoldk-1 for the free enzyme, the value of Km was 115±4 μM and the value of Vmax was 0,092±0,04 μmoldk-1 for the immobilized enzyme. According to thermal stability results, the free G6PD sustained 32% of its activity, though the immobilized enzyme sustained 39% of its activity after incubation at 40°C for 5 h,. Whereas, free G6PD sustained 23.5% of its activity, the immobilized G6PD sustained 34% of its activity in the end of 5 h incubation at 50°C. The retained activity of the free and immobilized G6PD was 33% and 72% over a period of 9 days on storage at 4°C, respectively. After eleven repeated uses, the immobilized G6PD sustained 29% of its initial activity.
