Bu tez çalışmasında, ticari faj preparatının çiğ sütten yapılmış beyaz peynirde patojen kontrolü üzerine etkisi araştırılmıştır. Bu amaçla Listeria (PhageGuard Listex) ve Salmonella (PhageGuard S) faj preparatları denemede kullanılmıştır. Ön deneme sonucunda Listeria fajının aktivite gösterememesi nedeni ile çalışmanın dışında tutulmuştur. Beyaz peynir üretimi 4 grupta yapılmıştır. Bunlar; 1-kontrol grubu (çiğ süt), 2-çiğ süt+patojen (Salmonella Typhi, %0.05), 3-çiğ süt+faj ilave edilen (%0,1), 4-patojen (%0,05) + faj (%0,1) grubu olarak belirlenmiştir. Faj ve patojen bakteri, süte mayalama aşamasından hemen önce ilave edilmiştir. Üretilen beyaz peynirler %10'luk salamuraya alınarak 4 °C'de 90 günlük olgunlaşmaya bırakılmıştır. Beyaz peynir örneklerinde olgunlaşmanın 0., 30., 60. ve 90. günlerinde fiziko-kimyasal ve mikrobiyolojik analizleri yapılmıştır. Yapılan fiziko-kimyasal analizler; pH, titrasyon asitliği (ºSH), kuru madde, kuru maddede yağ, tuz ve kuru maddede tuz tayinidir. Mikrobiyolojik analizler; toplam aerobik mezofil bakteri, laktik asit bakterileri, Stafilokok spp., koliform grubu bakteri, Salmonella spp. ile faj ilave edilen peynir örneklerinde faj titresi analizidir. Ayrıca faj ilave edilen örneklerin sütlerinde, peynir altı sularında ve salamurada da faj titresi durumu incelenmiştir. Salmonella spp. sayımının doğrulanması için de şüpheli kolonilere üreaz testi uygulanmıştır. Ön denemelerden sonra, peynirler 3 tekerrür, analizler ise 2 paralel olarak yapılmıştır. Peynir örneklerinde, faj uygulamasının veya söz konusu bir patojen bulaşışının örnekler arasında fiziko-kimyasal ile genel mikrobiyolojik özellikleri bakımından farklılık gösterici etkisinin olmadığı elde edilen veriler sonucunda belirlenmiştir. Bu değerler tüm peynir gruplarında sadece süreye göre değişkenlik göstermiştir. Salmonella spp. sayısında ise 90. günde 2 ve 4 numaralı grup arasındaki fark kontrol grubuna göre kıyaslandığında istatisiksel olarak önemli bulunmuştur. Bunun haricinde Salmonella spp. sayıları gruplar arasında farklı bulunmamış, sadece olgunlaşmanın doğal süreci olarak günlere göre azalış göstermiştir. Bu çalışma sonucunda, çiğ sütten yapılan beyaz peynirlerde bakteriyofaj ilavesinin peynirin fiziko-kimyasal ve genel mikrobiyolojik özelliklerine etki etmediği ve faj etkisi ile patojen bakteri sayısında önemli bir fark olmadığı tespit edilmiştir. Faj ile biyokontrolde en önemli noktalardan biri fajın konakçıya özgül olmasıdır. Bu çalışmada kullanılan faj preparatının, yabancı kökenli olması, kullanılan patojen bakterinin ise yerli izolat olan Salmonella spp. için spesifikasyonun yetersiz olmasından dolayı Salmonella spp. sayısında önemli bir fark görülmemiştir. In this thesis study, the effect of commercial phage preparation on the pathogen control in beyaz cheese made from raw milk was investigated. Listeria (PhageGuard Listex) and Salmonella (PhageGuard S) phage preparations were used for this purpose. The Listeria phage is excluded from the study because it does not show activity in preliminary experiments. Beyaz cheese production was carried out in 4 groups; 1-control group (raw milk), 2-raw milk and pathogen (Salmonella Typhi, 0,05%), 3-raw milk and phage (0,1%), 4-pathogen (0,05%) and phage (0,1%). The phage and pathogenic bacteria were added to milk just before rennet inoculation. The beyaz cheese was taken in 10% brine and left to ripening for 90 days at 4 ° C. Physico-chemical and microbiological analyzes were performed on at 0, 30, 60 and 90 days of ripening in beyaz cheese samples. Physico-chemical analyzes were made; pH, titratable acidity (ºSH), dry matter, fat in dry matter, salt and salt in dry matter. Microbiological analyzes were made; total aerobic mesophilic bacteria, lactic acid bacteria, Staphylococcus spp., coliform group bacteria, Salmonella spp. and phage titre analysis in cheese samples added phage. In addition, the phage titer status of the samples added with phage was investigated in milk, whey and brine. For confirmation of the Salmonella spp. count, the urease test was applied to the suspect colonies. After preliminary assays, the cheeses were analysed in triplicate and the analyzes were carried out in two parallels. It has been determined that in cheese samples phage application or a pathogenic contamination didn't show difference effect between the samples in physico-chemical and general microbiological properties. These values varied only according time in all cheese groups. Salmonella spp. count was statistically significant when comparing the difference between groups 2. and 4. on day 90 compared to the control group. Apart from this, Salmonella counts were not different among the groups but only decreased as compared with the days of ripening as a natural process. As a result of this study, it was determined that the addition of bacteriophage in white cheeses made from raw milk did not affect the physico-chemical and general microbiological properties of the cheese and there was no significant difference on count of pathogenic bacteria by the phage effect. One of the most important points in biocontrol with phage is host specifity. There is no significant difference in the count of Salmonella spp. because the phage preparation used in this study is of foreign origin and the used pathogen bacterium is inadequate for the native isolate Salmonella spp. Keywords: Bacteriophage, Beyaz cheese, Pathogen, Phage titer, Ripening.
