Turunç; üzerine aşılı çeşidin büyümesini, gelişimini, verimini, meyve kalitesini, biyotik ve abiyotik streslere karşı dayanımını olumlu yönde etkileyen bir anaçtır. Bu nedenle turunç anacı ülkemizde ve dünyada turunçgil yetiştiriciliğinde en yaygın kullanılan anaçlardan biridir. Fakat turunçgillerin en önemli virüs hastalığına yol açan Turunçgil tristeza virüsü (Citrus tristeza virus,CTV)'ye duyarlı olduğu için virüsün yaygın olduğu bölgelerde kullanılamamaktadır. Bu nedenle CTV'ye dayanıklı turunç geliştirilmesi ülkemizde ve dünyada turunçgil ıslahının ana amaçlarından birini oluşturmaktadır. Bu çalışmada CTV'nün hastalık oluşturmada etkili p23 geninin RNAi yöntemiyle susturularak CTV'ye dayanıklı anaç geliştirmek için turunç anacının genetik transformasyonu yapılmıştır. Öncelikle GenBank veritabanında yeralan 400'e yakın CTV izolatının p23 geninin dizlimi karşılaştırılarak p23 genine spesifik kısa engelleyici RNA (short interfering, siRNA) kodlayıcı DNA tasarlanmış ve bu bölge polimeraz zincir reaksiyonu (PCR) yöntemi ile çoğaltılmıştır. PCR ile çoğaltılan bölge Karnabahar mozaik virüsü (Cauliflower mosaic virus, CaMV) 35S promotör ve terminatör bölgeleri arasına klonlanmıştır. Elde edilen gen yapısı daha sonra pCAMBIA1304 binary plazmidin T-DNA bölgesine klonlanarakAgrobacterium tumefaciens'eaktarılmıştır. Bu gen yapısı daha önce geliştirilen ve turunçgilerde yaygın olarak kullanılan Agrobacterium transformasyon yöntemi kullanılarak tohumlardan yetiştirilen turunç epikotil parçalarına aktarılmıştır. Transformasyon yapılan epikotil parçalarından doğrudan organogenez yoluyla önce sürgün oluşturulmuş daha sonra köklendirme veya mikro aşılama yapılarak transgenik bitkiler elde edilmeye çalışılmıştır. Elde edilen bitkilerde beta-glukuronidaz (GUS) boyama testi ve PCR testiyle transgen çoğaltımı yapılarak CTV p23 genini hedefleyen siRNA kodlayıcı gen yapısının turunç anacına aktarılıp aktarılmadığı belirlenmiştir. Çalışma kapsamında toplam 1250 epikotil parçasına genetik transformasyon yapılmıştır. Transformasyon yapılan epikotil parçalarından 76 sürgün elde edilmiştir. Bu sürgünlerden elde edilen 58 adet sürgüne GUS testi yapılmıştır Test edilen örneklerin hiç birinde GUS raportör gen aktivitesi görülmemiştir. Bu bitkilerden 25 tanesi mikro aşılama yoluyla aşılanarak tüm bitkiciler elde edilmiştir. Aşılanarak gelişmeye devam eden 18 adet bitkiden alınan yeni sürgünlerden DNA izolasyonu yapılarak CTV p23 genini hedefleyen siRNA kodlayıcı gen yapısını içerip içermediği PCR yöntemi ile test edilmiştir. PCR analizleri sonucunda 4 bitkinin aktarılan gen yapısını içerdiği tespit edilmiştir. Elde edilen sonuçlar CTV p23 genini hedefleyen siRNA kodlayıcı gen yapısının genetik transformasyon yoluyla turunç bitkisine aktarıldığınıgöstermiştir. Sour orange (SO) is the rootstock that positively affects the growth and development, yield, fruit quality, and resistance to biotic and antibiotic stresses of, citrus varieties grafted on it. Thus, itis one of the most commonly used rootstocks in citrus production in Turkey and in the world. However, it is sensitivity of the most important viral disease of Citrus, Citrus tristeza virus (CTV) and connot be used asrootstocks in regions where CTVis common. For this reason the development of SO rootstock resistant to CTV is one of the main objectives of citrus breeding programs in Turkey and the world. In thisstudy, SO rootstock was genetically transformedwith a construct coding for short interfering RNA (siRNA)targeting p23gene involved in pathogenicity of the virus. For this, the first DNA sequences of p23 genes from about 400 strains of CTV in the GenBank were compared. Then, DNA sequence coding for siRNAspecific to p23 gene was designed and these DNA sequences of were amplified with polymerase chain reaction (PCR) method. The PCR product was cloned between 35S promoter and terminator sequences of Cauliflower mosaic virus, (CaMV) to generate p23specific siRNA construct. The gene construct with the promoter and terminator sequences wasthen cloned into T-DNA region of binary plasmid pCAMBIA1304 and the vector was transferred to Agrobacterium tumefaciens. Epicotyl fragments obtained from in vitro grown SO seedlings were was transformed with the construct in binary plasmid pCAMBIA1304 using Agrobacterium transformation method widely used in citrus.Transgenic plants were regenerated from epicotyl segments by direct organogenesis with shoot and root inductions. Potential transgenic plants were tested by beta-glucuronidase (GUS) staining and PCR test. A total of 1250 epicotyl segments were transformed and 76 shoots were obtained from transformed epicotyl pieces. 58 of these shoots were tested using GUS and none of them showed GUS reporter gene activity. Among these shoots 25 were grafted onto in vitro grown SO seedlings using micro-grafting method. 18 of micro-graft were successful and shoots were established on SO rootstock. DNAs was extracted from plants established by micro grafted shoots and used for testing the presence of the siRNA construct targeting p23 gene of CTV using PCR method. Based on the PCR analysis, 4 plants containing thesiRNA construct were identified. The results showed that the siRNA construct targeting p23 gene of CTV was successfully transferred to SO rootstock by genetic transformation.
