Domates (Solanum lycopersicum), ülkemizde ve dünyada yaygın olarak yetiştirilen, tüketilen ve ticareti yapılan tarımsal üründür. Domates pek çok hastalık ve zararlıdan etkilenmektedir. Ancak kimyasal mücadele yöntemi bulunmamasından dolayı virüs hastalıkları domates üretiminde verim ve kaliteyi düşüren faktörlerin başında gelmektedir. Domates kloroz virüsü (Tomato chlorosis virus, ToCV) başta Akdeniz ülkeleri olmak üzere dünya domates üretim alanlarında yayılmaktadır. Son yıllarda ülkemizde ve dünyada önemli kayıplara neden olmaktadır. Günümüzde virüs hastalıklarına karşı en etkili mücadele yönteminin genetik dayanıklılık olmakla birlikte ToCV'ye karşı doğal dayanıklılık bulunmamaktadır. Doğal dayanıklılığın olmaması nedeniyle bitki–virüs etkileşimleri araştırılarak alternatif yöntemlerin geliştirilmesi gerekmektedir. Domates-ToCV ilişkisi üzerine yapılan bir çalışmada; bitkide ToCV enfeksiyonu sırasında ekspresyonu azalan yüzlerce gen belirlenmiş olup bu genlerin büyük bir kısmının kloroplast ile ilişkili olduğu görülmüştür. Bu çalışmada kloroplast ile ilişkili 10 farklı domates geninin ToCV enfeksiyonu sırasında ekspresyonları ayrıntılı olarak incelenmiştir. Bu amaçla öncelikle ToCV'ye duyarlı MoneyMaker domates çeşidine aşılma yoluyla ToCV inokulasyonu yapılmıştır. İnokulasyon yapılan ve kontrol bitkilerinden inokülasyondan 1, 8, 15, 30, ve 45 gün sonra yaprak örnekleri alınarak total RNA izolasyonu yapılmıştır. Daha sonra elde edilen RNA'lardan oligo(dT) primeri kullanılarak cDNA sentezlenmiştir. Son olarak seçilen 10 gen SYBR Green bazlı real-time RT-PCR yöntemiyle çoğaltılarak inokulasyonun farklı aşamalarında genlerin ekspresyon düzeyleri aktin referans genine göre incelenmiştir. Yapılan analizler sonucunda kloroplast ile ilişkili 10 farklı genin ekspresyonlarının inokulasyonun farklı aşamalarında baskılandığı belirlenmiştir. Ancak bazı genlerin ekspresyonlarında inokulasyonun farklı aşamalarında artışlar gözlemlenmiştir. Bu çalışma sonucunda; Ferritin-2, Demir-sülfür Cluster assembly protein 1, Cytochrome b6-complex demir-sülfür subunit genlerinin ekspresyonlarında inokulasyonun 15. günde azalma meydana gelmiştir. Protein CHUP1, Ferredoxin-2, Taylokoid formation 1, Curvature Tylokoid 1A, Fotosistem II CP47 klorofil apoprotein, Superoksit dismutase [Fe], L-askorbate perokidaz S genlerinin ekspresyonunda sadece 1. günde azalma olduğu ancak ekspresyonun daha sonra arttığı gözlemlenmiştir. Yapılan ekspresyon analizleri bu genlerin ekspresyonlarının baskılanmasının kloroz oluşumunda etkili olabileceğini göstermiştir. Bu çalışmada elde edilen bulgular domateste ToCV enfeksiyonu sırasında kloroplast sentezi ve fonksiyonlarıyla ilgili genlerin ekspresyonlarının olumsuz etkilendiğini göstermiştir. Ancak kloroz oluşum mekanizmasının anlaşılması için daha fazla chloroplast geninin ekspresyonunun incelenmesi gerektiği sonucuna varılmıştır. Tomato (Solanum lycopersicum) is the most widely grown, consumed and traded agricultural product in our country and in the world. Tomatoes are affected by many diseases and harmful factors. Because at the absence chemical control methods, virus diseases are the leading cause of decrease yield and quality in tomato production. Tomato chlorosis virus (ToCV) spreads throughout the world's tomato production areas, especially in the Mediterranean countries. It causes serious economical losses in Turkey and the world in recent years. Currently, the most effective method of combating viral diseases is genetic resistance, but there is no natural resistance against ToCV. Due to lack of natural resistance plant-virus interactions were investigated for development of alternative resistance stategies. In a previous study on tomato-ToCV interaction; the expresion of hundreds of genes was reduced during ToCV infection in tomato plant. A large proportion of these genes were associated with chloroplast. In this study, 10 chloroplast-related tomato genes were selected and their expression in respose ToCV infection of was examined in detail. For this purpose, first ToCV-sensitive MoneyMaker tomato seedlings were inoculated with ToCV by grafting. Then leaf samples were collected from ToCV inoculated and control plants at 1, 8, 15, 30, and 45 days after inoculation. Total RNA was isolated and cDNA was synthesized from the RNAs obtained using oligo (dT) primer. Lastly, selected genes were amplified by SYBR Green based real-time RT-PCR method and the expression levels of these genes were compared using actin as the reference gene at different stages of inoculation. As a result of these analyses, it was determined that the expression of 10 different genes related to chloroplast was suppressed at different stages of inoculation. However, increases in the expression of certain genes were observed in different stages of inoculation. The results showed that the expression of Ferritin-2, iron-sulfur cluster assembly protein 1, was decreased at 15 day after inoculation. The expression of Cytochrome b6-complex iron-sulfide subunit, L-ascorbate peroxidase, Superoxide dismutase [Fe], CHUP1, Ferredoxin-2, Tylokoid FORMATION1, Curvature Tylokoid 1A, Photosystem II CP47 chlorophyll apoprotein, genes was decreased only on 1 day after inoculation and its expression is then increased. Expression analyses revealed that supression at these genes by ToCV may have no effect on chlorosis formation. The results of this study showed that the expression of genes involved in chloroplast synthesis and functions were adversly afected by ToCV infection. However, understanding the mechanism of chlorosis formation requires expression analyis of many more chloroprast related genes. Keywords: Tomato, ToCV, Gene expression, Real time PCR, Chlorosis
