Doğal kefir danesi, kendine özgü bir mikrobiotaya sahiptir. Bu yapı polisakkarit bir matriks içinde laktik asit bakterilerini (LAB), asetik asit bakterilerini ve mayaları belli oranlarda bulundurmaktadır. Kefir; sütte bulunan besin maddelerinin tümünü içeriğinde bulundurmasının yanında, 22-24 saat süren fermantasyon esnasında bazı vitaminlerin sentezlenmesi, proteinlerin, yağların ve laktozun ortamdaki enzimlerin etkileriyle kısmen parçalanmasıyla çeşitli biyoaktif özelliklere sahip olan bileşenler oluşmaktadır. Kefirin, antikanserojenik özelliği, bağışıklık sistemine, sindirim sistemine, laktoz intoleransa, kolesterol düşürücü ve diyabete karşı olumlu etkileri yapılan araştırmalarla desteklenmiştir. Bu tezin amacı; kefir fermantasyonu 0., 4., 8., 12., 16., 18., saatlerde ve soğuk depolamada 1. ve 7. günlerde alınan örneklerle laktozun fermantasyonu, pH, mikrobiyal içerik, galaktooligosakkaritler dahil karbonhidrat profilindeki değişimin tespit edilmesidir. Lactobacillus spp., fermantasyon sonunda 7,63 logkob/mL Lactobacillus acidophillus 10,18 logkob/mL' ye ulaşmıştır. Depolamanın 7. gününde Lactobacillus acidophillus, 9 logkob/mL, Bifidobacterium spp., 7,01 logkob/mL tespit edilmiştir. pH, fermantasyonun tamamlanmasına kadar kademeli olarak azalarak pH 4.6' ya ulaşmıştır. Laktoz içeriğinde 0., 8., 12., 16., 18. saatlerde ve 1. Gününde fermantasyon ve depolama süreci boyunca anlamlı bir fark tespit edilmiştir (P˂0,05). Likid kromatografik yöntemle Galaktooligosakkarit (GOS) potansiyeli bulunan pikler tespit edilmiştir. The natural kefir consultancy has its own microbiota. This structure contains lactic acid bacteria (LAB), acetic acid bacteria and yeast in certain proportions in a polysaccharide matrix. Kefir; In addition to having all of the nutrients contained in the milk, it also contains components that can have various bioactive properties by partially degrading the proteins, fats and lactose by the effects of the enzymes in the environment, during the fermentation for 22-24 hours and synthesizing some vitamins. Kefirin has been supported by studies that have positive effects on anticarcinogenic properties, immune system, digestive system, lactose intolerance, cholesterol lowering and diabetes. The purpose of this thesis is; kefir fermentation is the determination of lactose fermentation, pH, microbial content, changes in carbohydrate profile including galactooligosaccharides with samples taken at days 0, 4, 8, 12, 16, 18, and cold storage on days 1 and 7. Lactobacillus spp. reached 7,63 log kob / mL Lactobacillus acidophillus 10,18 log kob / mL at the end of fermentation. On the 7th day of storage, Lactobacillus acidophilus, 9 log kob / mL, Bifidobacterium spp., 7.01 log kob / mL were detected. pH gradually decreased until fermentation was completed. There was a significant difference in lactose content during the fermentation and storage periods at 0, 8, 12, 16, 18 hours and 1st day (P˂0,05). Liquid chromatographic method was used to detect peaks with Galactooligosaccharide (GOS) potential.
