Yaşlı sıçanlarda ılımlı yüzme egzersizinin öğrenme-bellek üzerine etkisi ve Sirtuin-1'in rolünün araştırılması İnsanın yaşlanma sonrası kognitif fonksiyonlarını en üst düzeyde etkin yapmak için etkili, ucuz ve düşük riskli bir strateji olarak fiziksel egzersiz önerilmektedir. Direnç eğitimi veya aerobik eğitimlere tabi tutulan deneklerde bilişsel kapasite ve plastisite gelişiminin olduğu gösterilmiştir. Çalışmadaki amacımız, orta ılımlı yüzme egzersiz uygulamasının sıçanlarda öğrenme-bellek etkinliğini Morris su labirent testi ile değerlendirildi. Egzersizin SIRT1'i aktiflediği alternatif yolağın CREB ve BDNF proteinlerinin ekspresyonundaki rolü araştırıldı. Çalışmada 32 adet erkek Sprague-Dawley (350-500 gr) 11-12 aylık ve 15-16 aylık yaşlı sıçanlar kullanılmıştır. Çalışma dört gruptan ve her grupta 8'er adet sıçan olacak şekilde randomize olarak oluşturulmuştur. Gruplar; kontrol (K1) (11-12 aylık), Egzersiz (E1; 11-12 aylık), Kontrol (K2; 15-16 aylık), Egzersiz (E2; 15-16 aylık) olmak üzere ayrılmıştır. 30 dk/gün, 5 gün/hafta yoğunluğunda eğitim periyodu 8 hafta sürecek tarzda ılımlı orta dereceli egzersiz şiddeti uygulandı. Morris verilerinden, platform bulma parametresinde zamana göre istatiksel olarak gruplar arasında fark yoktu (p>0,05). Platform zonuna giriş parametresinde Egzersiz 1 ile Egzersiz 2 arasında istatistiksel olarak anlamlı fark vardı ve giriş sayısı arttı (p=0,026). Hipokampus dokusunda egzersiz gruplarında kontrol gruplarına göre SIRT1, BNDF, CREB immunoreaksiyonlarının arttığı ortaya konuldu. SIRT1'in BDNF'yi onunda CREB'i etkinleştirip nöroplastisiteyi aktive eden bir sinyal yolağı olduğunu öngörmekteyiz. Ilımlı egzersizin yaşlı sıçanlardaki plastisiteye olan bu geliştirici etkisi için alternatif bir yolak olarak SIRT1 ile aktive edilen bu yolağın yaşlılıkta meydana gelen demans için yeni bir terapötik hedef olabileceğini düşünmekteyiz.
The effect of moderate swimming exercise on learning-memory and the role of Sirtuin-1 in elderly rats Physical exercise is recommended as effective, inexpensive and low-risk strategy to make human cognitive functions effective after aging. Cognitive capacity and plasticity development have been shown in subjects subjected to aerobic training. The aim of the study was to evaluate the learning-memory efficiency of moderate swimming exercise by Morris water maze test. The role of exercise on the expression of CREB and BDNF proteins in the alternative pathway in which SIRT1 was activated was investigated. The study included 32 male Sprague-Dawley rats (350-500 g) aged between 11-12 months and 15-16 months old. The study was randomized into four groups and 8 rats in each group. Groups; Control (11-12 months), Exercise (11-12 months), Control (15-16 months), Exercise (15-16 months). 30 minutes / day, 5 days / week training period 8 weeks, moderate exercise was applied. There was no statistical difference between the groups according to time in the platform finding parameter from Morris data (p> 0.05). There was a statistically significant difference between Exercise 1 and Exercise 2 in the platform zone input parameter and the number of entries increased (p = 0.026). In the hippocampus tissue, it was revealed that SIRT1, BNDF and CREB immunoreactivity increased exercise groups according to the control groups. We foresee SIRT1 is a signaling pathway activates BDNF and activates CREB and activates neuroplasticity. We think that this pathway activated by SIRT1 as an alternative route for enhancing effect of mild exercise on the plasticity in elderly rats may be a new therapeutic target for dementia.
